Semen samples were collected by masturbation under asceptic conditions from men who had sired children within the past 6 mo (group A) and asymptomatic men attending an infertility clinic who had not sired children and whose wives were asymptomatic for infertility (group B). These 109 semen samples were analyzed and cultured for isolation of aerobic and anerobic organisms. Overall, 68% of the specimens had positive bacterial cultures: 54% of the samples from group A were positive and 73% from group B were positive. Mixed bacterial flora were isolated from both groups but in group B they were more varied and present in a higher colony count than in group A. Staphylococcus epidermidis, Staph. aureus, Corynebacterium species, Mycoplasma hominis, and Ureaplasma urealyticum were isolated from group A. Group B revealed these organisma plus Streptococcus pneumoniae type III, Strep. pyogenes group A, Strep. feacalis, Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, Pseudomonas aeruginosa, Candida albicans, Bacteroides species, Peptostreptococcus species, and Eubacterium species. Semen samples from group A were of better quality than those from group B. In addition, the antibacterial effect of seminal plasma from group A was greater than that from group B.
Rats were vasectomized, vasectomized and injected in the epididymis with Freund's complete adjuvant (FCA), or only injected with FCA without vasectomy. All were positive for sperm agglutinins, in contrast to sham-operated or untreated controls. Generally, the highest titers were found in vasectomized or unoperated animals given FCA, indicating that local granuloma formation promotes the production of sperm autoantibody, even in nonvasectomized rats.

Male Wistar Strain rats of known fertility were given subcutaneous dosage of three drugs, alpha-chlorohydrin, amino-alpha-chlorohydrin, and busulphan alone or in combination for a period of thirty days. Males were housed overnight with females at various times during the treatment and ninety days after cessation of treatment. There were significant differences in levels of glycerylphosphorylcholine in the epididymides of two treatment groups as well as differences in numbers of spermatozoa recovered from the reproductive tracts of female rats mated with males from four treatment groups. Treatment had no effect on the weights of the testes or sex accessory organs or sialic acid levels in the epididymis, fructose levels in dorso-lateral prostate and coagulating gland, or citric acid levels in seminal vesicle and ventral prostate of the treated males when compared to controls. The antifertility activity was probably mediated by an effect on epididymal spermatozoa and thereby subsequent change in sperm transport within the female genital tract.
Serum testosterone levels, testicular LH binding and the spermatogenic cycle were analyzed in rats 4 and 22 mo of age. With age, serum testosterone levels decreased from 3.2 to 0.63 ng/ml serum. There was no age related decline in testicular LH binding or changes in the spermatogenic cycle.

Scanning electron microscopy was used to observe surface fine structure of germinal epithelium and granulosa cells of human ovaries from 24 patients with polycystic ovaries and primary infertility. Synthetic LH-RH was administered intravenously and serum LH was measured at frequent intervals before and after LH-RH injection. The PCO patients were arbitrarily classified into two groups on the basis of ovarian morphology: typical PCO (Type I) with greater LH response than the lower LH response of a typical PCO (Type II). The germinal epithelium which did not completely cover the surface of the normal ovary was characterized by patchy areas of cells with and without dense microvilli. In normal preovulatory follicles, most granulosa cells were polyhedral in shape with smooth surfaces, whereas those facing the follicular cavity were elongated and flattened and were covered with material having a filamentous/reticular texture. In PCO the germinal epithelium surrounding the whole surface possessed dense microvilli, solitary cilia, and blebs, resembling the fetal ovary. In normal ovaries, the follicular cells were uniform in size and shape with microvilli and evaginations. The PCO cells have irregular size and shape with few microvilli.

Ureaplasma urealyticum (T-mycoplasma) was isolated more frequently and in heavier growth from cervical mucus (49%) than from vaginal fluid (34%). It was isolated in 24% of vaginal fluid samples and in 35% of cervical mucus samples from fertile women, and in 29% of vaginal fluid samples and in 47% of cervical mucus samples from infertile women. The incidence of infection was high following abortion or total hysterectomy and during pregnancy or oral contraceptive use. T-mycoplasma was also isolated from the vaginal fluid and cervical mucus of a woman with tubo-ovarian abscess, but was not present in women with Trichomonas vaginalis infection. U. urealyticum did not alter the physiophysiologic characteristics of vaginal fluid and cervical mucus or the sperm penetration and sperm viability in cervical mucus. Treatment with tetracycline eradicated the organism in 88% of the infected women. Pregnancies were recorded during a 6-month follow-up in 1 of 19 infertile women who were treated with tetracycline.

The epithelium of the female reproductive tract undergoes cyclic morphologic alterations in cell shape, apical microvilli, ciliation, and secretion. These changes are hormone-dependent. The endometrial fluid is made of (a) components from the transudation of the blood serum and (b) protein, carbohydrate and other metabolites synthesized within the endometrial cells and discharged through the apical cell membrane. Endometrial cells have several mechanisms of transport processes across the plasma membrane. These processes regulate cellular volume, nutrition, excretion and communication along the surface of the cell as well as the intracellular and extracellular communication. The endometrial secretions play a major role in the capaciation of spermatozoa, and the nutrition of the blastocyst.
In 26-day-old rats, follicles capable of ovulation were present 48 h after PMSG injection and they degenerated if not exposed to an ovulating dose of HCG. In such follicles 125I-labelled LH bound to the thecal and granulosa cells. By 60 h after PMSG, LH binding to the granulosa cells was reduced by 46% although these follicles retained their ability to ovulate. LH binding to the granulosa cells was lost in most follicles by 72 h and ovulation could not be induced. The thecal cells still possessed LH binding sites at 72 h after PMSG. HCG stimulation of these follicles resulted in disruption of the granulosa and the invasion of blood cells into the antrum.
