[Preliminary study of acidification of the pH of the aqueous humor in rats treated with timolol maleate].

The Authors have studied in a homogeneous group of rabbits the variations of intraocular pressure and of aqueous humour pH caused by instillation of timolol maleate at different concentrations. In the animals treated with drug concentration of 0,25% and 0,5% a constant reduction of IOP has been observed in comparison with controls, associated to acidification of aqueous humour: no evidence of this correlation it seems to be at low doses of timolol. Further studies are in course to indicate an eventual action of intermediate substances in the mechanism of IOP depression caused by timolol.

Demonstration of traumatic pleuro-biliary fistula with 131I-rose bengal.

Penetrating thoracic trauma may result occasionally in communication between the pleura and biliary tree. A case of traumatic pleurobiliary fistula secondary to stab wound of the right chest was demonstrated with (131)I-rose bengal scan. The diagnosis was made 15 days after trauma and was managed conservatively with a thoracostomy tube drainage.

IgG, IgA and IgM concentration in human sera from different age groups: statistical evaluation.

A statistical analysis is presented of the results obtained from the measure of the serum concentration of IgG, A, M in 773 blood donors. In each of the two groups identified by sex, four subgroups were established according to age in ten years intervals from 20 to 59. On the basis of the results of the analysis, different fiducial intervals of normal values (calculated as mean +/- 2 X standard deviation) have been proposed. 1) A unique interval for IgG: 87-234 U.I. 2) Two intervals for IgA: 54-236 U.I. for subjects under 40 years and 64-270 U.I. for subjects over 40 years. 3) Four intervals for IgM: one for males under 40 years: 65-364 U.I., one for males over 40 years: 57-314 U.I., one for females under 50 years: 47-363 U.I., and one for females over 50 years: 53-434 U.I.

Immediate endoscopic diagnosis of upper gastrointestinal bleeding. Its accuracy and value in relation to associated pathology.

Two hundred sixty-two patients with active upper gastrointestinal (GI) bleeding underwent panendoscopy between July 1970 and March 1973. There was 100% accuracy of endoscopic diagnosis as to the anatomical site of bleeding; the etiopathologic definition was 94.7% accurate. The series was divided into two groups, 116 with "liver disease" and 146 with "no liver disease." There were 107 patients with varices: 21 fell into no liver disease (small varices) and 86 into liver disease (39 small and 47 large varices). All had associated gastritis. Three endoscopic bleeding patterns were identified in the liver disease group. Only 27% of the patients in the liver disease group with varices (cirrhotics) had frank variceal hemorrhage, whereas 57% bled from hemorrhagic gastritis. The diagnostic unit provided early diagnosis, meaningful therapy, organized data gathering, and rough estimates of ultimate prognosis.

New tricyclic enamine derivatives with CNS depressant properties.

63 tricyclic enamine derivatives were synthetized and pharmacologically tested. Many of the screened compounds showed remarkable CNS depressant chlorpromazine-like activities. The compounds no. 9 (substituents CH3, Cl, H), 18 (substituents CH3, F, H) and 27 substituents CH3, OCH3, H) resulted the most interesting and were studied in comparison with chlorpromazine and chlordiazepoxide. They were submitted to further studies in view of trials in humans.

Pharmacological properties of new neuroleptic compounds.

RMI 61 140, RMI 61 144 and RMI 61 280 are newly synthetized N-[8-R-dibenzo(b,f)oxepin-10-yl]-N'-methyl-piperazine-maleates which show interesting psychopharmacologic effects. This work contains the results of a study performed with these three compounds, in order to demonstrate their neuropsycholeptic activity in comparison with chloropromazine (CPZ) and chlordiazepoxide (CPD). The inhibition of motility observed in mice shows that the compounds reduce the normal spontaneous motility as well as the muscle tone. The central-depressant activity is evidenced by increased barbiturate-induced sleep and a remarkable eyelid ptosis can also be observed. Our compounds do not show any activity on electroshock just as do CPZ and CPD. As to the antipsychotic outline, our compounds show strong reduction of lethality due to amphetamine in grouped mice and a strong antiapomorphine activity. They show also an antiaggressive effect and an inhibitory activity on avoidance behaviour much stronger than CPZ. We have also found extrapyramidal effects, as catalepsy, common to many tranquillizers of the kind of the standards used by us. As for vegetative phenomena, the compounds show hypotensive dose related action ranging from moderate to strong, probably due to an a-receptor inhibition. Adrenolytic activity against lethal doses of adrenaline, antiserotonin and antihistaminic effects, as well as other actions (hypothermia, analgesia, etc.) confirm that RMI 61 140, RMI 61 144 and RMI 61 280 are endowed with pharmacologic properties similar and more potent than those of CPZ. Studies on the metabolism of brain catecholamines show that they are similar to CPZ, although with less effect on dopamine level.
