Electron microscopical investigation of citrate lyase single molecules.

Electron micrographs of citrate lyase from Rhodopseudomonas gelatinosa and Klebsiella aerogenes reveal two characteristic molecular forms. The "basket" form and the "star" form were subjected to two-dimensional image reconstruction using a technique involving averaging of superposed single molecular images after rotational correlation. A three-dimensional image reconstruction shows that the images of these forms can be interconverted by rotation and that they therefore represent different views of the same structure.

[Status of fibrinolysis in systemic lupus erythematosus].

In patients with systemic lupus erythematosus (SLE) both a haemorrhagic diathesis and a tendency to thrombosis of the venous and arterial vessels can be observed. In the course of the disease, thrombosis of the leg or pelvic veins developed in 20 per cent of 188 patients. The levels of alpha 2-plasmin inhibitor, plasminogen, fibronectin and of factor VIII complex were increased in patients with SLE compared with a control group. Fifty per cent of the patients showed no increase in fibrinolytic activity after venous occlusion measured with the fibrin plate method. This suggests a reduced fibrinolytic capacity in SLE probably caused by alteration of the endothelial cells through immune complex vasculitis. In addition, the lupus anticoagulant and an acquired antithrombin III deficiency in nephrotic syndrome in SLE are to be considered thrombophilic mechanisms. In the individual case there is an overlapping of hyper- and hypocoagulability.

[Thrombocyte counting with the Laborscale (PSL-1/PSA-1) particle-counting instrument by Medicor (Budapest/Hungary) and thrombocyte volume analysis in the diagnosis of selected thrombocytopathies].

A thrombocyte counting technique is presented in comparison with the chambre counting according to the German Book of Medicaments (DAB 7 D.L.) GDR on the particle counting device "Laborscale" (PSL-1/PSA-1) of Medicor by using a thrombofuge (70 g). The composition of reagents accounts for problems of measurement which are caused by the device and influences the separation of particles. The possibility of recording the curve of thrombocyte volume distribution and calculating the average thrombocyte volume (VM) is demonstrated by means of a normal collective of test persons as well as by intravascular disorders of haemostasis and during cytostatic therapy.

Protein and glycoprotein abnormalities in an unusual subtype of Glanzmann's thrombasthenia.

A patient with Glanzmann's thrombasthenia has been analysed by high resolution two-dimensional polyacrylamide gel electrophoresis and crossed immunoelectrophoresis. Although clot retraction is subnormal, glycoproteins IIb and IIIa are completely absent and platelet fibrinogen is strongly reduced indicating an unusual subtype of type II thrombasthenia. No changes in the pI of glycoproteins have been observed. Heterogeneity in Glanzmann's thrombasthenia is discussed.

[The ultrastructure of megathrombocytes in May-Hegglin's anomaly].

A clinical and extensive electron microscopic study was made of the circulating granulocytes and thrombocytes from three female patients of a family with May-Hegglin anomaly. There have been observed inclusion bodies (Döhle-bodies) in a high proportion of polymorphonuclear leukocytes as well as a thrombocytopenia and giant platelets (so-called megathrombocytes). Other family members did not show hematologic abnormalities. The investigated giant platelets show ultrastructurally a different electron density of the alpha-granules, however, these structures present no typical formation of "bull's eye granules" which has been observed in other cases of May-Hegglin anomaly. Peripheral or central situated net-like structures of the endoplasmic reticulum have been found frequently. There were seen no microtubuli whereas glycogen particles were scattered within the peripheral cytoplasm.

[Gout in the age (author's transl)].

The importance of the gout is growing in the GDR as its frequency has been increasing since the sixties. The gout is a disease of metabolism with the following accompanying phenomena: renal lesion in gout, hypertension, cardiac diseases and peripheral arterial diseases. Besides, there are proved relations between hyperuricemia and obesity, hyperlipoproteinemia, diabetes mellitus as well as steatosis hepatis. In describing the nature of the gout the peculiarities of age are stressed. The treatment of the gout depends on the clinical state.
