Olfactory experience modulates Bcl-2 expression in the developing olfactory bulb.

The effect of olfactory deprivation on cellular expression of the Bcl-2 gene in the olfactory bulb of young rats was investigated. Restriction of olfactory stimuli caused an overall increase in Bcl-2 mRNA expression, with increases seen in the lateral aspects of glomerular, external plexiform, mitral and granule cell layers, as well as the medial aspects of external plexiform layer. No differences were found in the unoperated control group. In addition, we found an inverse relationship between the incidence of apoptosis induced by olfactory deprivation and the magnitude of increase in Bcl-2 mRNA expression in the glomerular layer. These data raise the possibility that Bcl-2 may be involved in olfactory experience-related neural plasticity by regulating cell survival.

The capillary transport system for free fatty acids in the heart.

The nature of the process by which free fatty acids, which are tightly bound to albumin, traverse the endothelium of cardiac capillaries to reach the cardiac muscle cells, so that they are extracted to a net extent of approximately 40%, needs clarification. Previous studies have indicated that a membrane fatty acid-binding protein provides for carrier-mediated uptake of free fatty acids by isolated hepatocytes, cardiomyocytes, and jejunal mucosal cells. A monoclonal monospecific antibody was prepared against purified membrane fatty acid-binding protein from rat liver. Multiple-indicator dilution experiments were carried out in the isolated rat heart with labeled albumin, sucrose, and palmitate in the presence of control perfusate or perfusate containing either specific antibody or comparable nonspecific myeloma cell supernatant (each of the latter containing additional albumin, in identical concentrations). Analysis of the labeled-sucrose curves provided a permeability-surface area product for sucrose to which that for palmitate could be compared. In comparison with control supernatants, myeloma supernatant produced a minor inhibition of palmitate uptake, as a result of the increase in albumin concentration. The specific antibody, which contained identical albumin concentrations, produced a major inhibition of palmitate uptake, significantly greater than with the myeloma supernatant. The data indicate that the membrane fatty acid-binding protein mediates the transfer of free fatty acid across the endothelial cells of cardiac capillaries for presentation to heart muscle. Passive intramembrane lateral diffusion of palmitate could not provide an explanation for the findings.

Pattern of chronic liver disease in the eastern province of Saudi Arabia. A hospital-based clinicopathological study.

During the period 1982-1990, 544 patients with clinical evidence of liver disease were admitted to King Fahd University Hospital, Al-Khobar, Saudi Arabia. Besides routine laboratory and sonographic investigations, all were subjected to either a needle liver biopsy, laparoscopy or a laparotomy. The tissue diagnoses were as follows: liver cirrhosis 17.3%, periportal fibrosis 14.3%, metastatic cancer 12.9%, primary hepatoma (hepatocellular carcinoma: HCC) 12.1%, hepatic granuloma 11.2%, chronic active hepatitis 7.7%, chronic persistent hepatitis 2.2%, fatty liver 7.2%, hydatid liver disease 4.6% and others 2.8%. In 7.7% the histology was normal. These results will be discussed and compared with results reported in local and international literature.

Determination of the nucleotide sequence and chromosomal localization of the ATP2B2 gene encoding human Ca(2+)-pumping ATPase isoform PMCA2.

The plasma membrane Ca(2+)-pumping ATPase (Ca(2+)-ATPase) is responsible for maintaining calcium homeostasis in eukaryotic cells. The Ca(2+)-ATPase is a family of pumps that are encoded by at least four genes. A cDNA for the human version of Ca(2+)-ATPase isoform PMCA2 was isolated and characterized. Comparison of the human and rat cDNA sequences showed that they were 95% homologous in the coding domain, and this homology was reflected in the deduced protein sequence where greater than 98% homology between the human and rat sequences was found. The amino acid differences that were found were almost all conservative. The PMCA2 cDNA was used to probe Southern blots of human-rodent somatic cell hybrid DNAs; the results indicated that the human PMCA2 gene was located on chromosome 3.

Tumor localization of newly developed hematoporphyrin (DHP) using a bladder tumor model: a novel hematoporphyrin derivative.

Application of laser irradiation with porphyrin(s) or their derivatives for the destruction of tumors in humans requires preliminary studies of their localization in normal and malignant tissues. A novel derivative of hematoporphyrin (HP) was prepared. The newly developed hematoporphyrin (DHP) was administered to Fisher rats with bladder tumors and showed greater accumulation in the tumoral tissues. Comparative data on (HP) and (DHP) are presented and discussed in light of the enhanced tumor porphyrin uptake caused by these agents. The homogeneous intense fluorescence noted with DHP-treated animals suggests that total tumor kill curative therapy will be more feasible. The study paves the way to refining increased porphyrin augment phototherapy and laser application in the field of oncology.

Skin testing in an allergy affected population of Saudi Arabia.

Two hundred and ten (210) patients attending the Allergy Clinic of King Fahd Hospital, KFU, with a history of allergy related problems were tested for skin sensitivity. A total of 52 allergens were used. Only 55 patients were found to be positive to one or more of the allergens. House dust and inhalants were the most common allergens. Sixty percent (60%) of the patients had elevated IgE levels though all had evidence of allergy-related clinical disease. Clinical history appeared to be the most sensitive indicator of allergic problems. Skin testing and IgE levels were less sensitive.

Neuroepithelial (colloid) cyst of the third ventricle in identical twins. Case report.

Colloid cysts of the third ventricle are described in middle-aged twin brothers. One of them presented with recurrent attacks of headache. In this patient the cyst had reached a size large enough to obstruct the cerebrospinal fluid pathway, resulting in hydrocephalus. The twin brother, although asymptomatic, was suspected of the anomaly and investigated because of the similarity of his ocular signs. The diagnosis was confirmed by computerized tomography in both the patient and his brother. The latter proved to have a smaller colloid cyst situated anteriorly in the third ventricle with no obstructive hydrocephalus. The patient was successfully operated on, while the brother is still under observation. Both brothers have had bilateral cataracts, retinal detachments, and left lateral rectus palsies. The familial occurrence of colloid cysts and their association with these ocular findings have apparently not been described before.

[Radiographic signs of radiolesions of the pelvis (excluding the femur) after irradiation for epithelioma of the cervix uteri (author's transl)].

Radiolesions of the pelvic bones (excluding the femur) were observed in 29 patients after irradiation therapy for cervix uteri cancer. Three regions can be affected corresponding to the areas irradiated: the sacrum and internal part of the iliac wings, the pubis, and the cotyles. In most cases the lesions appeared after 1 to 4 years. Their radiological appearances are characteristic, the principal sign being irregular bone condensation in a demineralized bone, without true cavitation. The only lytic lesions observed were in the pubis. Calcification of soft tissues may occur and fractures are frequent. There is a slow progression of the lesions over long periods. Clinical, radiological, and progression signs differentiate radiolesions from other affections: metastases, invasion by contiguity, infections, and radio-induced sarcoma.

[Evolution of the dosimetric criteria of the curietherapy in carcinoma of uterine cervix (author's transl)].

The difficulties which arise when comparing the different therapeutic attitudes concerning the treatment in the curable stages of cancer of the uterine cervix have led us to the formation of dosimetric criteria to be employed in the analysis of the intracavity therapy. We have established points of reference which can be utilized irregardless of the intracavitary technique therapy. Furthermore, we have analysed those patients who have received the same dose of external irradiation in an effort to establish as the only variable the intracavitary treatment. Forty patients, receiving 4,000 rads tumor dose in 4 weeks, were reviewed and demonstrated a correlation between the reference volume of the intracavitary treatment (as defined by the three dimensions W, H, T) and eventual complications (4 cases of Rectitis 1). We now feel that with the utilisation of these dosimetric criteria we will be able to: minimize the risk of complication without augmenting the indicence of local-regional failures; facilitate the objective comparison of the efficacity of the different intracavitary techniques presently employed at the major treatment centers.
