[Contemporary possibilities of clinical immunology in oncology].

Clinical immunology involves studies of immunological diagnosis and monitoring of cancer patients, their immunity and treatment-induced changes, including those associated with immunotherapeutical exposures, immunodiagnosis of leukemias, estimation of solid tumor markers. Immunophenotyping of peripheral blood cells from patients with solid tumors treated with chemo- and immunotherapy has elucidated some relationships to evaluate the efficiency of treatment. Detailed characterization of the structure of leukemic clone has promoted development of individual treatment regimens. The authors' experimental studies will evaluate the effects of cytokines on the mechanisms of multidrug resistance caused by mdr-1 gene hyperexpression and on apoptosis processes diminished by bcl-2 gene hyperexpression.

Role of interleukin-2 and mononuclear cells in platelet-tumor interactions.

Interleukin-2 (IL-2) has been reported to induce mononuclear cell-dependent platelet perturbations and to reduce the incidence of metastases in rodent models. In this study, MC-1 leukemic cell-induced platelet aggregation was associated with a significant increase in thromboxane B2 (TXB2) secretion, quantified by RIA, but a decrease in leukotriene B4 (LTB-4) secretion compared to incubations of tumor cells alone. Whereas MC-1 cells also activated platelets as observed by a significant increase in scanning electron-microscopic spheroidal forms distributed throughout the platelet pool, increasing concentrations of IL-2-activated peripheral blood mononuclear cells (PBMC), but not IL-2 or PBMC alone, reversed this effect with a reduction in activated spheroidal forms. The addition of IL-2-activated PBMC to MC-1 platelet incubations inhibited platelet-tumor cell adhesion and suppressed MC-1-induced aggregation. In order to investigate the mechanism resulting in inhibition of MC-1-induced platelet aggregation, short-term PRP-MC-1 preparations were incubated with IL-2-activated PBMC and assayed for TXB2 and LTB-4 by RIA. LTB-4 levels were significantly elevated in these preparations while levels of TXB2 were significantly reduced compared with control platelet-rich plasma (PRP)-MC-1 incubations. Our results showed that inhibition of MC-1-induced platelet activation, aggregation and tumor-platelet adhesion by the addition of IL-2-activated PBMC was consequent to a cellular effect of IL-2 on PBMC. These findings may be important in understanding the observed antimetastatic effect of IL-2.

[Modeling of the "universal" bone marrow using monoclonal antibodies].

The data on the application of monoclonal antibodies (ICO-10) and rabbit complement for working the conditions of allogeneic bone marrow transplantation are presented in the paper. The treatment with monoclonal antibodies and bone marrow complement from BALB/c mice for 2 times prevented the development of transplant versus host reaction and completely protected lethally irradiated (CBA X X C57B1/6)FI mice-recipients from death. Thymus atrophy and the absence of T-cells in the peripheral blood was observed in these mice. The erythrocytes had markers characteristic of BALB/c and (CBA X C57B1/6)FI mice. Mouse splenocytes did not respond to the cells of donors and recipients in mixed lymphocyte culture reaction.

[Ultradian fluctuations in the natural antitumor resistance of mice undergoing transplantation and chemical carcinogenesis].

A study was made of the mouse life span in relation to the time of tumor cell injection on an hourly basis, while the carcinogenic effect was studied in relation to the time of urethan injection. Changes in both the values were shown to vary with ultradian rhythms. It was found that natural killer cell activity against tumor cells, migration index of normal spleen cells in the presence of tumor cells and the activity of carcinogen-metabolising enzyme system of intact animals also exhibited ultradian oscillations. It is assumed that oscillations of natural resistance against transplantable or chemically-induced tumor may be due to the changes of these parameters.

[Effect of preliminary administration of cyclophosphamide and pretransplantation irradiation on the growth of syngeneic tumors in mice].

The growth of the syngeneic tumor Acatol was retarded and the minimal number of tumor cells necessary for inoculation was increased in BALB/c mice preinjected with cyclophosphamide (100 mg/kg) and/or totally irradiated (5 Gy). Administration of spleen cells from intact mice abolished the antitumor effect of the above factors. The natural killer activity of spleen cells was estimated within 2-9 days after tumor inoculation. The natural killer activity in intact recipients was elevated on the 2nd day and then decreased to the control level. In irradiated recipients, the natural killer activity remained high throughout the entire observation period.

[Effect of the joint use of BCG and antithymocyte serum on the growth of a syngeneic tumor in mice].

The growth of the syngeneic tumor Acatol in BALB/c mice was retarded if the animals were pretreated with BCG or antilymphocyte serum (ATS). Combined use of BCG and ATS led to a significantly more powerful retardation as compared to the effect produced by each factor alone. Using the adoptive transfer of splenocytes from treated mice it was shown that tumor growth suppression is effected by the cell types other than T lymphocytes and macrophages. It is probable that the effector cells within the given system are K cells and natural killer cells. The results attest to a possibility of search for a two-directional action on the immune system of the tumor host, which would stimulate antitumor effector cells and inhibit the activity of suppressors, particularly that of T suppressors.

[Effect of various forms of human leukocyte interferon on the natural killer activity of human lymphocytes].

The effect of the preparations of human leucocytic interferon (HLI) of various purity levels and of various concentrations was studied with respect to the natural killer activity of human leucocytes. It was shown that the process of purification and concentration of the preparation of HLI with the Kantel method entailed a disproportionally high loss of the activity stimulating the natural killer cells as compared to the antiviral activity.

[Detection of an antigen, associated with cells during the blast crisis of chronic myeloleukemia, in a cytotoxic test using xenogenic antibodies].

The immune antimyeloblast serum (AMS) was obtained from horses immunized with white blood cells from patients suffering from chronic myeloid leukemia (CML) at the blast crisis stage; the serum was completely absorbed with normal red blood cells and white blood cells (WBC). The absorbed antiserum remained cytotoxic to blast cells from 20 of 42 patients with CML at the blast crisis stage. AMS failed to react with the WBC from patients with CML in its chronic phase, and from patients with other types of leukemia Morphological studies indicated a possibility of identification of the antigen associated with myeloblasts from the blood of patients with CML blast crisis, by means to AMS.

Specific xenogenous antiserum to cells of chronic myeloid leukemia (CML) in blastic crisis.

The antiserum was obtained from horses immunized with cells from patients with blastic crisis of CML and completely absorbed with normal white blood cells (WBC). The absorbed antiserum remained cytotoxic to blast cells from nearly half of the patients in blastic crisis and did not react with WBC from patients with Acute Myeloid Leukemia (AML), Acute Lymphoid Leukemia (ALL), Chronic Lymphoid Leukemia (CLL) and Chronic Myeloid Leukemia (CML) in its chronic phase as well as with cells of human normal bone marrow or fetal liver.

An attempt of treatment of malignant melanoma with bovine pineal tissue.

Seven incurable patients with disseminated malignant melanoma were injected (subcutaneously, sometimes intravenously) with homogenate of bovine pineal tissue. The daily injections of large quantities of pineal tissue appeared to be harmless for patients. Many-fold injections (during 9.5 months) of large quantities of pineal tissue (up to 3950 g) in 1 of 7 cases produced certain effect on subcutaneous metastases of malignant melanoma, i.e. causing reduction and even disappearance.

Rapidly acquired cytotoxicity of lymphoid cells from ice inoculated with allogeneic spleen cells.

Spleen cells from C57BL/6J or CBA mice inoculated iv with spleen cells from BALB/c mice produced a strong nonspecific cytotoxic effect on target cells (mouse L-cells). Lymph node cells from CBA or C57BL/6J mice inoculated sc with BALB/c spleen cells also destroyed L-cells. Lymph node cells from mice inoculated with syngeneic spleen cells were not cytotoxic. The cytotoxic effect was observed ion of allogeneic but not syngeneic spleen cells. This effect was considerably reduced or completely suppressed after partial or total removal of plastic-adherent cells.
