THE ISOLATION AND BIOLOGICAL ACTIVITIES OF RABBIT GAMMA M- AND GAMMA G-ANTI-SALMONELLA TYPHIMURIUM ANTIBODIES.

Rabbit gammaM- and gammaG-anti-S. typhimurium antibodies were isolated by combined immune specific and physical methods and some of their properties in immunological systems were measured. gammaM-Antibody was detectable at lower concentrations and revealed a higher specific activity than the gammaG-globulins. Indirect studies indicated that the gammaG-globulin was the more "avid" immunoglobulin. Treatment of the gammaM-globulin with the reducing agent 2-ME decreased but did not destroy the immune activity of the subunits. The results confirm the necessity of analysis of purified immunoglobulin antibodies to evaluate the significance of their biological properties secondary to their interaction with antigens.

LYSOSOMAL ACID HYDROLASES IN MICE INFECTED WITH BCG.

Experiments are reported dealing with the increase of lysosomal acid hydrolases induced by BCG infection. Acid hydrolases were determined quantitatively in peritoneal MP, liver homogenate, and plasma of normal and BCG-infected mice. A significant increase of acid phosphatase, beta-glucuronidase, and cathepsin was found in MP and liver homogenate of BCG-infected mice. In plasma also a significant increase of acid phosphatase and beta-glucuronidase was noticed. The results of the determination of the enzymes in centrifugally separated subcellular fractions of liver homogenate indicated clearly that the acid hydrolases associated mainly with the "large granular" fraction, which consists of mitochondria, lysosomes, and microsomes and that infection with BCG caused significant increase of the enzymes specifically in this fraction. Differences in the pattern of location among centrifugally separated fraction of liver homogenate were observed between acid phosphatase and the other two acid hydrolases. MP cultured in vitro doubled their acid phosphatases content within 24 hours, whereas beta-glucuronidase rather decreased in the same cells.

LYSOSOMAL ACID HYDROLASES AND HYPERREACTIVITY TO ENDOTOXIN IN MICE INFECTED WITH BCG.

Experiments are reported dealing with the correlation between activities of lysosomal acid hydrolases and hyperreactivity to endotoxin induced by BCG infection. Acid hydrolases were determined quantitatively in peritoneal MP, liver homogenate, and plasma of normal and hyperreactive mice. Mice infected with BCG not only exhibited a hyperreactive state to lethal effect of endotoxin, but also responded to endotoxin by rapid increase of acid hydrolases, especially of beta-glucuronidase, in the plasma; whereas control mice responded to endotoxin by almost no change in plasma acid hydrolases. The extent of increase of beta-glucuronidase in plasma of hyperreactive mice was shown to correlate fairly well with the degree of hyperreactivity to the lethal effect of endotoxin. Desensitization of such animals with endotoxin was found to cause a decreased response of plasma beta-glucuronidase parallel with decreased mortality. A large amount of PPD exerted the similar effect to that of endotoxin in hyperreactive mice. Furthermore, the effect of PPD was decreased by desensitization of such animals with endotoxin, a fact which suggests contamination of PPD with endotoxin.

Metabolism in infection: study on the enzymatic damage in kidney of guinea pig infected with Mycobacterium tuberculosis.

Malic dehydrogenase activity of the kidney homogenate from the normal and the tuberculous guinea pigs has been estimated. A defect in the electron transport chain has been detected at the level of flavoprotein-diaphorase system. A significantly high DPNase activity of kidney homogenate has also been found in the infected group. Niacin and phenazine methochloride could correct both the defects and improve the tetrazolium reduction of the homogenate in the infected group to the level of the normal activity. Oxidative phosphorylation by the kidney mitochondria from the tuberculous guinea pigs was found to be low and could not be improved by niacin and phenazine methochloride, unlike their effects on the reduction of tetrazolium. Results have been discussed in the light of the over-all intercellular economy and its relation to the symptom complexes in tuberculosis.

The effect of antibody on intracellular parasitism of Salmonella typhimurium in mononuclear phagocytes in vitro: prolonged survival of infected monocytes in presence of antibody.

The effect of antibody on the fate of Salmonella typhimurium within mononuclear phagocytes (MN) of rabbits was studied in vitro. Monocytes and bacteria were incubated either in absence or presence of antibody. After 45 minutes during which phagocytosis occurred infected cells were washed to remove extracellular bacilli and free antibody. The cells were then reincubated in a medium without addition of antibody, and the interaction between the MN and bacteria was followed, correlating bacterial viability and the morphology of the mixture. The following results were obtained. The anti-Salmonella antibody was not bactericidal even in presence of complement and did not enhance phagocytosis. Regardless of whether antibody was present or absent during phagocytosis, the bacteria appeared to multiply within the cells. When no antibody was present during phagocytosis the infected cells were severely damaged within a few hours of incubation, and extensive extracellular multiplication was dominating. When antibody was present during phagocytosis MN packed with bacteria persisted for a long time. Little or no extracellular growth occurred. It was possible to demonstrate the presence of the antibody within the infected MN, using the fluorescent antibody technique. The antibody appeared as a coat around the bacteria. Antibody entered the cells only during phagocytosis, presumably attached to the bacteria. The active factor of the immune serum was found in the gamma globulin fraction and reacted specifically with the somatic antigen of Salmonella typhimurium. The antiflagellar portion of the antiserum was not involved in the phenomenon described. It is concluded that this antibody protects monocytes against the effect of intracellularly located Salmonella.
