Microactuators based on conducting polymers.

In this contribution we report the use of polyaniline and polypyrrole for miniaturized actuators fabricated by microstructural and electrochemical technologies. The potential necessary to drive the actuator is typically less than 1 V, i.e. 2-3 orders of magnitude lower than that necessary for the widespread piezoelectric actuator devices. This low voltage is imperative for future application of actuators of micrometer dimensions. The volume variation of polymers substantially exceeds that of piezoelectric materials. Different contributions to the actuator characteristics are discussed and evaluated semi-quantitatively.

Transmission line probe based on a bow-tie antenna.

A high transmission probe for scanning near-field optical microscopy is discussed that is based on a bow-tie antenna. The proposed design of the transmission line probe relies on batch-fabricated hollow pyramidal silicon dioxide tips that are partly coated with aluminium to accomplish the tapered dipole antenna. Theoretical calculations of the field distribution were performed to investigate its optical properties. Results were compared with those of conventional aperture tips based on the same silicon dioxide tip configuration, and revealed unique properties with respect to the transmission efficiency.

Determination of preferential binding sites for anti-dsRNA antibodies on double-stranded RNA by scanning force microscopy.

The monoclonal anti-dsRNA antibody J2 binds double-stranded RNAs (dsRNA) in an apparently sequence-nonspecific way. The mAb only recognizes antigens with double-stranded regions of at least 40 bp and its affinity to poly(A) poly(U) and to dsRNAs with mixed base pair composition is about tenfold higher than to poly(I) poly(C). Because no specific binding site could be determined, the number, the exact dimensions, and other distinct features of the binding sites on a given antigen are difficult to evaluate by biochemical methods. We therefore employed scanning force microscopy (SFM) as a method to analyze antibody-dsRNA interaction and protein-RNA binding in general. Several in vitro-synthesized dsRNA substrates, generated from the Dictyostelium PSV-A gene, were used. In addition to the expected sequence-nonspecific binding, imaging of the complexes indicated preferential binding of antibodies to the ends of dsRNA molecules as well as to certain internal sites. Analysis of 2,000 bound antibodies suggested that the consensus sequence of a preferential internal binding site is A2N9A3N9A2, thus presenting A residues on one face of the helix. The site was verified by site-directed mutagenesis, which abolished preferential binding to this region. The data demonstrate that SFM can be efficiently used to identify and characterize binding sites for proteins with no or incomplete sequence specificity. This is especially the case for many proteins involved in RNA metabolism.

Modified fabrication process for aperture probe cantilevers.

We report the development of cantilever- and fibre-based probes for scanning near-field optical microscopy. Both probe concepts rely on the integration of a microfabricated aperture tip with reproducible optical and mechanical properties. Numerical calculations were carried out using a finite integration code to investigate the polarization-sensitive transmission behaviour of aperture tips. In order to establish technological guidelines for the optimization of the properties of the optical tip the distinct influence of the tip geometry on the intensity distribution in the vicinity of the aperture is studied in detail.

Coaxial probes for scanning near-field microscopy.

This paper deals with the development of coaxial aperture tips integrated in a cantilever probe for combined scanning near-field infrared microscopy and scanning force microscopy. A fabrication process is introduced that allows the batch fabrication of hollow metal aperture tips integrated on a silicon cantilever. To achieve the coaxial tip arrangement a metal rod is deposited inside the hollow tip using the focused ion beam technique. Theoretical calculations with a finite integration code were performed to study the transmission characteristics of coaxial tips in comparison with conventional aperture probes. In addition, the influence of the geometrical design parameters of the coaxial probe on its optical behaviour is investigated.
