The role of cisplatin in chemotherapy of advanced breast cancer.

Cisplatin containing regimens as first-line, second-line or as a third-line chemotherapy were administered in 26 and 36 patients, respectively. The overall response rate in patients on first-line chemotherapy was 53.9%, in patients on second or third-line chemotherapy 30.6%. The differences both in overall and disease-free survival between patients on first-line and on second/third-line chemotherapy were statistically significant in favor of women treated with first-line chemotherapy (p = 0.05). Hematologic and nonhematologic toxicities were mild to moderate and were more pronounced in patients on second and third-line chemotherapy. The overall response rate, DFS and OS were significantly better and longer in the group of patients treated with "bolus" CDDP in comparison to the group of patients treated with CVI CDDP. Our results confirm the activity of cisplatin-containing regimens (mainly CAP schedules) in patients with advanced breast cancer not only as a first-line therapy but also in heavily pretreated patients by chemotherapy and/or radiation therapy and endocrine manipulation.

[The induction of latent liver damage by cyclophosphamide].

The effect of the cyclophosphamide on the changes of the mitotic index (MI), metaphase-prophase ratio (M/P) and chromosomal aberrations in postmetaphase in regenerating rat liver was studied. Cyclophosphamide was injected in the single doses of 100 and 200 mg/kg i.p. 2 hours before partial hepatectomy (PHE). The results have shown that cyclophosphamide caused latent liver damage which was manifested after PHE in reduced MI and increased frequency of chromosomal aberrations. The extent of the changes induced by cyclophosphamide corresponds to that due to the effect of 3 or 6 Gy of X-rays, respectively.

Morphology of mononuclear cells in the peripheral blood of rats after single irradiation.

The morphology of nucleated cells in the peripheral blood of rats after single dose irradiation with a dose of 5.5 Gy was followed during 28 days after irradiation. During profound agranulocytopenia and granulocytopenia the number of lymphocyte-like mononuclear cells was increased from the days 7-10 after irradiation and the number of monocyte-like mononuclear cells increased from day 14. The cell population discussed in the paper differed markedly from typical lymphocytes and monocytes in particular cytomorphologic parameters.

[The effect of chronic gamma irradiation in an exponentially decreasing dose rate on the nucleic acid content in the hematopoietic organs and blood of rats].

The effect of continuous gamma irradiation at exponentially decreased dose rates (from 562 mGy/h to 13 mGy/h with a total cumulative dose of 14.355 Gy delivered over a period of 10 days) on the nucleic acid content of rat hemopoietic tissues and blood was followed up. The radiation model used simulated a decrease in the radioactivity of a fission mixture in the contaminated environment resulting from a nuclear device accident. We have found that the dynamics of the changes seems to be similar to that observed after acute exposure, and the hemopoiesis recovery starts just at the time of irradiation. In evaluating the damage and recovery extent after accidental irradiation, we consider it expedient to complement the biological dosimetry with the indices studied work including the determination of DNA and RNA concentrations in blood of irradiated human beings.
