The effect of thymic hormones (TFX) on lymphocyte subpopulations and proliferation after maximal physical effort.

The studies were performed on healthy well-trained cyclists. Maximal physical exercise was performed on a Monark bicycle ergometer according to individual schemes. Heart rate amounting to about 200 bts/min and oxygen consumption stabilization were considered as criteria for maximal physical exercise. In this study we have investigated the effect of short-term stimulation of conditioned sportsmen with thymic hormones and evaluated T cell subsets, DR antigen and transferrin receptor expression as well as mitogen-induced proliferation of lymphocytes before and after maximal physical effort. The results suggest that intensive physical exercise may be responsible for transient decrease of CD4/CD8 ratio and mitogen responsiveness, and increase of mononuclear cells number bearing HLA DR+ and CD71 antigens. These changes were modified by the treatment with thymic hormones.

T lymphocyte subsets and NK cell cytotoxicity in chronic hemodialysis patients. The effect of recombinant human erythropoietin (rHu-EPO) treatment.

We investigated subpopulations of T lymphocytes, NK cell number and cytotoxic activity in 14 chronic uremic patients on regular hemodialysis treatment. We observed a significantly decreased absolute lymphocyte number and percentage of CD3 cells. Relative numbers of CD16 cells were significantly elevated, but NK cell cytotoxic activity was within a normal range. Nine patients with chronic renal anemia on maintenance hemodialysis were enrolled in rHu-EPO treatment trial. The treatment was continued till the hematocrit level reached 30%. Each of the patients had corrected anemia and well-being. After 12 weeks of the treatment we observed in these patients decreases in CD3, CD4, CD8 and CD16 cell numbers and elevation of CD4/CD8 ratio. Cytotoxic activity of NK cells did not change significantly. Presented results indicate that chronic hemodialysis patients have significantly diminished lymphocyte number. rHu EPO treatment affects the T lymphocyte subsets inducing a deep decrease of CD8 and CD16 cell percentage leading to normalisation of the CD4/CD8 ratio.

Effect of thymic extract on allogeneic MLR and mitogen-induced responses in patients with chronic active hepatitis B.

Cell-mediated immune reactions are involved in the development of chronic active hepatitis (CAH-B). The present studies confirm that lymphocyte from patients with CAH-B have a decreased allogeneic mixed lymphocyte reaction (allo-MLR) and mitogen-induced responses. These abnormalities probably play a role in the immunological dysfunctions underlying chronic liver diseases. Twenty one patients with biopsy-proven CAH-B and a significantly lowered CD4:CD8 cell number ratio were treated with thymic extract (Thymomodulin-TFX) for 6 months. The results of immunological, serological and biochemical findings indicate that this preparation has an immunoregulatory action and produces beneficial clinical effect in patients with CAH-B.

[Comprehensive assessment of cell mediated immunity in petrochemical industry workers].

In the group of 29 workers of Acetobenzene Division of the petrochemical plant in Ptock with occupational exposure to toluene, acetone, and ammonia a set of tests were done evaluating the state of cell-mediated immunity which made possible finding out persons showing abnormalities in the studied parameters. It is suggested to follow-up these person continuously with control immunological testing once a year.

The effects of occupational exposure to hydrocarbons on some immune parameters of workers of the phenol division of a petrochemical plant.

The subjects of investigation included 34 workers at the Phenol Division of the Mazovian Refining and Petrochemical Plants and 18 persons working in administrative positions; the purpose was to elucidate some functions of the immune system cells. The lymphocyte subpopulations (CD3, CD4, CD8) and NK cells were evaluated using monoclonal antibodies and the immunofluorescence method. To search for the functions of lymphocytes and monocytes such as the in vitro production of interleukin-1 (Il-1) and interleukin-2 (Il-2), macrophage inhibitory factor generation (MIF), a cytotoxicity test and T cell proliferation in autologous mixed lymphocyte reaction (AMLR) were also examined. We have shown that workers from the Phenol Division exhibited pronounced deviations in the tested parameters as compared to the administration workers. We found a decrease in CD3 lymphocytes and in Il-1 production. After analysis of the individual results we selected three persons from the Phenol Division who showed abnormal values in four or more parameters (decreased CD4:CD8 ratio, abnormal values of lymphocyte subpopulations, impaired lymphocyte functions in functional tests). These cellular-immunity control tests are promising methods for studies of the biological effects of environmental and/or occupational pollution to toxic derivates of petroleum.

Effect of hemodialysis on T lymphocyte subsets, con-A-activated suppressor cell activity, and interleukin-2 receptor expression on lymphocytes in chronic uremic patients.

Effect of hemodialysis (HD) on some indices of immune response was studied in nine chronic uremics. Total lymphocyte, OKT4+, and OKT8+ cell numbers significantly decreased during the first 20 min of HD, and they were decreased till the third hour of the procedure, whereas the OKT4+/OKT8+ cell number ratio did not change significantly. Before HD, Con-A--activated suppressor cells exerted a stimulatory action on autologous responder cells measured in two-step culture. During HD, Con-A-activated suppressor cell activity transiently appeared, with its peak at 60 min after the start of HD. It was accompanied by a transient rise in lymphocyte count with spontaneous interleukin-2 (IL-2) receptor expression, whereas the number of cells expressing IL-2 receptor following phytohemaglutinin (PHA) stimulation was progressively decreased during HD. A significant correlation was found between the increment of Con-A-activated suppressor cell activity and the increment of spontaneous IL-2 receptor expression on lymphocytes during one single blood flow through the dialyzer. The results supply further evidence that HD may impose additional disturbances on immune regulation in chronic uremics.

T lymphocyte subsets, natural killer cell cytotoxicity and autologous mixed lymphocyte response in children after splenectomy because of hereditary spherocytosis.

Lymphocyte subpopulations and autologous mixed lymphocyte reaction (AMLR) studied in 13 hereditary sphereocytosis and 21 postsplenectomy patients; the operation was performed because of hereditary spherocytosis. Significant increases of Th cells and the Th/Ts ratio have been noted in children with hereditary spherocytosis. Splenectomy because of spherocytosis has resulted in an increase in Ts cell number and diminution of Th/Ts ratio. Hereditary spherocytosis patients showed both diminished natural killer cell activity and AMLR proliferation. Splenectomy normalized the above parameters. The presented data demonstrated disturbances in T lymphocyte number and functions both before and after splenectomy in children with hereditary spherocytosis.

Effect of maximal physical exercise on T-lymphocyte subpopulations and on interleukin 1 (IL 1) and interleukin 2 (IL 2) production in vitro.

Maximal physical exercise was performed on a Monark bicycle ergometer according to individual schemes. The investigations were carried out on 11 highly trained cyclists, aged 20 +/- 1 years. Heart rate (HR) amounting to about 200 bts/min and oxygen consumption stabilization were considered as criteria for maximal physical exercise. Lymphocyte phenotypes were determined using monoclonal antibodies. A significant increase in Ts (suppressor, cytotoxic) and a moderate increase in Th (helper, inducer) and NK (natural killer) cell numbers were noted 3 min after maximal physical exercise. At the same time, a significant diminution of the Th/Ts ratio was observed. A significant increase of interleukin 1 production and a diminished interleukin 2 production as well as spontaneous interleukin 2 receptor expression (Tac antigen) were observed at the same time. After a 2-h recovery, there was a normalization of most of the parameters investigated. The results suggest that maximal physical exercise in highly trained bicycle racers generates transient changes in immune cell function.

The immunoregulatory granulocyte factor (GF). I. Some antigenic and structure features.

Adherent PMNLs secrete protein factor termed granulocyte factor (GF). The antigenic and some structural features of GF were examined in the present paper. Molecular weight of GF was established on Sephadex G-75 column and is amounted to 40 kD. GF exhibited two isoelectric points in pH ranged from 4.7 to 4.9 and from 5.7 to 6.4. SDS polyacrylamide gel electrophoresis has displayed two bands, 20 and 68 kD, respectively. Initial analysis of amino acid composition of purified GF has shown 15 amino acids, but without aromatic and thioamino acids. Antigen-dependent species specificity was confirmed using rabbit anti-human, -murine, -guinea pig or-rat - GF IgG applying conventional precipitation methods in agar gel no crossreaction was noted. Using precipitation analyses GF was localized in the specific granules of PMNLs.

The granulocyte factor abrogates the alloantigen-induced delayed type hypersensitivity suppression in mice.

Adherent granulocytes secrete granulocyte factor (GF) in the course of the first 60 min of adherence. GF abolishes suppressor cell generation in mice induced by intravenous injection of irradiated allogeneic lymphoid cells. The effect of such induced suppressors was measured in delayed type hypersensitivity reaction induced by subcutaneously injected irradiated allogeneic spleen cells and evaluated in the foodpad test. Suppressor cell induction or GF treatment had no effect on antigen- or phytohaemagglutinin-induced production of migration inhibitory factor.

Enhancement of the autologous mixed lymphocyte reaction (AMLR) in children splenectomized after trauma.

Splenectomized individuals have been reported to have many immunological abnormalities. We have studied the proliferative response of T cells in AMLR of nine children who had undergone splenectomy after trauma. T cells from splenectomized patients showed increased proliferative response in AMLR as compared with that observed in parallelly studied controls (38.480 +/- 19.700 vs 11.160 +/- 3.760 dpm). The percentage of CD4 cells was diminished in the group of splenectomized patients. We found strong correlation between AMLR and time after splenectomy (r = -0.81, p less than 0.01). Possible mechanism of elevated AMLR following splenectomy and hypotheses on generation of AMLR are discussed.

Immunoregulatory granulocyte factor (GF). II. In vitro features of antibodies against GF+.

Activated polymorphonuclear leucocytes (PMNL) secrete a specific factor possessing numerous immunoregulatory properties. The rabbit IgG antibodies against this human granulocyte factor GF (anti-GF-IgG) have significantly changed the regulatory features of GF and the same antibodies alone have also exerted immunoregulatory potency when tested in vitro. Inhibition of mixed lymphocyte reaction (MLR) proliferation after GF treatment was abolished in the presence of anti-GF-IgG. GF displays a potentiating effect on interleukin-1 (IL-1) production by human monocytes in vitro. Anti-GF-IgG did not change the potentiating capacity of GF and these specific antibodies alone also enhanced IL-1 production to the same extent as GF. In peripheral blood mononuclear cell (PBMC) cytotoxicity test, the cytotoxic activity of effector cells was markedly reduced in the presence of GF. Anti-GF-IgG partially abrogated the observed suppression, although their own features indicate a strong inhibitory effect on PBMC cytotoxicity. These contradictory results are difficult to explain but they may enable us to understand the pleiotropic immunoregulatory mechanism of both GF and anti-GF-IgG. The results of the present and recently published studies support the idea that both GF and anti-GF-IgG modify T lymphocyte function at the same receptor level.

[Evaluation of various indicators of cellular immunity in persons exposed to work environment in the Masovian Refinery and Petrochemical Plants in Plock].

In 137 workers of the Mazovian Refining Petrochemical Plant cellular immunity parameters were examined. The following tests were performed: number of E rosette forming cells (T lymphocytes), TG and TM lymphocytes, EAC rosette forming cells (B lymphocytes), MIF generation and suppressor and cytotoxic activity of T lymphocytes. Workers of Asphalt Oxidation Division exhibited most pronounced deviations of the test parameters, as compared to controls.

Relative and absolute numbers of T lymphocyte subpopulations and NK cells in male population in relation to age.

Using BMA monoclonal antibodies and fluorescent microscope, percentages and absolute numbers of lymphocytes, T cell subsets and NK cells were enumerated in peripheral blood from 126 healthy men. Although absolute numbers of total lymphocytes did not differ according to age, the numbers and percentage of natural killer (NK) cells showed positive interrelationship with age. The percentage but not absolute numbers of cells reacting with BMA 030 (CD3) and BMA 040 (CD4) antibodies were significantly increased only in groups aged of 20-29 yrs and 30-39 yrs. The percentage and number helper/inducer T cells (CD4) were comparable in the four groups of subjects. These results indicate that peripheral lymphocyte populations and T cell subsets and NK cells remarkably vary in healthy men over a wide range of ages.
