Apoptosis of hypothalamic neurosecretory cells in stress mice at different stages of ontogenesis.

The aim of the present work was to identify the role of cycloferon in the apoptosis of cells in the neurosecretory centers of the hypothalamus in young and old mice in conditions of immobilization stress. Apoptotic cells were identified by staining with ethidium bromide. The optical density of the detection product of the antiapoptotic protein Bcl-2 was also studied in cells of the supraoptic and paraventricular (PVN) nuclei. Cycloferon was found to decrease the level of apoptosis in the neurosecretory centers of the hypothalamus via a Bcl-2-independent pathway. Administration of cycloferon before stress had no effect on the number of apoptotic cells, except in the PVN of old mice, where apoptosis was inhibited.

Involvement of interferon-alpha in the regulation of apoptosis of cells of the hypothalamo-hypophyseal-adrenocortical system of aged mice in oxidative stress.

The aim of this study was to identify the involvement of interferon-alpha (IA) in controlling apoptosis of cells of the hypothalamo-hypophyseal-adrenocortical system (HHACS) in young and aged mice in conditions of hyperoxia. Oxidative stress led to increases in the numbers of cells synthesizing the proapoptotic protein c-fos in the paraventricular nucleus in mice of both age groups. However, the protective actions of IA in stress were more marked at the earlier stage of apoptosis in young mice. Thus, the level of involvement of IA in controlling programmed cell death of hypothalamic cells depends on the age of the animals. In the fascicular zone of the adrenals in young mice, the number of dying cells was significantly greater after administration of IA, but remained at the control level in conditions of hyperoxia alone and in combination with IA. The proportion of apoptotic cells in the adrenals of aged mice was no different from that in young mice and did not change in response to any of the treatments used.

[Hypothalamic neurosecretory cell apoptosis during stress in mice at different stages of ontogenesis].

The aim of this work was to detect the participation of cycloferon in apoptosis control of cells of hypothalamic neurosecretory centers in young and old mice subjected to immobilization stress. Apoptotic cells were detected using ethidium bromide staining. Optic density of antiapoptotic Bcl-2 protein was determined in the immunoreactive cells of supraoptic and paraventricular nucleus (PVN). It was observed that cycloferon reduced activity of apoptosis in hypothalamic neurosecretory centers via Bcl-2-independent pathway. Cycloferon administration prior to stress had no influence on the number of apoptotic cells, except those ones in PVN of old animals, which demonstrated an apoptosis inhibition.

[Role of alpha-interferon in regulation of apoptosis in cells of the hypothalamo-hypophyseal-adrenocortical system of old mice in oxidative stress].

The aim of this work was to demonstrate the role of interferon alpha (IA) in apoptosis control of cells of hypothalamohypophysial adrenocortical system (HHAS) in young and old mice under conditions of hyperoxia. The increase of functional activity of neurosecretory cells of hypothalamic paraventricular nucleus in old mice treated with IA was demonstrated, while no effect was found in young mice. This differences could be associated with aging changes in HHAS in intact old animals. Oxidative stress caused an increase in the number of cells synthesizing proapoptotic c-fos protein in paraventricular nucleus of mice of both age groups. However, the protective action of IA against the stress is expressed more actively and in more early stage of apoptosis in young mice. Thus, the degree of IA participation in the control of programmed cell death of hypothalamic cells depends on the age of the animal. In the adrenal zona fasciculata of young mice the number of apoptotic cells was significantly increased after IA administration while after hyperoxia or its combination with IA it remained at control level. In the adrenal gland of old mice the proportion of apoptotic cells was not different from that found in young mice and remained unchanged after all experimental challenges.

Different pathways of neurohormonal hypothalamic control of the adrenal cortex function in young and old rats.

The pathways of hypothalamic control of the adrenal cortex function were studied in the experiments on young and old male Wistar rats. Immunohistochemical, morphometric, densitometric and radioimmunoassay methods were used. It was shown that in young rats under stress vasopressin (VP) is released into the portal circulation and probably in this way stimulates ACTH and enhances secretion of corticosterone in the adrenal cortex. In old rats stress-reaction is delayed and less adequate compared to young animals. It seems likely that in old rats VP is secreted into the general circulation mainly via the posterior pituitary lobe (PP), exerting a direct influence on the adrenal cortex. The conclusion is made that in old rats regulation of the adrenal cortex function is realised via the pathway phylogenetically more ancient than in young animals.

[Nucleolar apparatus of neurosecretory cells in the hypothalamus of rats of different age during acute immobilization stress].

To determine condition and adaptive capacity of cell nucleoli at ageing, the effect of acute immobilization on the hypothalamic neurosecretory cells was investigated in young and old male Wistar rats. Using immunohistochemical methods and nucleolometry we have shown that: 1) the nuclear volume in all neurosecretory cells is increased; 2) the share of cells, containing nucleoli with marginal position or multiple nucleoli in the nuclei, displays opposite changes in young and in old rats under stress condition. We suppose that adaptive mechanisms are different in young and old animals. Although, both kinds of morphological reorganization result in the increase in functional activity.

[Changes in the oxytocinergic neurosecretory cells in the accessory magnocellular neuroendocrine nuclei of the rat hypothalamus during aging].

Histophysiology of accessory magnocellular nuclei of hypothalamus was studied in young (3-6 months) and old (14-29 months) male Wistar rats. Using non-labeled antibodies to oxytocin and karyolometry activation of protein synthesis combined with inhibition of oxytocin transport along the fibres was shown to occur in 4 (circular, fornical, dorso- and ventrolateral) of 5 accessory nuclei in old rats. This was not observed in anterior commissural nucleus. A hypothesis on compensatory role of accessory magnocellular nuclei of hypothalamus in regulation of neuroendocrine homeostasis in old rats is discussed.

[The reaction to stress of the nonapeptidergic hypothalamic centers in old and young rats].

Diversities in reaction to immobilization stress in senile and juvenile animals were studied in 60 male Wistar line rats. Nonapeptidergic neurosecretory centres of hypothalamus were analysed morphometrically. Functional activity changes were found in vasopressin and oxytocin synthetic hypothalamic systems in senile and juvenile animals in different after-stress terms.

[Differences in the reaction of old and young rats to immobilization stress].

Studies have been made on the reaction of 55 male old and young Wistar rats to acute 1-hour immobilization. Corticosterone was determined in the blood serum using radioimmune assay. Similar phasic changes in the level of corticosterone in the blood serum were revealed in young and old rats, although in the latter these changes occurred 1 day later. The decrease in the body weight and in the mass of the adrenals was also observed which was more significant in old animals.
