Chromium levels in potable water, fruit juices and soft drinks: influence on dietary intake.

Potable water, fruit juices and soft drinks are some of the most widespread beverages in the habitual diet, and they can contribute to chromium dietary intake. We determined the concentration of chromium in 90 different samples of beverages widely consumed in Spain. Graphite furnace atomic absorption spectrometry was used to analyze samples processed with a HNO3-V2O5 acid digestion pretreatment. In water samples Cr was directly determined. We verified the sensitivity, accuracy and precision of the method and ruled out matrix interferences. In analyzed samples, chromium values ranged from not detectable to 11.80 micrograms/l in potable water, from not detectable to 17.60 micrograms/l in fruit juices and from 3.60 to 60.50 micrograms/l in soft drinks. The chromium levels we encountered are low and the contribution of non-alcoholic beverages to dietary intake of this element, have been estimated to be 0.41 microgram/day in the common Spanish diet.

Trace element determination in different milk slurries.

We have studied the contents of trace elements of nutritional or toxicological interest in 90 samples of whole, low-fat, skim, condensed, evaporated and powdered milks. Slurries of the samples were prepared with Triton X-100 and analysed using electrothermal atomic absorption spectrometry. The temperature-time programme of the graphite oven was optimized for each element, and the accuracy, precision, selectivity and sensitivity of the method were verified. Concentrations of the trace elements we investigated were: Pb 0-0.211 microgram/g, Cd 0-28.985 ng/g, Al 0.528-4.025 micrograms/g, Cu 0.041-0.370 microgram/g, Cr 0-0.177 microgram/g, Mn 0.024-0.145 microgram/g, Se 0-23.333 ng/g, Zn 0.297-0.827 microgram/g and Ni 0.058-1.750 micrograms/g. (A value of zero indicates that the element was undetectable by our methods.) Concentrations of the pairs of elements Cu-Cd, Mn-Cd, Mn-Cu, Zn-Mn, Ni-Cu, Ni-Mn and Ni-Zn were significantly correlated (P < 0.001). Linear discriminant analysis confirmed the separation between the six types of milk analyzed.

Molecular characterization of Brazilian infectious bursal disease viruses.

A reverse transcriptase-polymerase chain reaction (RT-PCR) procedure was used to amplify a VP2 gene fragment (248 bp) from infectious bursal disease virus (IBDV). The procedure allowed the detection of known IBDV strains from the United States, along with field isolates and commercial vaccines produced in Brazil. Amplified VP2 fragments were further characterized by restriction fragment length polymorphism (RFLP) analysis. From 55 Brazilian commercial flocks, 48 field samples were IBDV positive by RT-TCR. Vaccine RFLP patterns were found in 12 flocks, a pattern compatible with classic IBDV in one flock, four new patterns in 31 flocks, and a pattern compatible with very virulent (vv) IBDV in four flocks. Sequence analysis showed that the vvIBDV RFLP patterns were closely related to the vvIBDVs described in Europe and Asia. Phylogenetic analysis of the four new RFLP patterns showed that they were closely related to but distinct from other classic, variant, and vvIBDVs, suggesting a high prevalence of different IBDV strains in Brazilian commercial flocks.
