This chapter provides a brief but reasonably detailed group of well-defined but difficult problems (with their potential solutions) that are presented on a regular basis to veterinary practitioners. The author stresses that prior to instituting corrective measures there is a need for client education and communication. The potential for increased costs in meeting the needs of the patient and animal owner must be emphasized.

Inhibition of insulin binding by concanavalin-A.

Prolonged incubation of IM-9 lymphoblastoid cells with concanavalin-A (Con-A) reduces insulin binding at concentrations of the lectin that neither agglutinate cells nor acutely inhibit insulin binding. Scatchard plot analysis suggested that Con-A treatment reduces insulin receptor number. Con-A-induced reduction in insulin binding resembles insulin "down regulation" in the time required for appearance and reversal of the binding alteration. The effect of Con-A on insulin binding was inhibited when the sugar alpha-methyl mannoside was incorporated into the incubation medium at the same time as Con-A. Even when the sugar was added after hours of Con-A exposure, the depressed insulin binding was reversed proportional to the reduction of Con-A binding to the cells by the sugar. This latter observation suggests steric hindrance as the mechanism for Con-A-induced reduction in insulin binding. The slow association rate of Con-A to the cells under the conditions of the experiments probably explains the prolonged exposure time to Con-A required for inhibition of insulin binding. Additional support for steric hindrance was observed in experiments in which insulin was "trapped" on cells after exposure to Con-A. Bivalent succinyl Con-A was only 10% as effective as the natural tetravalent lectin in reducing insulin binding. Cytoskeleton and protein synthesis inhibitors failed to alter the Con-A-induced reduction in insulin binding.

The forensic taxonomic debate on Cannabis: semantic hokum.

It has been asserted that there are legal species of marihuana plants, and this contention has generated frequent court challenges of criminal prosecutions involving marihuana. Invariably the claim is made that the name C. sativa used in legislation is insufficiently comprehensive to proscribe all forms of marihuana. The maneuver being used, alarmingly, is potentially applicable to innumerable other materials, but its success is based on a failure to appreciate the subjective nature of taxonomy and the little-known but critical ambiguities which are inherent in scientific names. The complex principles and operational conventions of biological nomenclature are presented in elementary fashion. Despite important technical constraints on the use of scientific names, some facts are clear: these names are used subjectively, they may be highly ambiguous, the consensus on use of these names is liable to change with time and, most important, quite permissibly they may have substantially different meanings to different users. The claim that there are legal species of Cannabis merely amounts to a semantic ploy in which certain of the variants of Cannabis that have customarily been understood to be denoted by the species name C. sativa, and which are clearly understood to be proscribed, are simply arbitrarily redefined as different species. This ploy has proven unsuccessful in all cases where scientific evidence was adequately presented by the state and in all important court cases where the issue was critically examined.

Hypotrichosis in Miniature Poodle siblings.

Progressive hypotrichosis was detected in 2 silver male Miniature Poodle siblings at 5 weeks of age. A male and 2 female siblings had normal black coats. The sire, a 3-year-old black Miniature Poodle, and the dam, a 2-year-old silver Miniature Poodle, also had normal coats. Microscopically, the hypotrichotic skin had accumulations of keratotic debris and melanin in dilated hair canals and melanin deposits in and around inactive hair bulbs. Sex-linked or sex-limited inheritance may have been involved, inasmuch as both affected dogs were males.

Isolation in Illinois of a foreign strain of Ehrlichia canis, the causative agent of canine ehrlichiosis (tropical canine pancytopenia).

Ehrlichiosis (tropical canine pancytopenia) was diagnosed in Illinois in a 4 1/2-year-old mixed breed dog that had spent the previous 2 years in India. The dog had periodic epistaxis, anemia, leukopenia, hypoalbuminemia, and hypergammaglobulinemia. A titer of 1:160 for Ehrlichia canis was detected by the indirect fluorescent antibody test (IFAT). Subinoculation of blood into a clinically normal dgo resulted in typical signs of canine ehrlichiosis, and E canis was identified in circulating mononuclear cells. The recipient died 23 days after inoculation, and typical gross and microscopic lesions of ehrlichiosis were found at necropsy. Ecchymoses and petechiae were found on the gallbladder, kidneys, small intestinal mucosa, cerebrum, and cerebellum. Plasmacytosis and perivascular cuffing was evident in various organs, especially the cerebrum, cerebellum, and meninges.

Interaction between levodopa and methyldopa.

The interaction between methyldopa and levodopa was studied in 18 patients with Parkinsonism. Together they produced a fall in blood pressure in doses which when given alone had no effect or only a slight hypotensive effect. Severe hypotension never occurred. It is reasonable to give methyldopa to hypertensive patients on levodopa but the regimen should be initiated in hospital.
