Augmented production of gamma interferon in mice experimentally infected with Toxoplasma gondii.

The gamma interferon (IFN-gamma) production and the cell populations participating to this production were examined in Toxoplasma-infected mice. When spleen cells from Toxoplasma-infected mice were cultured with Concanavalin A (Con A) or OK-432, a Streptococcal preparation, they produced significantly high levels of IFN-gamma as compared with that of noninfected mice. Such enhanced IFN titers were observed as early as at 5 days postinfection, reached at the maximum levels on 20 days around and declined gradually thereafter. Treatment of spleen cells from the infected mice with either monoclonal anti-Thy-1.2 antibody plus complement or macrophage-blocking agents virtually abolished the IFN production. The spleen cells producing IFN-gamma were more susceptible to the treatment with monoclonal anti-Lyt-1.2 than anti-Lyt-2.2 antibodies, suggesting that CD4+ T cells are main producers of this lymphokine. When mice infected with Toxoplasma 10 days previously were injected with lipopolysaccharide (LPS), a well-known inducer of IFN-alpha/beta, the sequential production of IFN-alpha/beta and IFN-gamma was induced in their circulation.

Ovarian wedge resection with contact Nd:YAG laser irradiation used laparoscopically.

Twelve women with polycystic ovary disease were treated with Nd:YAG laser contact irradiation using a cutting-type probe. Bilateral ovarian wedge resection was performed in every case. After undergoing the procedure, seven patients achieved pregnancy, and another three experienced the return of a normal menstrual cycle. Serum testosterone and androstenedione levels were decreased postoperatively, while luteinizing hormone (LH), follicle stimulating hormone (FSH) and estradiol were increased. It seems that the decrease in intraovarian androgens might have reduced the negative feedback system and thus might have increased the secretion of gonadotropin and estradiol, which in turn promoted ovulation induction and pregnancy.

Nd:YAG laser therapy for infertility with a contact-type probe.

Laparoscopic Nd:YAG laser surgery for tubal adhesiolysis, hydrosalpinx and other disorders is an effective treatment for female infertility that causes relatively little tissue damage. In initial studies using contact-type probes, the 0.4-mm-diameter probe and 10 mm/sec incision speed were found to cause less tissue degeneration than did other combinations (P less than .05). Fifteen infertile women were treated at laparoscopy with a Nd:YAG laser; one of six with hydrosalpinx achieved pregnancy after salpingostomy using a contact-type probe, two of three with tubal adhesions achieved pregnancy after adhesiolysis with the Nd:YAG laser, and three of six with polycystic ovaries achieved pregnancy after wedge resection using a contact-type probe. Five of fifteen infertile women conceived after Nd:YAG laser surgery with a contact-type probe under laparoscopy. We confirmed that contact irradiation with a ceramic incising probe is capable of creating an adequate incision at low power levels and that laparoscopic Nd:YAG laser surgery for infertile women is an effective treatment.

An immunoglobulin G inhibiting lactate dehydrogenase activity.

We found extremely low (6 U/l) serum lactate dehydrogenase (LDH, EC 1.1.1.27) activity in a patient with uterine myoma. The patient's serum inhibited purified LDH isoenzymes. One milliliter serum neutralized 73 U purified LDH-1 isoenzyme, equivalent to 300-500-fold the amount of LDH in 1 ml of normal serum. The serum inhibitor was purified by ordinary procedures using chromatographies and identified as IgG which contains both kappa- and lambda-chains. The IgG is very unique in showing higher affinity to the H- than M-subunit of the LDH tetramer, in contrast with the IgGs already reported. After removal of the myoma, the anti-LDH activity gradually decreased with a half-life of 20 days corresponding to that of IgG and finally almost disappeared. This indicates a possibility that the myoma cells produce some factors such as B-cell growth and differentiation factors.

Immunological effects of mouse ceruloplasmin.

Mouse ceruloplasmin, the level of which in serum increases on administration of antitumor polysaccharides, was shown to have enhancing effects on the antibody production of mouse spleen cells toward sheep red blood cells and restorative effects on the suppressed mixed lymphocyte reaction of spleen cells of tumor-bearing mice. The proliferation of interleukin-2 dependent cytotoxic T cells and the suppressed concanavalin A response of the tumor bearing mice spleen cells were also enhanced by this serum glycoprotein, weakly but reproducibly. Furthermore, this serum glycoprotein was found to enhance glucose consumption and interleukin-1 production and cytotoxic activities of mouse peritoneal macrophages. These results suggest that mouse ceruloplasmin plays some roles in the natural resistance surveillance against tumors.

Identification of a mouse serum protein increased by administration of an antitumor polysaccharide, PSK, as a variant of mouse transferrin and some of its biological activities.

One of the so-called LC components, the content of which is increased in the serum of PSK (antitumor polysaccharide) treated mice, was purified by repeated ion-exchange column chromatography on DEAE-Sephadex A-50. The purified protein, designated as LC-2, was identified as a variant of mouse transferrin, which is a serum beta 1-globulin having an iron-binding capacity. The absorption spectrum and ultraviolet circular dichroism (CD) curve of LC-2 were identical with those of mouse transferrin. The molecular weight, isoelectric point and amino acid composition of LC-2 were in good agreement with the results obtained previously for mouse transferrin. However, the carbohydrate content of LC-2 was different from that of mouse transferrin. This protein restored the depressed spleen cell response of tumor-bearing mice to concanavalin A (Con A) and promoted the metabolism of proteose-peptone induced peritoneal macrophages obtained from tumor-bearing mice. Furthermore, it had weak but definite antitumor activity against Sarcoma 180 cells in vivo.

[Preclinical study of endoscopic laser coagulation of human ovaries].

The hemostatic potential of YAG laser irradiation was investigated in 21 hysterectomy and adnexectomy patients of childbearing age. Endoscopic forceps were used to induce hemorrhaging in the ovarian capsule. The hemorrhage was then irradiated from a distance of 2cm using a series of 1-sec. irradiations at either 30W or 50W until complete hemostasis occurred. In the 30W group, hemostasis was achieved after a total irradiation of 4.1 +/- 1.6 sec. while a significantly shorter time of 2.5 +/- 0.8 sec. was required in the 50W group (p less than 0.01). The irradiation energy in the 30W and 50W groups was 107 +/- 45 J and 109 +/- 35 J, respectively. Tissue temperature was measured during laser irradiation at 5mm and 10mm from the center of irradiation at a tissue depth of 5mm. In the 30W group, the tissue temperature at the 5mm and 10mm points increased 6.6 +/- 2.0 and 2.5 +/- 1.1 degrees C, respectively, while the temperature in the 50W group increased 8.5 +/- 3.5 and 3.1 +/- 0.9 degrees C respectively. No remarkable histological changes were noted in either group following irradiation. YAG laser irradiation during endoscopy is therefore considered to be a highly effective hemostasis for minor hemorrhages caused by instrument manipulation during endoscopic surgery.

Elevation of colony stimulating factors in mouse serum after injection of PSK, an antitumor polysaccharide.

The level of colony stimulating factors (CSF) in mouse serum was elevated by a single intraperitoneal injection of PSK, a host-mediated antitumor protein-bound polysaccharide obtained from Basidiomycetes. The assay for CSF activity was performed by employing a semisolid methylcellulose culture method using mouse bone marrow cells, and the activity was estimated by an equation well matched with the dose response curve obtained in the CSF assay. A temporary increase of CSF activity within 10 h after the injection of an antitumor polysaccharide, Krestin (PSK) (250-1000 mg/kg) was followed by a fast decline in the activity, but no significant increases were detected in the cases of 62.5 and 125 mg/kg PSK injections. The CSF activity in PSK (500 mg/kg)-treated mouse serum was separated into two active fractions by DEAE ion-exchange chromatography, and both fractions were found to induce colonies comprised of cells with the properties of macrophage in regard to morphology, cytochemistry of non-specific esterase, phagocytic function and expression of Fc receptors on the cell surface. The elevation of the serum CSF level due to administration of so-called host-mediated antitumor agents might be one of the tumor-defense mechanisms in vivo.

A day care program and evaluation of animal-assisted therapy (AAT) for the elderly with senile dementia.

We conducted a survey to clarify the evaluation methods of animal-assisted therapy (AAT) for the elderly with senile dementia in an adult day care center. AAT was implemented for a total of six biweekly sessions. The AAT group consisted of seven subjects and the control group numbered 20 subjects. In a comparison between Mini-Mental State Exam (MMSE) scores at baseline and those measured three months later, the average MMSE score before AAT (baseline) was 11.43 (+/- 9.00), and three months later it was 12.29(+/- 9.69). In the AAT group, the average score on Nishimura's Activities of Daily Living (N-ADL) at baseline was 28.43(+/- 14.00), and after ATT it was 29.57(+/- 14.47). In the AAT group, the average baseline score on behavioral pathology of Alzheimer's disease (Behave-AD) was 11.14(+/- 4.85), and three months after AAT it was 7.29(+/- 7.11) (p < 0.05). In the control group, the average baseline score was 5.45(+/- 3.27) and three months later it was 5.63(+/- 3.59). The evaluation of salivary CgA, as a mental stress index, showed a decreasing tendency in the AAT group. Our findings demonstrate the usefulness of using several methods for evaluation of the changes in patients given AAT.
