Local conformational changes in ferritins with variable iron content.

Conformational changes were induced in human spleen ferritin by partial or complete removal of iron, and the immunoreactivity of the ferritin samples with variable iron content was analyzed. We established that a decrease in iron content resulted in bimodal changes in immunoreactivity of the epitopes recognized by the monoclonal antibodies G10 and F11. Immunoreactivity demonstrated a 3-6-fold decrease on lowering iron content from 800 to 40 atoms per protein molecule, followed by a sharp (4-14-fold) increase that was observed when low-iron ferritin was converted to iron-free apoferritin. These bimodal changes suggest the presence of more than two conformational states of ferritin with local alterations of the epitopes recognized by the monoclonal antibodies. The global conformation of ferritin, however, remained essentially unaltered, as demonstrated by ferritin interaction with polyclonal antibodies. Together, the results indicate that local conformational changes in the ferritin protein shell occur on progressive iron removal that results in low-iron and iron-free forms of ferritin. These changes are most clearly seen in apoferritin when compared to low-iron ferritin.

[The mechanisms of the realization of the effect of glucocorticoids in aging].

The interaction of glycocorticoids with serum transport proteins, plasma membranes and rat liver cytoplasmic receptors progressively declines during ontogenesis, reaching its minimum at 24 months of age. Glycocorticoid receptor complexes (GRCs) binding to the rat liver nuclei and their residual fractions as well as the glycocorticoid-induced initiation of RNA-synthesis also decrease with age. The GRCs are shown to be capable of association with nuclear envelope, nuclear matrix and RNA-containing nuclear fraction isolated from the rat liver. The intracellular glycocorticoid receptor recycling requires fresh synthesis of the RNA and protein.

[The effect of nucleotides and molybdate ions on the activation of glucocorticoid-receptor complexes].

The effects of various nucleotides and sodium molybdate on the activation of glucocorticoid-receptor complexes (GRC) isolated from tissue cytosol of 6- and 25-month-old rats was studied. It was shown that nucleoside triphosphates activate GRC in the livers of 6-month-old rats, the activating effect being decreased in the following order: UTP greater than or equal to ATP greater than GTP greater than or equal to CTP. Nucleoside di- and monophosphates exert a far lesser stimulating effect. These effects of nucleotides decrease with ageing. Molybdate ions exert a 3-fold effect on the activation of GRC from various rat tissues, i.e., stimulating, inhibiting and zero effects.

[Study of the age-related features of the structural state of rat liver cell chromatin using micrococcal nuclease and circular dichroism].

The structural state of liver cell chromatin of young (6 months-old) and aged rats was evaluated, using circular dichroism and DNA hydrolysis by micrococcal nuclease. It was found that the chromatin condensation ratio in active and non-active fractions increased upon ageing. The length of the DNA nucleosomal repeat did not change upon ageing in either chromatin fraction. An elevated chromatin condensation ratio was observed in aged rats both for the unfolded nucleosome chain and the supernucleosomal structures. The mechanisms of age-related increase in the chromatin compactization ratio at different levels of organization and in dissimilarly active fractions are different. It was suggested that the age-related differences in the degree of compactization may stimulate a decrease of the transcriptional activity of liver cell nuclei upon ageing.

[Ischemic heart disease and the biological age of men over 40].

A population of 40- to 64-year-old males (normal subjects and coronary patients) were examined in order to estimate their biological age on the basis of cardiovascular and respiratory functional parameters and multiple regression analysis dynamometry in relation to hormonal status; it may be considered as a factor, contributing to cardiovascular pathology.

[Estrogen- and progestin-receptor complexes and their interactions with rat liver cell nuclei in ontogenesis].

Steroid-receptor complexes (SRC) of estrogen and progestin were isolated from rat liver and purified 1500-2000-fold. The SRC within the composition of cytosol and purified 2000-fold were characterized by gel filtration of Sephadex G-100 and by DEAE-cellulose chromatography. The purified SRC from rat liver were bound to isolated liver cell nuclei of rats of various age (1.5, 6, 12 and 24 month-old). The maximal binding of progestin and estrogen SRC from rat liver was observed in homologous nuclei of 1.5-month-old animals. The binding of SRC by the nuclei decreased progressively with age, reaching its minimum in 24-month-old rats. The observed differences in the SRC binding by cell nuclei of experimental animals may be the cause of functional changes at various stages of ontogenesis.

[Evaluation of radioprotective properties of a lipocarotenoid extract from mycelium of basidiomycetes during external irradiation].

Radioprotective properties of LPC extract from mycelium of Basidiomycetes fungus at doses 12.0; 3.0 and 1.0 Gy of external irradiation were investigated. The extract of LPC at 2.5 mkg/ml concentration promoted the survival of animals (to 40%) and increased the average life at the lethal dose. At 1.0 Gy dose the use of extract resulted in restoration of radiosensitive organs weights, the number of blood leucocytes and some indexes of lipid peroxidation in blood plasma. The extract has produced some effect on the amount of spermatogenic cells in the testis.

[Functional interactions of components of the adenylate cyclase system in the rat liver after prenatal exposure to gamma irradiation].

The effects of long-term prenatal gamma-radiation (0.5 Gy) on the glucagon signalling through adenylyl cyclase in adult rat liver have been investigated. The coupling of receptor/Gs-protein/adenylyl cyclase was tested using kinetic constants for stimulation of adenylyl cyclase by GTP, Gp(NH)pp, AlF4- and glucagon. The estimated data suggests that prenatal chronic gamma-radiation prompted (i) a decrease in rate of GTP hydrolysis on Gs-protein; (ii) a reduction of glucagon potency to accelerate the exchange GDP for GTP on Gs-protein.

[Effect of external irradiation with different intensity at 1 Gy dose on the DNA, RNA and total protein content in the testis and liver of rats].

The content of DNA, RNA and total protein in the rat testes and liver was studied 1 and 30 days after external chronic gamma-irradiation (dose rate 1.8 and 5.76 cGy/day) and acute gamma-irradiation with relatively low dose rate (5.4 cGy/min) up to a total dose of 1 Gy. The results obtained pointed to the specific metabolic reaction of radiosensitive (testes) and radioresistant (liver) tissues of organism at external irradiation at relatively low dose of different intensity due to unequal cell capacity for proliferation.

[The effect of acute external gamma-irradiation and thyroid glands dysfunction on rat reproductive system and frequency of structural mutations in somatic cells of pregnant females and their offspring].

Experimental hypofunction of thyroid gland, caused by mercazolil administration, and acute external exposure to 0.5 and 1.0 Gy of gamma-irradiation were shown to result in disturbance of reproductive function in puberal females and of estrogenreceptor interaction, in reduction of pregnancy probability, increase in pre- and postimplantation lethality of fetuses as well as in rise in the frequency of structural mutations in somatic cells of pregnant rats and their offspring.

[Morphofunctional state of the rat male reproductive system after chronic low intensity irradiation at a dose of 1.0 Gy].

It was studied the morphofunctional state of rat male reproductive system after chronically exposure to low intensive gamma-irradiation in early ontogenesis. After radiation with a total dose of 1.0 Gy it was revealed the decrease of the testes and epididymis weight, dysbalance in the ratio of different spermatogenic cells, changes in nucleic acid content and discoordination of bioenergetic metabolism in testes. Disturbance in rat male reproductive system retained in remote period (180 days after irradiation).

[Effects of prolonged low-intensity irradiation on organ weight of the male rat reproductive system].

The effects of prolonged irradiation at accumulated doses from 0.5 to 6.0 Gy (dose rate 3.03 cGy/day) on reproductive organs' weight (testes, epididymises, seminal vesicles, prostate) of male rats starting from the early ontogenetic period were studied. On the first day after the irradiation with 1.0 Gy dose a significant loss of the weight in testes and epididymises was revealed. This leaded to the marked atrophy of the organs with the increase of the radiation dose. Long-term restoration of the weight of testes and epididimyses was registered. It was not completed during three months after radiation exposure at 2.0 Gy and higher doses for epididimyses and 4.0-6.0 Gy for testes. The respective changes in the seminal vesicles and prostate weight were less pronounced and had more complicated character. However in the distant period (30-90 days of postreatment) after exposure to 2.0 Gy these parameters were noticeably decreased.

[The effect of iodine-131 on the binding characteristics of sex and thyroid hormones by blood plasma proteins in children with a functional thyroid disorder as a result of the accident at the Chernobyl Atomic Electric Power Station].

In studying characteristics of specific interaction of estradiol-, testosterone- and thyroid-binding blood globulins with the corresponding ligands in children from Gomel Province with endemic swelling of the thyroid gland (degrees I and II) affected by iodine-131 revealed were a reduced cooperativity in estradiol and T-3 binding and a halved affinity to androgens and thyroids as compared to healthy controls. In addition, there was a drastic decline in the binding capacity of estradiol- and testosterone-binding globulins in the blood plasma. The endemic thyroid gland swelling is supposed to be due to sexual malfunction in teenagers.
