[Proceedings: Peroxydase activity in benign thyroid diseases and in experimental rat goiters (author's transl)].

Peroxydase activity in various benign thyroid diseases has been investigated. In hyperthyroid patients treated with antithyroid drugs, the observed increase in iodide-peroxydase activity (7 times that of normal tissue) is double than that of the DNA content. In goiter, the activity is 3 times that of normal tissue while the DNA content is unchanged. These results are in agreement with those obtained in rats maintained on a low iodine diet suggesting that high cellular enzymatic activity is due to specific enzyme induction in excess of cell proliferation and protein synthesis.

[Importance of monoclonal antibodies in the analysis of tumor markers].

Due to their extreme specificity, monoclonal antibodies (Mabs) have been very useful for new analytical progresses in the field of tumor markers. For example, it has been possible to analyse only calcitonin, independently of its precursors in the patients with medullary thyroid carcinomas. These precursors have been estimated and found increased in the absence of calcitonin for many cancer patients, specially with small cell lung tumors. The assay of HCG and its subunits with a great specificity and sensitivity is an other interesting example of the analytical power of Mabs. The usefulness of the assay of beta subunit is now clear in the diagnostic and follow up of choriocarcinoma and perhaps in the screening of high risk pregnancies. An other example can be derivated of our works on neuropeptide Y (NPY). The assay of NPY is highly difficult, due to its great similarity with other neuropeptides (PYY and PP). Mabs have been used to solve these analytical problems. The assay of NPY is useful not only for patients with neuroendocrine tumors, but also experimentally in the rat hypothalamus for research in nutrition.

[Neuron-specific enolase in the aqueous humor. Its significance in the differential diagnosis of retinoblastoma].

Several studies support that retinoblastoma originates from a primitive neuroectodermal cell, the main argument being the expression of the neuron-specific enolase (NSE) by the retinoblastic cells. NSE was measured in the aqueous humor taken off: 1) in 19 patients with various eye disorders for which the diagnosis of retinoblastoma was definitely excluded (NSE = 5.0 +/- 7.7 micrograms/l); 2) at the time of the enucleation, in 7 cases of proved retinoblastoma (NSE = 83 to 17,900 micrograms/l); 3) in 2 cases of retinoblastoma held as cured for more than 5 years (NSE = 2.8 and 8.7 micrograms/l). The high level of NSE in the aqueous humor of the subjects with retinoblastoma is very significant, so that it can play a great part in the diagnosis, particularly in the case of atypic tumors. Specific indications of this determination may be considered for the survey of tumors under treatment or of the controlateral eye after a first tumor.
