Depression and monoamine oxidase.

1. The first part of the paper is devoted to a critical review on the possible relationships between depression and monoamine oxidase. 2. This study describes the results of an investigation of MAO-activity in depression, using new approaches and methodology. This methodology was developed because the literature data indicated that a) previous results are difficult to compare because of varying methods and diverse target populations used; b) previous methodological deficiencies do not allow to draw definite conclusions about the relationship between MAO-activity and depression. 3. The present investigation selected 35 psychiatric patients according to clearly defined diagnostic criteria (20 endogenous depressive, 10 neurotic depressive, and 5 manic patients) matched to 25 healthy control-subjects. 4. The Michaelis-constant (Km), the maximum reaction speed (Vmax), and the 50% enzyme inhibition by tranylcypromine (IC50) of platelet-MAO were determined during and after recovery from the depressive or manic episode using 3 substrates (tyramine, tryptamine, and phenylethylamine). 5. The present investigation, in contrast to conventional methodology, utilized three different substrates at different concentrations. Significant correlations were demonstrated for the Vmax-values of each of the three substrates, whereas the Km and the IC50 (tranylcypromine)-values varied for each substrate. 6. The results show that there were no differences between the characteristics of the platelet-MAO in depressive or manic patients and those of normal subjects. Furthermore, treatment with tricyclic antidepressants had no effect on MAO-activity. A previous investigation indicated that the MAO-properties in human brain tissue were similar to that in human platelets.

[Clinical trial of mepiprazol in the treatment of neurotic inpatients (author's transl)].

Within a controlled double-blind clinical trial the psychopharmacological effect of mepiprazole was tested against placebo in the treatment of neurotic inpatients. Some results of the recorded psychophysiological parameters (tremor, reaction time and flicker fusion threshold) as well as the scores on a symptom checklist for neurotic patients (post hoc divided into three scales by means of factor analysis: anxiety, depression and weaping) indicate a tranquilizing effect of mepiprazol. These findings are discussed critically with regard of methodological problems of testing tranquilizers in psychiatric hospitals.

[Studies on a novel antidepressant (viloxazin) by means of time-series analysis of TV data].

19 depressed patients were treated with viloxazin (n = 9) or amitriptyline (n = 10) in a double-blind-trial. TV-tapes were produced for each patient on day 0, 10 and 20. These tapes were analysed with respect to the intensity of depression by 4 independent raters under the condition of a time-blind analysis. These 3 TV-tapes could easily be discriminated in the sense of a decreasing depressivity. The results suggest that the effect of viloxazin has a shorter delay than that of amitriptyline.

[Practice and further development of the AMP-system (a report about a symposion and a seminar) (author's transl)].

During two days in November 1974 TV-tapes of psychiatric interviews were demonstrated and the psychopathology was documented on AMP-record sheet 3. It was a purpose of the discussion to get stimulation for the improvement of the AMP-manual and to decrease the interrater-variance. It was only the first tape which was not followed by a discussion. This film was presented a second time at the end of the seminar in order to examine the training effects. Both record-sheets were compared with each other. Generally the variance decreased. The discussion as to an improvement of the AMP-manual lead to the following suggestions: In defining a psychopathological item it is important to differentiate more precisely between the patient's subjective experience and his behaviour, to find better criteria concerning the rating of different intensities and to improve discrimination between singular items. Furthermore the logic of documentation procedure should be unequivocal and more practical. Following the seminar there was a symposion discussion more general and some specific methodological problems of the further development of the AMP-system. Finally study groups were formed to elaborate on the topics "AMP-manual", "rater-training", "social data" and "electronic data processing".

The rate of excretion of VMA in depressed patients before and after the administration of methionine.

The results of a previous experiment in which the relationship between VMA-excretion in the 24-hr-urine and a depressive syndrome was found were examined on the basis of the following hypothesis: 1. The patients of the present study show an increased VMA-excretion, because there are more depressed patients than in the previous experiment. 2. The amount of excreted VMA correlates positively with the degree of severity of their depression. The mean value of the VMA-excretion in the urine of 20 depressive patients of the present study corresponds to that obtained from the depressive patients in the previous exploratory experiment. The degree of severity of depression of the patients correlated with the VMA-excretion. Patients with an increased VMA-excretion were significantly more often treated with antidepressive drugs than those with normal VMA-values.
