Detection of adenovirus DNA in peripheral blood mononuclear cells by polymerase chain reaction assay.

Adenovirus can establish persistent infections which may reactivate and cause disease in immunocompromised hosts. Lymphocytes have been postulated to serve as a site of adenoviral persistence based upon the ability to isolate adenovirus from tonsils and to detect adenovirus DNA by Southern blot hybridization in peripheral blood mononuclear cells (PBMC). To test this hypothesis, a more sensitive and specific polymerase chain reaction (PCR) assay was developed to detect adenovirus DNA. Two sets of nested primers were designed to conserved sequences in the adenovirus E1A and hexon genes. The E1A and hexon primers amplified DNA from representative adenoviral serotypes in all six adenoviral groups (A-F). Both primers detected a single copy of the adenovirus type 2 genome but were less sensitive for the group B type 35. None of 33 PBMC specimens from healthy adults and only one of 40 pediatric samples was positive (at a low level) for adenovirus DNA by nested PCR assay. In comparison, PBMC from two children with fatal adenoviral infection were both strongly positive for adenovirus DNA. It is concluded that, in contrast to a previous study, PBMC are not a common site of persistent group C adenoviral infection. In addition, assay of PBMC by the adenovirus-specific PCR may help detect early invasive disease and warrants further evaluation.

A study of the chemical composition of Erythroxylum coca var. coca leaves collected in two ecological regions of Bolivia.

Coca-Erythroxylum coca Lamarck var. coca-remains one of the most common plants of the folk medicine of Bolivia used as a general stimulant. Aymara and Quechua natives prefer to chew the sweeter coca leaves from the Yungas (tropical mountain forests of the eastern slopes of the Andes) rather than those from the Chapare lowlands. The contents in cocaine and minor constituents of leaf samples cultivated in these regions does not rationalize this choice.

Effects of supplemental oxygen on forearm vasodilation in humans.

Supplemental O2 reduces cardiac output and raises systemic vascular resistance in congestive heart failure. In this study, 100% O2 was given to normal subjects and peak forearm flow was measured. In experiment 1, 100% O2 reduced blood flow and increased resistance after 10 min of forearm ischemia (flow 56.7 +/- 7.9 vs. 47.8 +/- 6.7 ml.min-1.100 ml-1; P < 0.02; vascular resistance 1.7 +/- 0.2 vs. 2.4 +/- 0.4 mmHg.min.100 ml.ml-1; P < 0.03). In experiment 2, lower body negative pressure (LBNP; -30 mmHg) and venous congestion (VC) simulated the high sympathetic tone and edema of congestive heart failure. Postischemic forearm flow and resistance were measured under four conditions: room air breathing (RA); LBNP+RA; RA+LBNP+VC; and 100% O2+LBNP+VC. LBNP and VC did not lower peak flow. However, O2 raised minimal resistance (2.3 +/- 0.4 RA; 2.8 +/- 0.5 O2+LBNP+VC, P < 0.04). When O2 alone (experiment 1) was compared with O2+LBNP+VC (experiment 2), no effect of LBNP+VC on peak flow or minimum resistance was noted, although the return rate of flow and resistance toward baseline was increased. O2 reduces peak forearm flow even in the presence of LBNP and VC.

Mercury determination by CV-AAS in wastewater and sewage sludge from a stabilization pond system.

The mercury concentrations in wastewater and sewage sludge of a stabilization pond system have been evaluated. The system is built by three parallel facultative ponds followed by two systems of three maturation ponds in series. The samples of wastewater and sludge were digested using nitric acid and placed into a Parr-type bomb for 4 h at 110 degrees C. Mercury was measured by Cold Vapour Atomic Absorption Spectrometry (CV-AAS) at 253.7 nm with sodium tetrahydroborate as reductant. The methodologies were checked with an USEPA quality control sample, a standard reference material from NIST and with another method of mineralization (cold mineralization) showing good results. Concentrations of mercury in wastewater between 1.47 +/- 0.75 microgl(-1) have been found at the entrance of the system and 0.74 +/- 0.0 microgl(-1) at the exit, while in sludge the results were between 0.29 +/- 0.12 microgkg(-1) in the facultative pond and 0.04 +/- 0.02 microgkg(-1) in the second maturation pond (exit).

Determination of anions in human and animal tear fluid and blood serum by ion chromatography.

An important factor contributing to the development of ion chromatography (IC) has been the need for repetitive analyses of samples with high ionic contents and samples available in microvolumes. IC was selected for the determination of Cl, NO3, SO(2-)4 and PO(3-)4 anions in tear fluid and serum from ten human volunteers of both sexes, seven young-adult black vultures (Coragyps atratus) and three young-adult chickens (Gallus gallus domesticus). The samples were analysed on a Dionex Model 2000i/SP ion-exchange chromatograph equipped with an anion guard column (Dionex IonPac AG4A), anion separator column (Dionex IonPac AS4A), suppressor column (Dionex AMMS-II) and a conductivity detector. The flow-rte of the mobile phase, 1.7 mM NaHCO3 - 1.8 mM Na2CO3 was set at 2.0 ml/min. The R.S.D. was calculated to be less than 1.5% for all anions. In the human, black vulture and chicken serum samples, the NO3, PO(3-)4 and SO(2-)4 anion contents were higher than in tears; for Cl the reverse was found. No correlation was found amongst the anion concentrations present in the tear fluid and blood serum in all samples (p > 0.05). With no sample treatment, column maintenance was required.

Conformational analysis and biological activity of leptocarpin and leptocarpin acetate.

This paper deals with the conformational study in solution of leptocarpin, a germacranolide with the heliangolide structure isolated from Leptocarpha rivularis D.C. The results obtained agree with the single crystal X-ray diffraction studies. Leptocarpin and leptocarpin acetate show 50% inhibition of [35S]-methionine uptake without affecting the (methyl-3H)thymidine and (5,6-3H)uridine incorporation, when assayed in HeLa cells.

Subcortical U-fibers layer preservation in brain edema.

The mechanism involved in the relative preservation of the subcortical U-fibers in the arcuate zone was studied in a post infarct edema after sagittal superior sinus occlusion. Superior sagittal sinus (SSS) of 36 mongrel cats were occluded by polymer injection. Immediately before the occlusion Evans-blue (EB) was administered intravenously. The cats were killed 1, 2, 3, 6, 12, 24, 72 and 120 hours after sinus occlusion. In 20 cats in which cortical veins were occluded, in addition to the SSS, EB was extravasated. In 9 of these cats, which had moderate edema, EB-staining was present only in the cortex. In 11 cats with severe edema, massive EB extravasation was observed also in the white matter. The U-fiber layer was free of EB, suggesting that the extension of edema was blocked by this zone. Our findings demonstrated that the U-fibers act not only as a resistance against extension of edema from white to gray matter, but also in a reverse direction. The characteristics of the spread of brain edema is not yet completely understood; both anatomical and biochemical peculiarities from its basis. Different morphological patterns in the astrocytic reaction as well as the U-fibers sector vascularization are important. To evaluate the role of each one of these factors in the preservation of subcortical U-fiber layer in brain edema further investigations should be done.

Identification of class I MHC regions which bind to the adenovirus E3-19k protein.

The adenovirus early region 3 glycoprotein E3-19k binds to and inhibits expression of class I major histocompatibility complex (MHC)-encoded molecules, which may help infected cells evade immune recognition. The role of specific regions of the class I MHC molecule in the interaction with E3-19k was evaluated using a series of HLA-A2.1-, HLA-A2 variant-, and HLA-B7-expressing cell lines. The monoclonal antibody (mAb) W6/32, which recognizes a monomorphic epitope on class I MHC molecules, readily co-immunoprecipitated E3-19k with HLA-A2.1 and 14 different HLA-A2 variant molecules that differ from HLA-A2.1 by single amino acid substitutions. Thus, no single residue tested in the regions of the class I MHC molecule that bind peptide or the T-cell receptor controls the binding to E3-19k. Additional immunoprecipitations performed with mAbs directed against well-defined epitopes on the surface of HLA-A2.1 revealed a dichotomy in the ability of the mAbs to co-immunoprecipitate HLA-A2.1 and E3-19k. The mAbs LGIII-220 (directed against the C-terminal end of the alpha 1-helix), CR11-351 (directed against the N-terminal end of the alpha 2-helix), and PA2.1 (directed against the middle of the alpha 2-helix and an underlying beta-loop) readily co-precipitated HLA-A2.1 and E3-19k. In contrast, mAbs MA2.1 (directed against the N-terminal end of the alpha 1-helix and the C-terminal end of the alpha 2-helix) and HO-2 (directed against the N-terminal end of the alpha 1-helix) did not co-precipitate E3-19k with HLA-A2.1. Similarly, mAb MB40.2 (directed against residues 169-182 of HLA-B7) also did not co-precipitate E3-19k with HLA-B7. These studies lead to the conclusion that the N-terminal end of the alpha 1-helix and the C-terminal end of the alpha 2-helix play an important role in dictating the ability of the E3-19k protein to bind to the class I MHC molecule.

Physiological variations in the pigmentation of newborn infants.

Skin pigmentation is a physiological feature of great value and usefulness from both an anthropological point of view and a clinical point of view. This article is a first approach to the study of the skin pigmentation of 126 full-term newborn infants (37-42 weeks gestational age) at the Maternity Hospital "La Paz" (Madrid, Spain) using the reflectance method. We provide the normal patterns, which have to be taken into account when applying this method to detect transcutaneous bilirubin and some physiopathological conditions, such as hyperbilirubinemia, anemia, plethora, and hormonal disturbances. Measurements of the reflectance were taken at the forehead, the internal and external aspects of the arm, the thorax, and the gluteus with blue, green, and amber tristimulus filters. With all filters the least pigmented zone was the thorax, and the most pigmented zone was the forehead; the internal aspect of the arm, the gluteus, and the external aspect of the arm were intermediate in increasing order. Our data confirm a higher pigmentation at birth than during the first year. We think that the idea of cephalocaudal progression of hyperbilirubinemia must be rejected. Our results also indicate that the thickening of the skin with gestational age is one of the most important factors in the skin color changes of newborn infants with an apparently different rhythm in males than in females.

MR imaging of benign prostatic hypertrophy using a Helmholtz-type surface coil.

MR examinations of the prostate were performed on six healthy volunteers and 18 patients with well-documented symptomatic benign prostatic hypertrophy, using an organ-encompassing Helmholtz-type surface coil at 1.5 T. The healthy volunteers were also imaged with a standard circumferential body coil. The morphologic features and signal intensity characteristics of the prostate and adjacent structures were analyzed in the patient group. Several recognizable patterns of benign prostate hypertrophy were identified including bilaterally symmetrical nodules in the central gland, multiple central gland nodules, and a diffusely heterogeneous central gland without appreciable nodules. The peripheral zone was of moderate to high signal intensity on T2-weighted images, and was diffusely heterogeneous in 78% of patients. The false prostatic capsule, peripheral venous plexus, and seminal vesicles were also characterized. A good correlation was shown between prostatic glandular volume and prostate-specific antigen. Calculated signal-to-noise ratios (S/N) were significantly greater on images acquired with the Helmholtz-type receiver coil than on those acquired with the body coil. We conclude that the hyperplastic prostate gland has a variety of MR appearances, but that recognizable patterns are frequently seen. High resolution imaging with a Helmholtz-type surface coil provides excellent anatomical depiction of the prostate and adjacent structures.

Role of the adenovirus E3-19k conserved region in binding major histocompatibility complex class I molecules.

The adenovirus early region 3 glycoprotein E3-19k binds to and down regulates major histocompatibility complex (MHC) class I molecules in infected cells. We previously identified a 20-amino-acid conserved region in E3-19k by comparison of protein sequences from four different adenovirus serotypes. The roles of the E3-19k C-terminal and adjacent conserved regions in the interaction with MHC class I molecules have been examined. A functional class I-binding glycoprotein was expressed from the cloned E3 18.5-kDa open reading frame of adenovirus type 35. Truncations and single-amino-acid mutations in the adenovirus type 35 glycoprotein were created by site-directed in vitro mutagenesis and tested for the ability to associate with MHC class I molecules. Deletion of most of the transmembrane domain and cytoplasmic tail did not affect binding to class I molecules. However, removal of an additional 11 amino acids eliminated binding and changed the conformation of the adjacent conserved region. Separate mutations of residues Asp-107 and Met-110, within the conserved region, severely reduced or eliminated binding. These data indicate that the E3-19k conserved region plays a crucial role in binding to MHC class I molecules.

Dynamic gadolinium-enhanced rapid acquisition spin-echo MR imaging of the liver.

Rapid acquisition spin-echo (RASE) magnetic resonance (MR) imaging allows for coverage of the entire liver with highly T1-weighted SE images during a single 23-second breath-holding period. The RASE sequence was implemented in conjunction with rapid intravenous injection of gadopentetate dimeglumine to enable performance of dynamic contrast material-enhanced MR imaging of the liver. Prospective evaluation of 24 patients with 62 liver lesions 1 cm or greater in diameter was performed. Images obtained with RASE were devoid of respiratory-related ghost artifacts or edge blurring. The dynamic contrast-enhanced RASE technique resulted in contrast-to-noise and contrast-to-artifact values and time efficiency measures significantly greater (P less than .05) than those obtained with use of conventional T1- and T2-weighted pulse sequences, indicating a higher likelihood for lesion detectability. Lesion conspicuity was maximal during or immediately following bolus administration of gadopentetate dimeglumine, with lesions often becoming obscured at delayed postcontrast imaging.

Normal abdominal enhancement patterns with dynamic gadolinium-enhanced MR imaging.

The objective of this study was to quantitatively and qualitatively determine contrast enhancement patterns of normal abdominal organs with dynamic gadolinium-enhanced magnetic resonance (MR) imaging. Dynamic gadolinium-enhanced, T1-weighted, spin-echo imaging was performed during a 23-second breath hold in 38 patients, with images acquired before, during, and at 1,2, and 5 minutes after bolus injection of gadopentetate dimeglumine. Enhancement patterns of normal liver, spleen, pancreas, adrenal gland, kidney, aorta, inferior vena cava, and fat were determined by visual evaluation and by performance of signal intensity measurements with an electronic cursor. Time-intensity curves demonstrated peak enhancement of all abdominal organs during or immediately after bolus injection of gadopentetate dimeglumine. MR enhancement patterns included visualization of renal cortical nephrogram and heterogeneous enhancement of the spleen during the bolus phase of contrast material administration. Peak enhancement of normal liver was 72%; spleen, 172%; pancreas, 82%; adrenal gland, 85%; and kidney, 291%. This study established reference data regarding abdominal organ enhancement that will be useful as dynamic gadolinium-enhanced MR imaging becomes clinically implemented.
