Physicochemical, nutritional, and microstructural characteristics of chickpeas (Cicer arietinum L.) and common beans (Phaseolus vulgaris L.) following microwave cooking.

Microwave cooking of legumes such as chickpeas and common beans was evaluated by assessing the cooking quality (cooking time, firmness, cooking losses, and water uptake) and the physicochemical, nutritional, and microstructural modifications in starch and nonstarch polysaccharides. Compared to conventional cooking, microwave cooking with sealed vessels enabled a drastic reduction in cooking time, from 110 to 11 min for chickpeas and from 55 to 9 min for common beans. The solid losses, released in the cooking water, were significantly less after microwave cooking than after conventional cooking (6.5 vs 10.6 g/100 g of dry seed in chickpeas and 4.5 vs 7.5 g/100 g of dry seed in common beans). Both cooking procedures produced a redistribution of the insoluble nonstarch polysaccharides to soluble fraction, although the total nonstarch polysaccharides were not affected. Increases in in vitro starch digestibility were similar after both cooking processes, since the level of resistant starch decreased from 27.2 and 32.5% of total starch in raw chickpeas and beans, respectively, to about 10% in cooked samples and the level of rapidly digestible starch increased from 35.6 and 27.5% to about 80%. SEM studies showed that the cotyledons maintained a regular structure although most of the cell wall was broken down and shattered by both cooking procedures. In addition, the ultrastructural modifications in the cotyledon's parenchima and cells are consistent with the chemical modifications in NSP and the increase in starch digestibility after cooking.

Rapid detection of meso-diaminopimelic acid in lactic acid bacteria by microwave cell wall hydrolysis.

Two innovative microwave hydrolysis procedures for rapidly detecting meso-diaminopimelic acid in lactic acid bacteria have been developed. Whole-cell hydrolysis was performed both in liquid phase and vapor phase with 6 N HCl in sealed vessels using a microwave oven equipped with pressure and temperature probes. The presence or absence of meso-diaminopimelic acid determined by TLC, after the application of liquid- and vapor-phase microwave hydrolysis procedures, gave the same qualitative results as those obtained by traditional hydrolysis. These standardized microwave hydrolysis procedures permit a drastic reduction in hydrolysis time, from 16-20 h to less than 10 min and, consequently, in the total time of meso-diaminopimelic acid analysis (less than 90 min). Microwave hydrolysis in vapor phase is particularly convenient because, besides reducing hydrolysis time, it also eliminates the successive troublesome step of HCl removal.

Rapid determination of lactulose in milk by microdialysis and biosensors.

A simple and rapid flow system for the determination of lactulose in milk samples was developed. It is based on the hydrolysis of lactulose to galactose and fructose by the enzyme beta-galactosidase immobilised in a reactor. The amount of fructose produced was measured with an electrochemical biosensor based on the fructose dehydrogenase enzyme, K3[Fe(CN)6] as mediator and a platinum based electrochemical transducer. Parameters such as the enzyme immobilisation in the reactor and under the electrode surface, the lifetime of the beta-galactosidase reactor and of the dehydrogenase biosensor and the flow parameters were studied and optimised. Fructose was determined in the range 1 x 10(-6)-5 x 10(-3) mol l-1 with an RSD of about 2% and a detection limit of 5 x 10(-7) mol l-1. The use of a microdialysis probe as the sampling system permitted the direct measurement of lactulose in milk samples without pre-treatment in the range 1 x 10(-5)-5 x 10(-3) mol l-1. The sensitivity of the procedure allowed pasteurised, UHT and in-container sterilised milk to be distinguished.

Color Doppler sonography.

The authors report the results obtained with color Doppler sonography in the study of breast conditions. Color Doppler allows to detect the following main features in breast conditions: the presence of blood flow, vessel arrangement, vascularization extent, the number of vascular poles. To investigate slow flows, it is better to use low PRF values (not above 1 KHz) and low filters, while amplification should be set immediately above the system's noise threshold; the size of color Doppler box should be adjusted as small as possible to maximize sensitivity and minimize flash artifacts. In May 1992 to September 1997, 252 patients with solid breast masses were examined with mammography, B-mode, color Doppler and power Doppler sonography (only 57 cases). We identified histologically (176 cases) or cytologically (77 cases) 141 carcinomas and 112 benign solid lesions. The diameter of the 141 carcinomas ranged 0.4-4 cm (mean 1.7), while the diameter of benign lesions ranged 0.7-3 cm (mean 1.5). The malignancy pattern was characterized by hypervascularity (92.9%), irregular and abundant (54.2%) vascularization and more than one vascular pole. Benign lesions were avascular (43.4%) with poor and peripheral vascularity (90%) and mostly showed only one vascular pole. The avascular cancers (10 cases) were three mucoid, five in situ and two small (0.7 and 0.9 cm) invasive ductal carcinomas. The six benign lesions with irregular and abundant vascularization and more than one vascular pole were proved to be two proliferating and three juvenile fibroadenomas and one phylloid tumor. These results are encouraging and suggest that this technique can be a useful adjunct to mammography and sonography in the differential, diagnosis of breast nodules.

Epidemic of cholera among the aborigines of northern Argentina.

In February 1992, an epidemic of cholera began in Argentina. The first known case appeared in an aborigine population living by the Pilcomayo river on the Bolivian border. By the end of that year, 551 cases and 15 deaths were reported to the health authorities. Epidemiological analysis helped identify the pattern of transmission of the disease in this region of South America. Polluted watercourses and little pools, formed after the rainy season, were identified as the single source of infection for subsequent cases. Then, the epidemic adopted a person-to-person pattern of transmission and was propagated over a long course with weekly occurrence. Anthropological, cultural, and other factors that contributed to the origin and spread of cholera in northern Argentina are described in this study.

[Use of vital statistics in maternal mortality prevention programs].

"In the first part of the article the vital statistics system of the Republic of Argentina is presented, highlighting its characteristics as a data source, the integrity of its death records, the quality of the medical certification as to the cause of death by means of indirect indicators and the under registration of maternal causes. In the second part, the results of a research on maternal mortality carried out in the Federal Capital from 1987 to 1989, which was later extended to other jurisdictions of the country (Cordoba, Tucuman and Corrientes), is synthesized.... The principal objectives were...to know the real structure of maternal mortality in the Federal Capital and to identify the factors associated with it, in order to orient the activity of the health services and reduce these deaths." [Neonatal hemoperitoneum: role of ultrasonography. Experience with 4 cases].

This study was aimed at investigating the role of US in the newborn with a clinical suspicion of hemoperitoneum. Hemoperitoneum is a dramatic clinical event which may be caused by some obstetrical and fetal factors--breech or complicated delivery, hepatomegaly, macrosomy, anoxic liver congestion being the most frequent causes. Direct or indirect trauma of an abdominal organ during delivery represents the major pathogenetic factor. Clinical symptoms are correlated with the appearance of shock whose intensity is proportional to trauma severity: symptoms may appear even 48 hours after delivery. The authors report their experience with 4 consecutive cases of neonatal hemoperitoneum which underwent US of the abdomen 24 to 72 hours after birth. In the first 3 newborns, US was performed on the basis of a clinical picture of bleeding shock, while in the extant newborn US was performed after a sudden increase in blood transaminase level associated with an obstetrical paralysis of the left arm. The US examinations were performed using a 7.5-MHz sectorial probe unit (Siemens). In all patients, US demonstrated the presence of a liquid effusion in the abdomen which could be referred to hemoperitoneum. In two patients, it was associated with a traumatic focal alteration of the liver parenchyma, in the third with a diffuse subcapsular liver hematoma and, finally in the fourth patient with a disconnected splenic vascular pedicle which was at surgery demonstrated. In conclusion, US is indicated as the most reliable and valuable diagnostic technique in the study of neonatal hemoperitoneum. The authors believe abdominal US to be essential in all the neonatal cases where hemoperitoneum is suspected and in selected cases presenting specific risk factors, to allow a rapid diagnosis and better treatment planning.

Protein quality and antinutritional factors in wild and cultivated species of Vigna spp.

The nutritional quality of wild and cultivated species of Vigna has been evaluated. Wild species showed significantly higher protein content (P less than 0.01), antitryptic activity (P less than 0.005) and tannin content (P less than 0.025) and significantly lower protein digestibility (P less than 0.005). No significant differences were found with regard to protein quality, measured as chemical score corrected by the digestibility; sulphur amino acids turned out to be limiting in both groups. In addition no correlations were found between sulphur amino acids and protein levels, or between sulphur amino acid and trypsin inhibitor levels. Selected lines showed an improvement from the nutritional point of view linked to lower antitryptic factors and tannin content, which may give rise to a decreased resistance to pests. For all the properties considered, whether physical or biochemical, wild samples presented wider variations and the types considered most primitive were those that differed most from the cultivated ones, whose properties were altogether more consistent. In vitro protein digestibility turned out to be a useful parameter in evaluating wild and improved samples.

NMR study of seed hydration: the role of the seed anatomical structures in water uptake of soaked cowpeas.

A NMR method based on the analysis of the transverse magnetization decay curve of water protons, was employed to study the hydration process of commercial cowpeas. In order to investigate the role of the different anatomical parts of the seeds, hilum and micropyle, or alternatively seed coat, were inhibited by a water resistant epoxy resin. The kinetic constant for the hydration of the different sets of beans was measured.
