Visualization of male and female characteristics of bovine fetuses by real-time ultrasonics.

Sexing of bovine embryos can enhance breeding programs at a rather early stage of pregnancy. The accurate predictability of the sex of the fetus can, for example, speed up breeding programs and allows an early selection of pregnant heifers for certain purposes according to the sex of their fetuses. Ultrasonic images were produced by a real image-producing sector scanner that was inserted per rectum into pregnant cows and heifers. Scrotal swellings and mammary teats were recognizable, allowing sexing of the fetuses between gestational day 73 and 120. The tests carried out showed an accuracy of 94%. Forty-nine of 52 ultrasonic diagnoses were confirmed as correct by subsequent calvings.

[Necessity for ECG triggering in digital subtraction angiography. Construction of a simple trigger device].

Movement unsharpness during digital subtraction angiography of the aortic arch and supra-aortic vessels can be avoided by means of ECG gating. The usual techniques resulted in exposures during the phase of least cardiac movement. The relatively long exposure time of about 60 to 300 msec caused marginal blurring during the exposure. An ECG-triggered device is described which produces short exposure times for the mask and filled frame during identical cardiac phases. An example with and without ECG gating shows marked improvement in detail rendering. The design of an ECG-triggered device with digitally adjustable delay and freedom from interference is described.

Human Ile-Ser-bradykinin, identical with rat T-kinin, is a major permeability factor in ovarian carcinoma ascites.

Ascites from patients with metastatic ovarian carcinoma contains high amounts of an activity that increases vascular permeability, as easily detected by a rat skin test. Ascites was fractionated by gel permeation and reversed-phase high-performance liquid chromatographies. The fractions were analysed for permeability-increasing activity. In this way a peptide was isolated and identified as Ile-Ser-bradykinin by sequence and amino-acid analyses. It is identical with T-kinin which has previously been detected as a product of an acute-phase protein, T-kininogen, in rats but never in human material. The so far identified human kininogens, i.e. high- and low-molecular mass kininogens, can only release Met-Lys-bradykinin or its degradations products, as Ile-Ser-bradykinin is not a part of their structure. However, the present results provide evidence that the permeability factor Ile-Ser-bradykinin under certain conditions can be produced in considerable amounts also by human tissues.

Calmodulin binding proteins in human erythrocyte membranes.

Human erythrocyte membranes reveal different calmodulin-binding proteins determined by a 125I-calmodulin gel overlay procedure. Beside the well-established Ca2+-transport ATPase, other proteins (205, 91, 72 and 42 kDa) bind calmodulin in a Ca2+-dependent manner. Two proteins of the human erythrocyte membrane are able to bind calmodulin only in the absence of Ca2+. One of them (76 kDa) is probably an integral, the other (240 kDa) a peripheral protein.

[Animal experiment and cell biology study of Saccharomyces cerevisiae Hansen CBS 5926 in the non-specific enhancement of resistance to infection].

In an animal experimental study the effect of Saccharomyces cerevisiae (S.c.) Hansen CBS 5926 (Perenterol) on bacterial infections has been investigated using different forms of application. A significant protective effect after oronasal, intragastric and subcutaneous application of S.c. Hansen CBS 5926 has been found by challenge with Streptococcus pneumoniae and Escherichia coli. In order to investigate the possible mode of action the effect of S.c. Hansen CBS 5926 on chemiluminescence activity of polymorphonuclear cells from porcine blood has been studied. Isolated cells as well as cells in whole blood showed a significant induction of chemiluminescence. In comparison to Zymosan A the yeast cells originating from the commercial formulation showed a less pronounced ability to stimulate the chemiluminescence of blood cells. However, the stimulation was significantly enhanced after inactivation by heat.

[Intrinsic oxygen affinity of hemoglobins: the hemoglobin of bisons (Bison bonasus,Bovidae)].

The hemoglobin from a European Bison (Bison bonasus) was analysed and the complete primary structures of the alpha I-, alpha II-and beta-chains have been determined. The alpha I- and alpha II-chains differ only at position alpha 19 (Asp----Gly). The beta-chains are homogeneous. The sequences are compared with the globin chains of Bison bison and bovine and the polymorphism of the alpha-chains is discussed. On the basis of the primary structure it may be concluded that the hemoglobin of Bison bonasus belongs to the group of hemoglobins with intrinsically low oxygen affinity.

[Is the alternating pulse a prognostically unfavorable sign for the postoperative course in patients with severe aortic defects?].

Pulsus alternans has been observed in patients with severe aortic valve disease and hypertensive or coronary artery disease, and has been considered a sign of severe ventricular dysfunction. Between 1974 and 1982 we observed 12 patients with severe aortic valve disease (10 aortic stenosis, 1 aortic insufficiency and 1 with a combination of both valve lesions) and pulsus alternans (group 1). Twelve patients (9 aortic stenosis, 1 aortic insufficiency and 2 with a combination of both valve lesions), but without pulsus alternans, served as controls (group 2). All 24 patients underwent surgery and were followed up for 36 months (group 1) and 50 months (group 2) respectively. One patient from group 2 died after surgery from cerebral hemorrhage. Mean age was similar in both groups (56 vs. 55 years). Preoperatively, the patients in group 1 were in a higher NYHA class (2.7 vs. 2.3; p less than 0.05) and had a higher heart rate (90 vs. 71 beats/min; p less than 0.005) than patients in group 2. M-mode echocardiography showed reduced left ventricular systolic shortening (23% vs. 35%; p less than 0.001) and increased left atrial diameter (4.6 cm vs. 3.8 cm; p less than 0.05) in group 1 compared with group 2. Systolic pressure gradient, aortic regurgitation, left ventricular end-diastolic and peak systolic pressure were, however, similar in both groups. Left ventricular angiographic ejection fraction was significantly reduced in group 1 (48% vs. 60%; p less than 0.01) compared with group 2. Postoperative follow-up was similar in both groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Alkaline phosphatase of the calf intestine hydrolyzes phospholipids.

Pure alkaline phosphatase of the calf intestine is able to hydrolyze phosphatidylinositol 4,5-diphosphate (TPI) to phosphatidylinositol and Pi and to dephosphorylate phosphatidic acid. This phosphomonoesterase activity shows a considerably high specific activity when an incubation medium at neutral pH containing 3 mM deoxycholate is used. The activity is inhibited by low concentrations of Ca2+. The enzyme has no detectable phosphodiesterase activity under the conditions tested.

A three-dimensional reconstruction of high endothelial venules in the mouse lymph node: an enzyme-histochemical study.

A three-dimensional reconstruction of high endothelial venules (HE-venules) of an entire mouse lymph node is presented. The reconstruction has been made by means of the histochemical technique for alpha-naphthylacetate esterase. The course of the HE-venules is shown in coherence with lymphocytic aggregates (follicles and unit), which were concomitantly reconstructed. Carbonic anhydrase, glutamate-, and alpha-glycerophosphate dehydrogenase were found in the high endothelia, while calcium-stimulated NA+, K+ ATP-ase and the acetylcholinesterase were localized to the endothelia and/or to the perivascular sheath of the HE-venules and submarginal capillaries.

Relationships between blood groups, isozymes and halothane reaction in pigs from a selection experiment.

German Landrace pigs (n = 1500) were halothane-tested and blood samples were taken for the determination of A-O and H blood types as well as for the determination of PHI and 6-PGD isozymes. The pigs originated from two generations (7th and 8th) of a selection experiment 'selection for activity of NADPH-generating enzymes in backfat of pigs'. The selection lines are E-, E+ (selection for low and high enzyme activity), U- (selection for low ultrasonic backfat thickness) and K (control). Preliminary results show an average proportion of halothane-susceptible animals of 49%. The frequencies of halothane-positive pigs amount to 60%, 46%, 70% and 30% in lines E-, E+, U- and K, respectively. The investigation shows a non-random combination of the marker genes caused by linkage disequilibrium, especially in line E-. Recombination frequencies between the loci vary from 0% to 18%.

Dermatoglyphic findings in patients with fragile X-chromosome.

Finger- and palmar prints of hemi- and heterozygote fragile X-patients with mental retardation (10 males and 5 females) were compared to dermatoglyphic findings in 20 mentally retarded patients (10 males and 10 females) without fragile X and to 200 healthy unrelated persons (100 males and 100 females). Characteristic whorls and double-loops with high ridge-counts on finger-tips and a pronounced transversal course of palmar ridges were restricted to males with fragile X. Female carriers of fragile X showed, corresponding to male patients, some abnormalities of the digital- and palmar ridge-pattern. Contrary to males, in carriers as well as in mentally retarded females without fragile X, fingerprints with low ridge-counts were found. Common to all mentally retarded patients, but more pronounced in males with fragile X, abnormal palmar creases and hand-measurements were observed. These findings probably are related to prenatal retarded growth of the length of the palma and of the middle-finger.

[Can oscillatory estimation of airway resistance in chronic obstructive patients be improved by the use of double reference resistance?].

Tests were carried out on the usefulness of doubling the reference resistor during airways resistance measurement by forced oscillation method (set frequency technique). Measurements on 110 random subjects using the standard reference tube (Zo = 10 hPa/1/s) for comparison showed almost identical Ros values using the experimental tube, while the latter gave higher Ros values as obstruction increased. Comparisons with the bodyplethysmographic Rt on 65 patients with a chronic obstructive airways disease yielded a closer approximation to the identity curve from 7.0 hPa/1/s using this modified technique. The practical conclusion is to recommend the use of the double reference resistor for the basic clinical diagnosis of obstructive airways disease, both in quantifying advanced grade obstructions and in assessing the damaged respiratory mechanism in chronically ill patients with pronounced ventilatory resistance.
