[A novel immunoregulating hexapeptide with antineoplastic effect].

A synthesized analog of myelopeptide HP-2-->M[symbol: see text]-2 (Leu-Val-Val-Tyr-Pro-Trp) caused a significant (60-80%) and prolonged inhibition of s.c. grafted tumors P388, Ca-755, B-16 and sarcoma 180 in isogenic mice but did not affect the growth of tumor B-16 in nude mice. Nor did it influence proliferative activity or viability of cultured human tumor cells. The best results were obtained with s.c. injections of 0.5-2 mg/kg HP-2-->M[symbol: see text]-2, twice or trice a day, at 96 hr intervals. No symptoms of severe poisoning were registered at doses of HP-2-->M[symbol: see text]-2 100 times the therapeutic one. A pharmacokinetic study in mice revealed prolonged circulation of HP-2-->M[symbol: see text]-2 in blood and a high affinity for the bone marrow (t 1/2 (130.1 hr and 431.6 hr, respectively). HP-2-->M[symbol: see text]-2 restored in vitro the ascites P388-suppressed cytotoxicity of murine T-lymphocytes. HP-2-->M[symbol: see text]-2 is regarded as a candidate for clinical studies of its potential of immunocorrection in cancer patients suffering T-lymphocyte immunity disturbances.

[Dextran-ferrite distribution in the body of animals in a nonuniform permanent magnetic field].

Dextran-ferrite (DF) was injected via the femoral artery to the dogs and followed by local magnetization (induction 0.3 T, grade 0.024 T/cm). It has been shown that the local retention of the iron in some tissues of the leg was 3-5 times as high as the control. The magnetization did not increase the local retention of the iron after intravenous injection of DF to mice and rabbits.

[Study of heterogeneity of the chromatin endonuclease fragment by free flow electrophoresis].

The oligomer chromatin fragments relatively uniform in size (8--11 nucleosomes) were prepared by a short-term endonycleolysis. The heterogeneity of these fragments with respect to their electrophoretic mobility was revealed using free flow electrophoresis. The individual fragmentated chromatin subfractions were obtained. These subfractions differed in their protein and RNA content per DNA weight unit, in quantitative ratios of different zones of high molecular weight non-histone proteins and in thermal and alkaline denaturation kinetics. It was also found that the parameters investigated are correlated with electronegativeity of the fragmentated chromatin subfractions.

[Study of heterogeneity of chromatin endonuclease fragments, using fractionation by step-wise endonucleolysis].

Isolated nuclei from rat liver were incubated at different time intervals under conditions, optimal for manifestation of the Ca, Mg-dependent endonuclease activity. After each of the 6 endonucleolyses chromatin was extracted and 6 chromatin fractions (I--VI) and "residual" chromatin were collected. A comparative analysis of the "early" (I--III) and "late" chromatin fractions revealed an increased RNA content in the "late" fractions and changes in the composition of the non-histone proteins. Electrophoresis in acrylamide gel concentration gradient demonstrated that fractions I--III predominantly contain high molecular weight fragments whereas fractions IV-VI are represented by highly fragmented chromatin. Each fraction was characterized by peculiar shapes of alkaline denaturation curves and by heterogeneity of charges of their constituent chromatin fragments.
