Tubular aggregates induced by anoxia in isolated rat skeletal muscle.

The fast-twitch extensor digitorum longus and the slow-twitch soleus muscles of the rat were incubated for 1 to 6 hours in vitro in oxygen-free medium or a medium containing potassium cyanide. Within 3 hours of incubation, characteristic structural alterations of the sarcoplasmic reticulum were observed in extensor digitorum longus muscle fibers. Concomitant with the disruption of the normal sarcotubular organization there was a selective clustering of sarcoplasmic reticulum elements with formation of tubular aggregates similar to those observed in human muscle under various pathologic conditions. The aggregates increased in size by apposition of new tubules at the periphery and acquired a regular hexagonal arrangement. Double-walled tubules, apparently derived from folding of sarcoplasmic reticulum tubules, were also contained in the aggregates. Electron-opaque material was seen connecting adjacent tubules. It is proposed that this material derives from the Z-bands which appeared largely extracted since early stages of anoxic injury. Tubular aggregates were not seen in the soleum muscle fibers incubated under the same conditions in vitro. The results indicate that tubular aggregates can be formed by simple rearrangement of the sarcoplasmic reticulum without proliferation of new sarcotubular elements.

Can a mixed damage interfere with DNA-protein cross-links repair?

Some photochemical and photobiological properties of 4,5',8-trimethylpsoralen (TMP) have been studied in comparison with 1,4,6,8-tetramethyl-2H-furo[2,3-h]quinolin-2 one (FQ) and 8-methoxypsoralen (8-MOP). TMP and FQ can photobind to mammalian cell DNA in vivo, by UVA irradiation, forming DNA-protein cross-links (DPC), but only TMP shows a strong capacity of inducing interstrand cross-links (ISC). The mechanism of DPC formation was studied using the double irradiation method in Chinese hamster ovary (CHO) cells, and DPC were detected by alkaline elution. Both TMP and FQ induce covalent diadducts linking together DNA and proteins. Studying the formation of double strand breaks (DSB) in CHO cells we observed that TMP induced a low amount of DSB, similar to 8-MOP. TMP and 8-MOP induced chromosomal aberrations in CHO cells to the same extent, while FQ appeared to be more active. Our data suggest that the ISC induced by TMP could trap enzymes involved in DPC repair.

Chromosomal aberrations in peripheral blood lymphocytes from patients with dermatomyositis and their first-degree relatives.

Chromosomal abnormalities in cultured peripheral lymphocytes from 14 dermatomyositis patients and their first-degree relatives were examined. Fifteen normal subjects were also studied. A low increase was observed in the frequency of chromosome aberrations in patients who had not received any medical treatment but not significantly higher versus the controls; the detected chromosome aberrations were in the form of acentric fragments and dicentrics. The patients who had received medication showed an increased frequency of dicentrics, acentric fragments and chromosome breaks, although the frequency was not significantly higher than that of normal subjects. There were no statistically significant differences in the number of chromosome aberrations between men and women patients with dermatomyositis. Only one family member, (2.22%) a boy of 18 years old who suffered from the same disease like his mother, was found to have chromosomal abnormalities in peripheral blood lymphocytes. The presence of autoantibodies found in the sera of Dm patients could not be correlated with the presence of chromosomal anomalies.

Micronucleus frequency in peripheral blood lymphocytes from patients with systemic lupus erythematosus.

In this study we evaluated the effect of immunosuppressive drug, cyclophosphamide, on lymphocytes from systemic lupus erythematosus (SLE), using the micronucleus test. Micronucleated peripheral blood lymphocytes were analyzed in cytochalasin B treated binucleated lymphocytes. There was significant increase in cells with micronuclei (P < 0.001) in SLE patients who had received cyclophosphamide (10.05 +/- 4.7) in comparison to the patients with SLE who had not received any medical treatment (6.24 +/- 2.42) and control normal subjects (5.48 +/- 2.24). The SLE patients who used cyclophosphamide showed a slightly higher frequency of micronucleated cells in the first 48 h after treatment. The present study emphasized the lack of correlation between age, sex and frequency of micronuclei.

Induction of chromosome aberrations in vivo in bone-marrow cells of mice by paracetamol.

The induction of chromosome aberrations by a common analgesic with antipyretic activity, Paracetamol, was investigated in vivo on bone-marrow cells of albino male mice. The drug was given orally or intraperitoneally in a single and cumulative dose series. The doses were the serial dilution of the highest one (400 mg/kg for intraperitoneal administration and 800 mg/kg for oral administration). Our study indicates that the cellular damage induced by Paracetamol leads to expression in chromosome aberrations such as gaps, chromatid breaks, acentric fragments and polyploid metaphases.
