A quantitative study of visual cortex synapses during the postnatal development of dark-reared rats.

The method of Aghajanian and Bloom (1967) was applied to the visual cortex of normal and neonatally visually deprived rats. The rats were kept with their mothers in total darkness since birth, in a ventilated and temperature-controlled animal quarter. Controls were rats from the same stock, maintained in a regular 12-h-light/12-h-dark rhythm. The animals were killed at 15, 23, 40, and 65 days of age, and the visual cortices fixed in 4% glutaraldehyde and processed for electron micrography with ethanol-phosphotungstic acid. A total of 6249 synaptic profiles were counted and their numerical density (DS) determined in both conditions. Features of the presynaptic grid were used for classifying the synaptic profiles in: type A [with one presynaptic dense projection (PsDP)]; type B (with two or three PsDPS); and type C (with four or more PsDPS). In the visually deprived rats the DS increases with a rate similar to controls, but the values for each age are slightly lower (P less than 0.01). Type B synapses predominate in the visually deprived group while types A and C are scarcer. The differences found between types were maximal at 65 days of age and the results were highly significant (P less than 0.001). It is concluded that major effects of dark-rearing are manifested when the individual features of the presynaptic grid are considered. It seems that a selected population of synapses is affected by the alteration of the normal epigenetic influence of early visual experience.

Effect of medium composition on the growth and asparaginase production of Vibrio succinogenes.

Asparaginase synthesis by Vibrio succinogenes is induced by ammonium ions. Synthesis occurs throughout exponential phase, and in early stationary phase asparaginase accounts for about 5% of the total soluble protein. The organism grows best when fumarate is provided as the terminal electron acceptor of the formate-oxidizing cytochrome system. Yeast extract or enzyme-hydrolyzed proteins are effective nutrient sources. In an ammonium formate-sodium fumarate medium, where maximum growth and asparaginase synthesis occurs, the total enzyme yield (international units per liter of culture) is about one-tenth that obtainable with a good asparaginase-producing strain of Escherichia coli. The energetic inefficiency of V. succinogenes appears to cause a low yield of cells and therefore low total enzyme yield. However, the levels of asparaginase accumulated within cells raise questions about the organism's protein synthesizing system.

[Communicating with patients: how good are our pamphlets and other written materials? An evaluation of the quality of the written information available to patients with asthma and diabetes in out-patients' clinics in Milan].

OBJECTIVES: 1) To identify the extent and the nature of information pamphlets available for asthmatic and diabetic patients in secondary care hospitals in Milan; 2) To investigate what patients think about the quality of pamphlets using a questionnaire developed ad hoc. METHODS: Pamphlets have been identified by contacting 8 specialised ambulatory hospitals. The questionnaire was developed to achieve a quality rate in 3 dimensions of the materials (Style and readability, The condition and Treatment options). Patients were recluted in the same ambulatory hospital. RESULTS: 4 ambulatory hospitals and 40 patients agreed to participate in our survey. We identified 8 pamphlets (5 diabetes, 3 asthma). 6 of these have been promoted byfarmaceutical companies. The mean length was 31 pages and the issue most covered was treatment options (33% of pages dedicated). 30 pages (12% of the total) presented with publicity advertisements (6% direct and 6% indirect). The dimension with the best satisfaction rate was Condition, followed by Style and readability. CONCLUSIONS: In Milan, in secondary care asthmatic and diabetic patients, are often invited to consult pamphlets. They positively judged this material. During the study ambulatory physicians were poorly attentive about problems concerning patient access to information.

Concomitant radiation-doxorubicin administration in locally advanced and/or metastatic soft tissue sarcomas: preliminary results.

Doxorubicin was administered by continuous infusion at a dosage of 12 mg/sqm/day for 5 days concomitantly with radiation treatment (150 or 200 cGy/day for trunk or extremity lesions, respectively) for 5 days. The 5-day cycles were repeated every 3 weeks. Seventeen patients, 5 of whom were pretreated, entered the study; all were assessable for toxicity and 15 for response. The overall objective response rate was 46% (7/15): 1 complete and 6 partial responses. Response rate reached 54% in only non-pretreated patients (6/11) and 75% in patients with PS less than or = 2 (6/8). No disease progression was observed during treatment. The median duration of complete or partial responses was 28 weeks (range 5-86). Toxicity was low and treatment very well tolerated. In our preliminary analysis, the response rate obtained with this combined chemo-radiotherapic regimen was encouraging and the toxicity was acceptable.
