[Structural changes in the microcirculatory bed and perivascular connective tissue in the myocardium during experimental hypercholesterolemia].

Light and electron microscopy were used in rabbits to study the effect of hypercholesterolemia (HC) on the microcirculatory bed (MCB), perivascular connective tissue and specialized cells of the myocardium. Within the first hours (1, 3, 6, 15, 24, 48 h) after cholesterol single administration to the animals, there were structural changes in the MCB, aggregation and superficial changes in red blood cells (echinocytes), stases, the presence of unusual ultrastructures in elastic membranes (EM) of the arterioles, in the amorphous substance of perivascular connective tissue, specifically near the venules, and focal dystrophy in cardiomyocytes. The ultrastructures detected in the EM of the arterioles and perivascular tissue seem likely to be the complexes of atherogenous lipoproteins and glycosaminoglycans. The transfer of the animals on the atherogenous diet entailed the reconstruction of small arteries of the myocardium and formation in them of so-called "Conti's pillows" as well as lipid deposition. A complex of structural changes revealed in the myocardium in the early stages of HC indicates the increased permeability of the microvessels, alterations in the blood rheological properties and circulatory disorders. A relationship was found between the intensity of the changes cited and blood cholesterol level. Injury to the MCB of an organ is likely to play an initial part in the mechanisms of atherogenesis.

[Effect of quercitrin on structural changes in the large and small intestines in experimental enterocolitis].

Sensitization of rats with the homologous antigen of the large intestine with Freund's stimulant caused marked immunomorphological shift in the large intestine of rats with the phenomena of enteritis in the small intestine. Oral administration of quercitrin in doses of 25 and 100mg/kg for 10 days promoted decline of focal lymphohistiomonocytic infiltrations, of eosinophilic in the mucosa and of scleroti changes in the large intestine. At the same time restoration of structural disturbances occurred in the small intestine. The influence of quercitrin on the morphological changes in the epithelium of the large and small intestine mucosa were insignificant. It is believed that the therapeutic action of quercitrin could be caused by restoration of local circulation deranged in animal sensitization.

[The effect of functional overstrain of the central nervous system and endocrine disorders on the development of experimental atherosclerosis in dogs].

In a chronic experiment (up to 6 years) on dogs (13) a protracted functional overstrain of the CNS was associated with an additional action changing the hormonal background (castration and mercasolyl injection). A steady hypercholesteremia, atherosclerotic changes in different intraorganic arteries, particularly in the myocardium, developed in dogs; dystrophic processes occurred in the parenchyma of various organs. An important role of functional overstrain of the CNS as one of the leading factors of the "risk" determining the development of disturbances of the regulation of lipid metabolism and formation of the atherosclerotic process in the vessels was demonstrated.

[Interrelationship of the cardiovascular and digestive systems in the genesis of experimental atherosclerosis].

In chronic experiments (up to 2 years) on dogs on atherogenic diets the motor activity of the gastrointestinal tract, cholesterol, the phospholipid level in the blood and in the intestinal secretion as well as the morphology of the small intestine and the vascular system were examined. At the early stages of atherosclerosis (up to 4 months) the rise of the blood cholesterol level was followed by an increase in the periods of the activity of the gastrointestinal tract, a rise of cholesterol discharge by the small intestine, and the development of the hypertrophic process in the mucous membrane. At the later stages a change in the mucous membrane of the small intestine was observed with development of a dystrophic-atrophic process, a disturbance of motor activity of the gastrointestinal tract and of the correlations between the cholesterol content in the blood and its discharge by the small intestine. This period was characterized by a high and persistent hypercholesterolemia and pronounced atherosclerotic changes in the vascular system.

[Morphofunctional state of the digestive tract in the early stages of atherosclerosis].

Cholesterol, alpha- and beta-lipoproteids, serotonin were determined in the blood serum of dogs which were on atherogenic diet for 2 months. The serotonin content was examined in various parts of the gastrointestinal tract (GIT). Parallel studies of the structural changes were carried out in the vascular system (VS) and various parts of the GIT. There was found a direct correlation between a rise in the cholesterol and serotonin level in the blood and serotonin in the GIT tissues. The initial stages of atherosclerotic changes were revealed in the vascular system. Along with compensatory-adaptive changes detected in the duodenum and the upper protions of the small intestine, initial stages of dystrophic-atrophic processes were observed in the lower portions. Comparative analysis of biochemical and morphological data indicated that disturbances of the morphofunctional state in the GIT played an important role in the genesis of the early stages of atherosclerosis.

[Effect of perftoran on microcirculation and target organs in early atherogenesis].

Effects of perfluoran, a plasma substitute with a gas-transport function, were studied at the stage of dyslipidogenic microangiopathy in rabbits. Changes in microcirculation, hemorheology, morphology of the liver, phospholipid composition of hepatocytic membranes were examined histologically, histochemically, electron microscopically and biochemically. Perfluoran produced a positive effect on microcirculation, phospholipid composition of hepatocytic membranes. Individual features of these parameters' dynamics were determined for animals.

[Influence of cell therapy and alimentary correction on the condition of microcirculatory system in experimental dyslipidemia].

A comparative analysis of the impact of cell therapy (allogenic fetal liver cells, stem cells of autologic bone marrow) on lipid homeostasis, microcirculatory disturbances and structural changes in some organs was made using the atherosclerosis model in guinea-pigs. A positive effect of cell therapy on these parameters was established. The effect was stronger and prolonged after transplantation of stem cells. Atherogenic diet weakens the effect of cell therapy. Thus, cell therapy is more effective in restriction of atherogenic risk factors.

[The correction of the pathological changes in the neuroendocrine system in hyperlipoproteinogenic microangiopathy].

Thymalin and alpha-tocopherol were studied for their effects on the morphofunctional status of the hypothalamopituitary neurosecretory system, thyroid, and adrenals in early atherogenesis. Correction of abnormal changes in the neuroendocrine organs restored lipid hemostasis, lowered the rate of lipid peroxidation, and prevented the progression of atherosclerotic changes in the great arteries.
