Splenic ultrastructural features in idiopathic thrombocytopenic purpura.

Spleen samples from idiopathic thrombocytopenic purpura (ITP) patients were investigated for ultrastructure and peroxidase activity. The white pulp was characterized by the presence of many large secondary lymphatic follicles and periarteriolar sheaths formed from lymphocytes and plasma cells. Their ultrastructural details substantiate the process of lymphocyte differentiation and the immunopathologic mechanism of the disease. The vascular lesions proved to be subendothelial hyaline deposits. The platelet destruction process and phagocytosis resulted in the occurrence of fibrin clusters. Macrophages were also described as stainable body macrophages, foamy macrophages and a third type dealing with fibrin phagocytosis.

Ultrastructural peroxidase pattern indicating cellular maturity in myelomonocytic leukaemia.

The ultrastructural pattern of endogenous peroxidase of monocytes has been investigated in bone marrow samples as well as in the blood of patients labelled as myelomonocytic leukaemia (MML) after clinical and laboratory investigation. The predominance or the exclusive presence of a granular pattern in the bone marrow proved to be consistent with chronic MML cases. On the other hand, the presence of the peroxidase in RER, nuclear envelope, Golgi complex and granules in the bone marrow and even in the peripheral blood occurred in acute MML patients.

Electron microscopic investigation of cell surface antigens using a multivalent hybrid antibody with double specificity.

Multivalent hybrid antibody (MHA) complexes with double specificity were prepared by combining two different antibodies, one specific against ferritin (Fer), the other against horseradish peroxidase using protein A from Staphylococcus aureus (SpA). Electron microscopy of mouse spleen lymphocytes and thymocytes (the latter coated with mouse anti-Thy-1 antibody) reacted with anti-HRP/SpA/anti-mIg complex and HRP showed the pattern of surface Ig receptors visualized by the specific peroxidase labelling. When IgG anti-Fer/SpA/IgG-anti-mIg complex was applied to the same cellular stuff, better resolution and higher accuracy were obtained although the distribution was similar. Surface Thy-1 alloantigen and Fe receptors (charged with hIgG) were simultaneously detected on the thymocyte surface by using in the same way a mixture of anti-Fer/SpA/anti-Thy-1 and anti-HRP/SpA/anti-Fab. The concomitant ultrastructural visualization of Thy-1 (with Fer) and Fc (with HRP) on mouse thymocytes clearly showed that their distribution was mainly independent and that the amount of Thy-1 antigen prevailed. These data show that electron microscopy with a mixture of MHA may be a useful technique for the concomitant location of two antigenic determinants on the cell surface.

Chemical sclerosis of the adrenal medulla with sodium salyclate (2.5 M) - histopathologic, histoenzymatic, electron microscopic and biochemical studies.

The selective inhibitory effect of sodium salicylate (2.5 M) on the adrenal medulla was investigated histopathologically, histoenzymatically, electron microscopically and biochemically. It was proved that the different behaviour of nicotinamide-adenine dinucleotide (NAD) and nicotinamide-adenine dinucleotide phosphate (NADP) linked dehydrogenases under the action of sodium salicylate can account for the survival of cortical cells, while the medullar ones are necrosed.

Ultrastructural correlation between follicular dendritic cells and the frequency of virus-like particles during the lymph nodes reaction of SIV/HIV 2 infected cynomolgus monkeys.

Lymph nodes from 18 SIV/HIV2 infected monkeys developing a severe AIDS-like disease were studied by electron microscopy. The lesions were correlated with the histopathologic staining and the anti-SIVp28 immunostaining. The investigation of follicular dendritic cells (FDC) and associated retroviral particles was focused on the light zones of germinal centres during the follicular hyperplasia, fragmentation atrophy and depletion. FDCs were mostly developed in the first two stages, while the highest amount of retroviruses was found in the two middle stages. During the last stage of follicular depletion FDCs were still present but deprived of dendrites and no viral particles were seen around. Associated lesions were small haemorrhages, binucleated FDCs, viral phagocytosis by macrophages and reticular paracrystalline inclusions.

Microscopic and ultrastructural aspects in retinoblastoma.

A number of 12 retinoblastoma cases were studied. We had in view a number of histopathological aspects (tumor type, neighbouring structures invasion) which are considered to be decisive in appreciating the vital prognosis. We mention that undifferential retinoblastoma of histopathological type was met in 8 cases and the neighbouring structures invasion was present in 7 of them. We also studied the ultrastructura of the tumor cells belonging to the malignant tumor.
