Suppression of transformed phenotype in hybrids of v-fgr and v-raf transformed rat-1 cells with rat embryonic fibroblasts is due to transcriptional inactivation of viral oncogenes.

Rat-1 cells that had been transformed to tumorigenicity by transfection with the retroviral oncogenes v-raf from 3611-murine sarcoma virus, or v-fgr from Gardner-Rasheed feline sarcoma virus were fused with rat embryonic fibroblasts at an early passage. In both fusion experiments hybrid cells were isolated that exhibited normal morphology, anchorage requirement for proliferation, and either no tumorigenicity (v-fgr) or extended latency periods for tumor growth (v-raf) in nude mice. Transcription of viral oncogenes is drastically reduced in hybrid cells (at least 30-fold compared to their transformed parental cells), while the half-life of the corresponding transcripts is not effected. In the chromatin of hybrid cells the integrated retroviral oncogenes are as sensitive to degradation with pancreatic DNase I as the endogenous actin gene. Thus the observed down regulation of proviral transcript levels does not correlate with changes in chromatin structure. We conclude that in hybrids of (v-fgr)- and (v-raf)-transformed Rat-1 cells with embryonic fibroblasts, transcription of the retroviral oncogenes appears to be repressed by trans-acting factors of the normal parental cell.

[Rare site of malignant melanoma].

Primary melanoma of the vagina is a rare tumor associated with a poor prognosis. We report on a 82 years old woman with a malignant melanoma of the vagina. The treatment performed was the removal of the vaginal tumor and a hemivulvectomy followed by a chemotherapy over 6 month. A recurrence of the tumor and the development of metastases in the brain and in the lungs was observed 3 weeks after the end of the chemotherapy. The patient expired 10 month after the primary treatment.

[Clinical experiences with treatment of granulosa cell tumors].

Symptoms, tumor morphology and clinical course were reviewed in 15 cases of granulosa cell tumors of the ovaries. The main symptoms were irregular menstrual bleedings. In 2 cases were found signs of glandular cystic hyperplasia additionally and in one case an adenocarcinoma of the endometrium. The appropriate treatment was the hysterectomy with bilateral salpingo-oophorectomy performed in most of the cases, followed by additional irradiation and polychemotherapy in 4 cases each.

[Experimental comparative study on investment materials for casting high-melting dental alloys].

The physical properties of 12 investment materials produced in middle- and east european countries were tested with methods as usual in experimental practice. At that the following properties were evaluated: grain-size distribution by screen-analysis, setting behavior by penetration, setting and thermal expansion, primary and secondary hardness and porosity by water-absorption. On the basis of the results the tested materials are valuated.

[The influence of different electrolytes on the electrochemical polarization behaviour of dental amalgams].

The measurements of electrochemical polarization of dental amalgams was studied in 5% sodium chloride aqueous solutions, in an artificial saliva and in natural saliva with different pH-values. The non-gamma-2-amalgams have been reported to have higher corrosion resistance in comparison to gamma-2-phase containing amalgams. The results show that the electrochemical corrosion behaviour is strongly differentiated using natural saliva.

[Biological evaluation of In-Ceram-ceramics compared to cobalt-base-alloys and the metals titanium, tantalum and niobium in animal experiments].

The aim of the study was to evaluate the biocompatibility of the in-cream-ceramic-system in several technological phases by animal experiments. The comparative materials were the Co-Cr-alloys Remanium CD and Wirobond and the implant materials Titanium, Tantalum and Columbium and the high biocompatible material Teflon. The experiments were carried out in accordance with the DIN-standard 13,930 using subcutaneous implantation to verify the toxicity of the materials. 20 cylindrical test pieces were implanted into the subcutaneous connective tissue of the neck in rats. After 12 weeks the animals were sacrificed and the implants were removed with the surrounding tissues. The thickness of the fibrous capsule and the number of cells within the capsule were measured microscopically. The in-ceram-ceramic-system showed the same results like approved Co-Cr-alloys. The results of the implant materials were not attained.

[Determination of prolactin concentration in endometrium in the histologically normal secretory phase and in delayed transformation].

Endometrial biopsies of 92 infertile women were histologically investigated. In 46 cases the endometrium was in-phase. We found tissue prolactin by RIA examination of the endometrium on day 15 of the menstrual cycle and later. A plateau of the prolactin tissue concentration was seen between the 20th and 26th day of the cycle. A marked increase of the hormone concentration is seen at the end of the luteal phase. The endometrial prolactin tissue content was characterized by a large individual variation. A luteal phase defect was observed in 40 cases. There was a later rise of tissue prolactin concentration decreasing in contrast to the normal luteal phase at the end of the cycle. The difference was statistically significant at day 20, 26 and 28 of the menstrual cycle. Six biopsies were not included in the study. The estimation of endometrial prolactin in the late luteal phase appears to be an additional diagnostic test in case of a luteal phase-defective endometrium.

[Rare type of metastases in progressive cervix carcinoma].

There are demonstrated two cases of advanced cervical carcinoma with unusual localization of metastases. One patient showed a scalp metastasis and the other one had a ulcerated inguinal lymph node metastasis. X-ray therapy caused a sufficient remission of the metastatic tumors.

[Studies on the oral health status of elderly and old people in a rural district].

There is reported on an epidemiological study to estimate the gerostomatological treatment need by examining in a rural district all people older than 55 years. The studies were carried out in compliance with the WHO-Basic Methods for oral health services. The results demonstrate that the oral health status of the old people is determined by loss of teeth and its functional consequences. Caries, diseases of the oral mucosa and TMJ-disorders are unimportant in the epidemiological point of view. The condition of porsthetic treatment is quantitative satisfactory in the age-groups up to 75 years.

[Comparative study on the biological properties of plastics for temporary crowns and bridges].

Six different plastic materials for temporary crowns and bridges were tested comparatively concerning their biological properties. To this the temperature in the pulp cavity was measured in vitro while making temporary crowns. Also the cytotoxicity was determined by the hemolysis-test. The results obtained showed that there is no thermal irritation of the pulp if the temporary plastic material is applied in an alginate impression as usual in practice. Cytotoxicity of the hardened plastics was not provable. Nevertheless tissue irritation can not be justified when using polymethacrylates and epimine-resin.

[Maintenance of teeth by a preventive strategy of prosthetic care].

As a result of analyses of the oral health conditions and prosthodontics the demand for a preventive strategy is opposed to the traditional treatment. Its primary goal is not completing the rows of teeth in number, but structural conservation using prosthetic appliances. The possibilities are discussed by examples given by crown restorations, initial loss of teeth, interrupted and shortened row of teeth. The results of clinically controlled studies are presented for the assessment of the preventive effectiveness.
