Cerebellar control of eye movements studied with injection of harmaline in the trained baboon.

1. The alteration of the cortical cerebellar mechanisms resulting from the activation of the olivo-cerebellar pathway by Harmaline (H) administered at doses which were subthreshold for skeletomotor tremor (5 mg/kg, IM) yields, in the trained Baboon (Papio papio), a marked decrease of oculomotor system performance. 2. The mean angular excursion of the spontaneous eye movements produced in the dark over a given period of time is increased by 50%. The increase is basically due to an increase of the number of saccades in the 0-20 degrees amplitude range. 3. The eye movements in response to a slow target displacement are altered as evidenced by a decrease of the smooth pursuit gain, a 60% increase in latency, and by intrusive saccades. The effects are dose dependent so that around 8 mg/kg of H, smooth pursuit is totally suppressed. 4. The saccadic system is also altered, Stepping target tracking shows dysmetria and increase of the amplitude of the fixation eye movements. The velocity-amplitude relationship of the saccades is not modified but the saccadic reaction time is increased by 50%. The observed alterations are in no way similar to the tremor commonly induced by the drug, at skeletal level. In fact, no ocular tremor at 8 to 10 Hz was recorded. 5. In conclusion, administration of H alters considerably the control and the stability of the oculomotor system in a way similar to that resulting from partial or total cerebellectomy. It may thus be possible to use the drug to simulate, in a reversible manner, cerebellar dysfunctions and study the involvement of the cerebellum in oculomotor control.

Intracellular flow of particulate steroid-receptor complexes in steroid target cells.

The structural expression of nuclear transportation of receptor-bound oestradiol was investigated by immunoelectron microscopy on human breast cancer cells. In oestradiol-treated cells during transportation, oestradiol antibodies attached to cytoplasmic oestradiol-bearing particles which seemed to interact with the nuclear membrane. These particles, subserving the intracellular flow of steroid-receptor complexes, could constitute a special cellular system of macromolecular transport, communication and compartmentalisation.

Estrogen receptor cytochemistry by fluorescent estrogen.

A recently synthesized fluorescein-labeled estrogen (17FE, 1-(N)-fluoresceinyl-estrone-thiosemicarbazone) interacts with estrogen-target cells like the native hormone and visualizes the uptake, transport, and distribution of estrogen in intact target cells. Moreover, estrogen binding sites are traced by 17FE in cryostat sections of estrogen target tissues as well. Cell and tissue 17FE binding sites fulfill the accepted criteria for specific estrogen receptors (finite binding capacity, high affinity, steroid and tissue specificity). This fluorescent probe allows estrogen receptors to be studied in a wide variety of cell and tissue preparations under varying conditions of physiologic and pathophysiologic interest.

[Vulvovaginal candidiasis in pregnancy and its treatment with clotrimazole].

The Writers have carried out a study on 624 pregnant women pointing out that the incidence of vulvovaginal candidiasis is notably increased (35,5%) during pregnancy with a slight difference in the three three-monthly periods of pregnancy. Controls on 195 women after treatment with clotrimazole showed an average percentage of recovery of 83% with a maximum of 87% in the second three-monthly period.

[Evaluation of the degree of maturation of the cervico-vaginal epithelium in normal, inflammatory and dysendocrine states].

The AA. have determined the maximum percentage of the para-basal cells compatible with a phlogistic state resulting from the exam of 2400 cervical and vaginal smears from women of various ages that were not following therapies able to falsify the evaluations of the endocrine situations. At the objective exam the patients had no sure signs of erosions, thyroid illnesses, vitamin deficiency or cervical metaplasia. The aim of the research was to find a criteria of evaluation of the endocrine situations of the patients, that were not easily influenced by the variations of the karyopyknotic index and acidophilix that are met in the course of numerous illnesses.

[Statistical observations on the incidence of Trichomonas vaginalis in the female population of Terni from 1974 to 1978].

The casuistics refers to 1646 cases equal to 20,5% of vaginal trichomoniasis in a total of 7996 pap tests examined in the service of Anatomy and Histology Pathology in the hospital of Terni. The exams of the incidences of trichomoniasis in the various age classes, reveals that the main frequency is found in the 2nd., 3rd., 4th decades of life, that is in the period of maximum sexual attivity. Vaginal Trichomonas is rarer before puberty and after the menopause.

[Association between Trichomonas vaginalis and some types of lesions of the uterine neck based on data collected in the quinquennium of 1974--1978].

In our statistic the number of cases of vaginal trichomoniasi in which discariosi, displasie, metaplasie are found are represented in about 40%. While 37,5% of women affected with neoplastic lesions had Trichomonas vaginalis. It is certain that in the presence of flogosi from Trichomonas vaginalis we meet more frequently than normal, atipie cellulars and that often an exact valutation of this lesion is put back to an exam after the recovery of the Trichomonas vaginalis, because fortunately, more than often these disturbances disappear.

Concerning the localization of steroids in centrioles and basal bodies by immunofluorescence.

Specific steroid antibodies, by the immunofluorescence technique, regularly reveal fluorescent centrioles and cilia-bearing basal bodies in target and nontarget cells. Although the precise identity of the immunoreactive steroid substance has not yet been established, it seems noteworthy that exogenous steroids can be vitally concentrated by centrioles, perhaps by exchange with steroids already present at this level. This unexpected localization suggests that steroids may affect cell growth and differentiation in some way different from the two-step receptor mechanism.

N-banding and nucleolus organisers in Asellus aquaticus (Crust. Isop.).

The N-banding technique was used to stain the nucleolus organiser of the karyotype of Asellus aquaticus (crust. Isop.). Observations were made on the morphological expression of nucleolus organisers as secondary constrictions and the presence of nucleoli in mitotic prophase. An attempt was made to correlate the various results and it seems likely that N-banding is not a reflection of NO activity.
