Immunohistochemical analysis of estrogen and progesterone receptors in benign breast diseases.

Surgical specimens of 62 patients with fibrocystic disease, 39 with fibroadenoma and 11 with intraductal papilloma were evaluated with ER-ICA and PgR-ICA methods. Positive nuclear staining was only observed in the epithelial components. The myoepithelium, myofibroblasts, fibroblasts and the so called apocrine epithelium, however, did not contain steroid receptors. Some steroid receptor negative cases occurred among the atrophic and non-proliferating fibrocystic diseases. In the proliferating benign alterations, particularly in those considered preneoplastic, e.g. radial scar, ductal and lobular atypical epithelioses, such as carcinoma lobulare in situ and florid intraductal papilloma there was a positive staining for estrogen receptor and progesterone receptor in the epithelial cells. Authors' results seem to serve as a theoretical basis for the hormonal influence of precarcinomatous cases and to explain the success of antisteroid therapy in patients with fibrocystic disease.

Clinical correlations of steroid receptors and male breast cancer.

Estrogen receptors (ER) were present in tumor specimens from 29 of 34 cases of male breast cancer. There was a significant negative correlation of ER concentration with age. The quantity of ER tended to correlate directly with progesterone receptor levels, disease-free interval, and response duration among responders, but not to a statistically significant extent. In 13 patients for whom response data were available, no significant correlation was observed between ER levels and either frequency or duration of orchiectomy response. Among the six patients with tumor ER levels of less than 30 fmol per mg of protein, however, only two brief responses to orchiectomy occurred that were of little clinical benefit, while three of seven patients with higher ER responded more favorably. Thus, although this suggests that a relationship between low ER and unfavorable orchiectomy response may emerge as more patients are studied, currently available data do not justify basing therapeutic intervention on ER status of a biopsy in a manner analogous to that used for female breast cancer. Nine of 14 male breast cancer patients had positive progesterone receptor assays and several had androgen or glucocorticoid receptors. Tissue from only three of ten men with gynecomastia had measurable ER, and these were limited to the 4S component on sucrose gradients.

Purification of an estrophilic protein from calf uterus.

A method has been devised for purifying the estrogen-binding protein ("uptake receptor") of uterine cytosol which circumvents the marked tendency of this receptor to aggregate during attempted isolation. Ammonium sulfate precipitation of the estradiol-receptor complex of calf uterine cytosol in the presence of calcium ions yields both an 8S complex and its 4S subunit which are stable during further purification by gel filtration and ion exchange chromatography. The major product is the 4S complex which does not revert to 8S in the absence of salt and which has been purified about 2500-fold. The approximate isoelectric points of these partially purified 8S and 4S complexes are 5.8 and 6.4, respectively.
