Human naive and memory T-helper cells display distinct adhesion properties to ICAM-1, LFA-3 and B7 molecules.

In this paper the contribution of different accessory molecules to the adhesion of resting, naive and memory CD4+ T cells was examined utilizing a panel of CHO cell transfectants as model antigen-presenting cells (APCs). CD4+ T lymphocytes demonstrated strong adhesion to HLA-DR4 transfected CHO cells co-expressing B7, ICAM-1 or LFA-3 molecules, suggesting that all three adhesion pathways is utilized by resting CD4+ cells. Monoclonal antibodies (MoAbs) against the corresponding receptors on T cells, e.g. anti-CD28, anti-LFA-1 beta and anti-CD2, inhibited completely T-cell adhesion to natural ligands expressed on transfected CHO cells. Pretreatment of CD4+ T cells with NKI-L16 MoAb, which interact with an activation epitope on LFA-1 alpha chain, enhanced adhesion to ICAM-1 but not B7 or LFA-3-expressing CHO cells. Analysis of T helper-cell subsets revealed that memory T cells bound several fold stronger to ICAM-1 expressing transfectants compared to the CD4+ 45RA+ naive T cells, whereas adhesion to B7, LFA-3- and B7/LFA-3-expressing CHO cells was similar in both T-cell subsets. The kinetics of adhesion of naive and memory CD4+ T cells to ICAM-1 was rapid and similar in both subsets. The NKI-L16 MoAb multiplied several times ICAM-1-dependent adhesion in naive compared to memory cells, which enabled the naive cells to reach a similar adhesion level as memory cells. The results suggest that resting naive CD4+ T cells utilize preferentially the CD2/LFA-3 or CD28/B7 adhesion pathways upon adhesion to APCs, while memory CD4+ T cells utilize the CD2/LFA-3, CD28/B7 and LFA-1/ICAM-1 adhesion pathways. The NKI-L16 MoAb-induced upregulation of adhesion involves an increased affinity of LFA-1 for its ligand and not a change in the number of LFA-1 molecules. This is compatible with a view that naive cells express a large number of inactive LFA-1 molecules, whereas memory cells express preferentially activated LFA-1 molecules. The inherent low number of active LFA-1 molecules on naive CD4+ T cells may be important in keeping these cells in a resting state.

AIDS2: a decision-support tool for decreasing physicians' uncertainty regarding patient eligibility for HIV treatment protocols.

We have developed a decision-support tool, the AIDS Intervention Decision-Support System (AIDS2), to assist in the task of matching patients to therapy-related research protocols. The purposes of AIDS2 are to determine the initial eligibility status of HIV-infected patients for therapy-related research protocols, and to suggest additional data-gathering activities that will decrease uncertainty related to the eligibility status. AIDS2 operates in either a patient-driven or protocol-driven mode. We represent the system knowledge in three combined levels: a classification level, where deterministic knowledge is represented; a belief-network level, where probabilistic knowledge is represented; and a control level, where knowledge about the system's operation is stored. To determine whether the design specifications were met, we presented a series of 10 clinical cases based on actual patients to the system. AIDS2 provided meaningful advice in all cases.

Formal and population genetic studies of AHSG: further data from Galicia.

A sensitive immunodetection method for Alpha-2-HS glycoprotein (AHSG) after ultrathin layer polyacrylamide gel isoelectric focusing has been applied to a family study of 126 matings including 292 offspring. Formal genetic studies are in agreement with an autosomal mode of inheritance for this system. A population study of 506 unrelated individuals from Galicia (NW Spain) gave the following frequencies: AHSG*1 = 0.7559 and AHSG*2 = 0.2441, which correspond to a exclusion chance for non-fathers of 0.1505.

Distribution of F13A phenotypes in Spain: a particularly high frequency of the F13A*2 allele.

The distribution of the phenotypes for coagulation factor XIIIA subunit (F13A) of autochthonous individuals from the following five Spanish populations was studied: Galicia, Castilla-León, Castilla-La Mancha, Extremadura and Western Andalusia. The frequency values obtained for F13A*2 ranged from 0.248 to 0.311. To date, these values are the highest recorded in the world.

Simultaneous diagnosis of coagulation factor XIIA (F13A) and plasminogen (PLG) phenotypes by polyacrylamide gel isoelectric focusing.

In this paper, we report a simple rapid method for simultaneous determination of Coagulation Factor XIIIA (F13A) and plasminogen (PLG) phenotypes by PAGIF with a nominal pH range of 3.5 to 10, followed by immunofixation and silver stain. Critical considerations concerning the conditions of molecular separation and detection strategies are also presented.

Synthesis and conformational studies of carrabiose and its 4'-sulphate and 2,4'-disulphate.

Methyl alpha-carrabioside (13), and its 4-sulphate (19) and 2,4-disulphate (20) have been synthesised via glycosylation of methyl 3,6-anhydro-2-O-benzyl-alpha-D-galactopyranoside with 2,3,6-tri-O-acetyl-4-O-benzyl-beta-D-galactopyranosyl bromide and subsequent partial or complete debenzylation, sulphation, and deprotection of the resulting disaccharide derivatives. Conformational studies have been carried out on 13, 19, and 20 on the basis of 1D and 2D 1H-n.m.r. spectroscopy and molecular mechanics calculations.

Effects of ethnicity and child's age on maternal judgements of children's transgressions against persons and property.

Mothers (N = 256) of four different cultural groups judged the culpability of children whose actions were described in Piaget-type stories depicting commission of harm under various conditions. Child's age, harm to persons vs material objects, intentions, and amount of harm were systematically varied. All main effects (culture, intent of transgressor, age, and magnitude of harm) were significant, but several effects were qualified interactions. A cognitive social learning explanation is proposed to account for group differences in judgement, with special attention to cultural differences in values attached to harm to persons vs damage to property.

False-positive and indeterminate human immunodeficiency virus test results in pregnant women.

Guidelines from the Centers for Disease Control and Prevention, Atlanta, Ga, recommend that all pregnant women be offered human immunodeficiency virus (HIV) testing to ensure that they have the opportunity to use currently available therapeutic interventions to reduce the risk infecting their offspring with HIV. These recommendations have resulted in an increased number of low-risk women being tested and a significant rise in the percentage of false-positive results from HIV antibody screening tests and ambiguous (indeterminate) findings from confirmatory tests. Women receiving such results are generally in emotional turmoil yet must make treatment choices if they prove to be infected. This article provides guidelines to help general medical practitioners to understand the nature of HIV testing, to assess a woman's infection status when initial tests are ambiguous, and to determine when treatment is appropriate.

Alpha-2-HS-glycoprotein gene frequencies in Galicia: further evidence for a cline in European populations.

alpha 2-HS-glycoprotein (AHSG) phenotypes were determined in 506 unrelated autochthonous individuals from the Galician population (NW Spain) by means of isoelectric focusing followed by silver stain immunofixation. The phenotype frequency distribution fitted with the Hardy-Weinberg law. No evidence of intrapopulation heterogeneity was observed. The allele frequencies obtained were: AHSG*1 = 0.7559 +/- 0.013 and AHSG*2 = 0.2441 +/- 0.013. Our findings lend support to the hypothesis of an AHSG gene frequency cline in European populations. Other population genetics' considerations have also been discussed.

Plasminogen (PLG) gene frequencies in the Iberian Peninsula: heterogeneity analysis.

The distribution of Plasminogen (PLG) phenotypes in five populations of Spain (Galicia, Castilla-La Mancha, Castilla-León, Extremadura and Western Andalucia) is analysed by means of polyacrylamide gel isoelectric focusing and immunofixation. PLG*A frequencies, ranging from 0.7281 in Extremadura to 0.8038 in Castilla-León, are similar to those reported for other Southern populations of the European continent and higher than those in Northern and Central European populations. Heterogeneity analysis of the PLG allele distribution among different regions of the Iberian Peninsula have also been carried out.
