[Polarographic analysis of cobalt in biological specimens].

References to different methods of cobalt detection in biologic material are given. Polarographic detection of cobalt in human urine and animal biologic material worked out by authors was used for evaluation of biologic MAC. Biologic material was limed by nitric and sulfuric acids. Sulfuric acid was carefully distilled off the burnt material. Mineralized material was dissolved in 10 ml of 1N hydrochloric acid, filtered and again steamed into moist salts. 2.5 ml of water, 0.5 ml of dimethylglyoxime (0.02 M), 2 ml of Na4P2O7 (0.2 M) and 5 ml of buffer solution (0.1 M NH4Cl and NH4OH) were added to limed mineral, than pH was driven to 9 by 1 = 2 drops of ammonia solution and after blowing through, the specimen was exposed to polarography in alternating current. ++Redox potential of cobalt is 1.18 V. The method can be used for detection of cobalt in urine of workers contacting with the stated metal. Sensitivity of the method is 0.02 Mkg/ml.

Toxicokinetics of cobalt and the problems of biological monitoring.

Toxicokinetics of cobalt was studied on rats by the method of radioactive isotopes using single or repeated administrations. Co was used in the form of sulphate in doses of 0.1 and 0.2 mg/kg administered by 4 different routes (intragastric, intraperitoneal, intratracheal, subcutaneous). Good absorption was established in the parenteral routes of administration, the regularities of absorption, retention in the internal organs and elimination of cobalt were determined and mathematically expressed. Clear dependence was established between the doses administered and the content in biomedia. The liver and the kidneys were found to be the organs of highest accumulation of cobalt. Elimination half-lives and the time of establishment of a constant level during long-term exposure were determined. It is proposed to carry out diagnostic examination in the course of a few hours after the work shift. Urine and blood are the test objects in which direct correlation has been established. Delayed elimination of cobalt from the organism under protracted exposure may be the essential cause of increased hazard for man, which conditions the necessity of determining maximum acceptable concentration for this substance.

Vanadium--toxicity, metabolism, carrier state.

By using the method of radioactive isotopes on albino rats, we studied the rate and degree of absorption of vanadium in to the blood, the time of its retention in the organs and the kinetics of its excretion, the influence of stable vanadium on the generative function and the correlation between the carrier state in workers who are in contact with vanadium in its production and the levels of exposure. Varying rates of absorption in to the blood in dependence on the route of administration, accumulation of vanadium in the kidneys, bone tissue, liver, lungs, placenta, testicles and the mammary gland, predominant elimination by the kidneys during the first two days, the gonodotoxic and embryotoxic effects were established. Correlation was revealed between the content of vanadium in the urine of workers and the levels of exposure.
