UNLABELLED: To assess the value of subrenal capsule assay (SRCA) for predictive chemo-sensitivity testing in breast cancer 24 primary tumours were examined in normal immunocompetent (NIC; AB- and B6D2-F1-) mice. In 6 cancers the effect of 0.25 mg Prednisolone mous X day (applicated with drinking water) was compared to the control group. Independent of the mice-species a significant increase of the grafts size occurred in 14 of 20 evaluable cancers (p less than 0.05). Histologically the increase of tumour size was caused by infiltrated host cells, only in 22 of 107 (20.6%) grafts vitale tumour cells were detectable. After immunosuppression the increase of the grafts size was significant lower, than in the no treated control group (p less than 0.05). In groups treated with Prednisolone only a thin inflammatory host cell infiltration was observed, but vital tumour cells were only seen in 3 of 29 (10.3%) grafts. CONCLUSION: Using normal immunocompetent mice subrenal capsule assay is not suited for predicting responsiveness of breast cancers to cytostatic drugs.
A cell line (NCE-G28) was established from the biopsy material of a human gliosarcoma of low histological differentiation. The initial cultures showed a mixed population of cells which in later stages became more uniform due to loss of slower growing constituents. The cells have been growing steadily for 20 months. A suspension of NCE-G28 cells injected s.c. as well as i.p. into nude mice produced solid tumors in all cases. Histologically these tumors closely resembled the original tumor. The original tumor, the nude mouse tumor, and NCE-G28 cells were immunochemically positive for glial fibrillary acidic protein as well as for neural plasma membrane antigen A2B5 expression. Two cell strains, 9B2C and 9B2E, were obtained by cloning of the initial cultures and another strain, NCE-G28T, was derived after explantation of a mouse heterotransplant. The two subclones were negative for glial fibrillary acidic protein expression but stained for cell surface fibronectin. NCE-G28T cells initially were positive for glial fibrillary acidic protein but lost this property within 8 months of cultivation. Karyotype analysis of NCE-G28 and the three strains revealed hyperdiploidy and six structurally altered marker chromosomes five of which were shared by nearly all cells. Receptors for epidermal growth factor were detected in all cell lines with the highest levels (about 300,000 receptors/cell) in the parental cell line. The epidermal growth factor receptors had an affinity of 2.5 nM (Kd) and by affinity cross-linking analysis a molecular weight of 170,000 was found. Initially, NCE-G28 cells responded to epidermal growth factor as well as fibroblast growth factor with increased rates of proliferation, while platelet derived growth factor had no effect. In higher passages the growth factor sensitivity was reduced. Using antibodies directed against synthetic protooncogene peptides the production of c-sis immunoreactive material was detected. NCE-G28 cells produce an autocrine factor which stimulated proliferation. This factor is present in conditioned medium and is active on cultured meningiomas and other glioma cell lines. NCE-G28 cells can be maintained in serum-free defined medium on plastic coated with fibronectin or an extracellular matrix from bovine corneal endothelial cells. The NCE-G28 cell line with its strains provide an in vitro model system in which the complexity of gliosarcoma cell populations and the interaction of the cloned cellular constituents can be studied.
Gliomas are heterogeneous in their cellular composition, affecting therapeutic efforts such as surgical removal, radiotherapy, chemotherapy and immunotherapy. 106 gliomas were taken into culture in our laboratory and 12 cell lines could be established there from. Experiments were carried out in as many early cultures as possible and with the constant experimental system of the cell lines. To subdivide and possibly classify the heterogeneous group of gliomas the following approaches emerged: Immunostaining of cells for glial markers such as GFAP, A4, A2B5, Leu 7 as well as fibronectin will allow one to distinguish different groups of glial cultures. Performance of growth factor sensitivity tests allows the assessment of major aspects of growth control in cultured gliomas. Cytogenetic evaluation in early cultures and correlation with the expression of oncogenes may yield useful information on mechanisms of escape from normal growth control. One of our cell lines (NCE-G28) in which cells switch from GFAP to fibronectin expression and transiently express the x-hapten may serve as a model to study crucial aspects of cellular differentiation. Using different extracellular matrices for the initiation of cultures even from very benign lesions with low proliferative potential it is possible to include these into comparative studies with glioblastomas.
Primary cell cultures were initiated from surgically obtained specimens from human gliomas. After 4-10 passages in vitro the cells from 18 individual cases were seeded onto cluster dishes and left to attach for 24 h in medium containing fetal calf serum. Thereafter the medium was changed to serumfree, defined conditions and the following growth factors were added: EGF 1.5 x 10-8 M, PDGF 35 ng/ml, FGF 80 pg/ml. The growth factors were added fresh every 2 days. After 9 or 11 days in culture, the cells on one multiwell-plate were counted. In 8 cases another set which was cultured also in a multiwell-plate but on glass cover slides was immunostained for glial fibrillary acidic protein (GFAP), fibronectin (FN), A2B5 and galactocerebroside (galC). The proliferative response pattern in consequence to the growth factor addition varied greatly between different cases. In all cases a pronounced proliferative response was accompanied by marked changes on the culture morphology. Usually the best proliferative response was obtained with PDGF (10 cases) or FGF (8 cases) whereas EGF was most effective only in one case. Two cases showed only minimal response, one of which was an oligodendroglioma D according to Ludwig and another a gliosarcoma which was re-evaluated after 1 year in culture. It could be noted, however, that FGF had a tendency to be less effective in the group of malignant astrocytomas. The staining pattern of the cultures with the afore mentioned markers was not affected by the growth factor treatment. The proliferative response usually resulted in increased staining for fibronectin and never an induction of GFAP. A2B5 staining was positive only in one case of gliosarcoma and galC staining was regularly negative.
Cytologic slides from 11 women with clinical, cytological and histological signs of a papillomavirus infection of the cervix uteri are investigated by cytophotometry and cytomorphometry. The mean values of DNA content, chromatin density, nuclear area and nuclear circumference of koilocytes were notably higher than for atypical or normal squamous cells. In those patients with concurrent cervical intraepithelial neoplasia the DNA content of koilocytes was inversely related to the degree of atypia. Squamous cells with HPV-induced lesions can be characterized by cytophotometry and cytomorphometry as a distinct cell population forming a continuum with the preneoplastic epithelium.

The effect of Cordemcura, a new cardiotonic substance, was tested on retrograd perfused isolated rabbit hearts in comparison with equieffective doses of dopamine. The substances were applied continously via the perfusion medium in the following dosages: Cordemcura 3.34 X 10(-4) mol/l, 2.67 X 10(-3) mol/l; dopamine 3.9 X 10(-6) mol/l, 1.6 X 10(-5) mol/l). The effects of the substances were studied during normoxic and hypoxic perfusion. The heart rate was increased under normoxic perfusion of Cordemcura about 37.1% (3.34 X 10(-4) mol/l) and 17.9 (2.67 X 10(-3) mol/l). The oxygen consumption in relation to the performance of the hearts (contractility. heart rate) was increased by Cordemcura on 1.45 and 1.93 (3.34 X 10(-4) mol/l and 2.67 X 10(-3) mol/l) in the same way like by dopamine (1.30 and 2.66; 3.9 X 10(-6) mol/l and 1.6 X 10(-5) mol/l). The content of adenine nucleotides decreased in the hypoxic perfused hearts on nearly 20% of the control values. There was no difference between control, Cordemcura and dopamine treated hearts. Cordemcura increased the level of cAMP in the myocardium after 90 min of hypoxic perfusion in comparison with control hearts (control hearts 2.8 +/- 0.7 nmol/g protein; Cordemcura 4.7 +/- 0.8 nmol/g protein). In comparison to control hearts and dopamine treated hearts the hypoxic perfusion with Cordemcura resulted in a significant lower release of lactate dehydrogenase from the heart.
The cardiotonic activity of Cordemcura was investigated in anesthetized pigs, cats, and rats. Hemodynamic measurements were made under normal conditions and under drug-induced heart failure (hexobarbital, isoproterenol). Cordemcura caused significant positive inotropic effects with rapid onset and relatively short duration of action (0.2-3.2 mg/kg i. v.). In the pig dp/dtmax was increased by 31.4% (p less than 0.02) at 1.6 mg/kg, in the cat by 34.2% (p less than 0.05) at 0.5 mg/kg, and in the rat by 14.4% (ns) at 0.5 mg/kg. The inotropic response was associated with a significant reduction of total peripheral resistance (TPR) (pig: -21.4%, p less than 0.02; cat: -13.7), and relatively small changes in heart rate, cardiac output, and blood pressure. Under drug-induced heart failure there was no further increase in the cardiotonic activity of Cordemcura. The hemodynamic effects of Cordemcura were compared with the effects of dopamine (5-100 micrograms/kg i. v.) and ouabain (10-40 micrograms/kg i. v.).
Report about a case of an ovarian metastasis of a small intestinal adenocarcinoma in a 25 year old woman. Diagnosis was difficult because of a simultaneous ovarian pregnancy on the same ovary. This is very seldom.
In this epidemiological, cytological and histological study signs of papillomavirus infections were found in 128 cases (16,6 per cent) and dysplasia or carcinoma in situ in 713 cases in a series of 773 cone biopsies. Epithelial changes caused by papillomavirus were combined with preneoplastic lesions in 116 cases. Pretherapeutic cytologic examinations of the condylomatous epithelial changes secured histologically later on referred to cervical papillomavirus infections in 64 per cent. Epidemiological inquiries of patients with cervical papillomavirus infection correspond extensively to those with invasive carcinoma of the cervix and premalignant cervical lesions.
In 8 patients with hypothalamic primary amenorrhea aged from 16 to 23 years (average age 19,4 years) a sequential stimulations test was performed with 0,5 g arginine hydrochloride per kg body weight, 25 micrograms gonadotropin-releasing hormone (GnRH) and 200 micrograms thyreotropin-releasing hormone (TRH). The response of the lactotropic, gonadotropic, thyreotropic and somatotropic cells of the pituitary was investigated. Serum levels of PRL, LH, FSH, TSH and HGH were determined by RIA. In all women hypoplastic ovaries were found by laparoscopy. In 7 patients tissue biopsies showed primordial follicles or primordial and secondary follicles, respectively. Investigations point to, that in hypothalamic primary amenorrhea at first the function of the gonadotropic and lactotropic cells of the pituitary is injured. The somatotropic cells could not be stimulated in 3 of 8 patients, the function of hypothalamo-pituitary-thyroid-axis in the stimulations test was normal in all women.

Report about the rare occurrence of ovarian metastases from a thymoma. Exstirpation of a lymphoepithelial thymoma was performed in a female patient at the age of twenty. 10 months afterwards a large tumour in the abdomen was the cause for a laparotomy. The situs showed a gigantic ovarian tumour (size 20 X 20 X 10 cm). The histological diagnosis was a metastatic tumour from the lymphoepithelial thymoma. Salpingo-Oophorectomy, radiotherapy and immunosuppressive therapy with glucocorticoids were used.

In 138 tokolytically treated cases of imminent abortion or premature birth the secundines were examined histologically and compared with 390 cases of pathological gravidities or births without tokolysis therapy. The frequency of occurrence of morphologically manifested circulatory disturbances following tokolysis therapy was equal to that in the untreated cases, the rate of ascendent infections was only slightly increased. In tokolytically treated placentas with the histological pattern of "dissociated disturbed maturation" and "Maturitias retarda" are prevailing. Thus in most cases sufficient supply of the fetus is possible and the critical phase of uteroplacental insufficiency with the danger of abortion or premature birth may be overcome. There were no hints at direct influences of tokolysis on placental morphology.
Cytologic findings and the histologic diagnosis are compared in 326 cervical cones. Out of the Pap III ("with cytologic control") group, cases named "Pap III with necessity for histologic diagnosis" were evaluated separately. In this subgroup of Pap III, we have found prestages or early stages of cervical cancer.
