Is an amphiphilic region responsible for the haemolytic activity of Bacillus thuringiensis toxin?

The amino acid sequence of the 27 kDa protein responsible for the haemolytic activity of Bacillus thuringiensis subsp. israelensis toxin has been analysed by secondary structure prediction, helical wheel/net diagrams and molecular mechanics calculations. We found that segment 116-126 presumably forms a strongly amphiphilic alpha-helix. This is supported by the findings that the synthesized segment 116-126 (a) has a significant alpha-helical content in water, and (b) displays an in vitro haemolytic activity comparable to that of bee venom peptide melittin. As segment 116-126 is present in the haemolyzing, but not present in the non-haemolyzing proteins from B. thuringiensis toxins, we suggest that this segment is responsible for the lytic potential of the B. thuringiensis subsp. israelensis protein.

Evaluation of thyroid function and anti-thyroid autoantibodies in systemic sclerosis.

Parameters of thyroid metabolism, and the presence of anti-thyroid antibodies were investigated in 43 patients with systemic sclerosis. Anti-thyroid antibodies were detected in 14 cases. Elevated levels of anti-thyroglobulin antibodies were determined in 4 cases, anti-thyroid peroxidase (TPO) antibodies in 11, and anti-microsomal antibodies in 5. The detection of anti-TPO antibodies gave the most remarkable information about the presence of autoimmune thyroiditis. The patients with anti-TPO and/or reduced T3 concentration tended to have secondary Sjögren's syndrome. Our results provide further evidence that anti-thyroid antibodies might be responsible for the remarkable appearance of autoimmune thyroiditis in systemic sclerosis.

[Smoking--II. Duration of smoking and factors affecting smoking as a social phenomenon].

According to the poll of 5,555 individuals (91% of the expected number) over 40 years of age, selected from electoral registers in all three districts of the community of Mali Idjos in 1987, the average duration of smoking among active smokers increases with age, divergently with regard to sex. The age of smokers at the time they started smoking increases with age groups, from 22.2 to 33.2 in males and from 28.1 to 49 in females. The onset of smoking in the oldest male group in this rural area occurred in the first years after the war (first land confiscation) while in the group from 70 to 74 years of age it occurred in the years of compulsory crop-purchase system. Agglomeration in the onset of smoking in two male age groups (60-64, 65-69) occurred at the time of the second land confiscation. The onset of smoking among females from all age groups, from 45 years on occurred in the first decade of the sexual revolution (1961-1971). The prevalence of smoking in the studied population was different in previous decades as well. It changes according to social factors: from political and economic changes among males to the changes in women's social status among females.

[Smoking--I. Smoking habits, number of cigarettes smoked].

A multiphase, oncologic, population screening was carried out in 5.555 residents (91% of the expected number) over 40 years of age, from three districts of the community of Mali Idos. The poll showed that 83.1% of males and 21.9% of females are familiar with smoking (ex-smokers or active smokers); 45.7% of males and 11.6% of females are active and 37.4% of males and 10.3% of females are exsmokers; 314 residents--11% of males and 1.3% of females smoke more than one pack of cigarettes daily; some of them smoke more than 60, 80, even 100 cigarettes daily. In two age groups (40-44, 45-49) 100% of male respondents is familiar with smoking; in the group over 75 years of age this percent is reduced to one third (21.8%). We can conclude that the distribution of smoking in this population differed in the middle and in the second half of the century.

[Results of sonographic hip examination of infants in a risk group].

During sonographic examination of 770 newborns and infants the diagnostic value and the possibility of valuation of singular risk factors were investigated. According our data an acetabular dysplasia, requiring treatment can be found most frequently in the background of a limitation of abduction motion. Pathologic alterations may be found less frequently in groups with familiar anamnesis and with breech presentation. It could be stated that the aspecific click is a clinical symptom of significance and cannot be neglected whereas we could not prove connections between asymmetry of the folds and the sonographic data. Our results prove that the anamnesis and the clinical symptoms alone do not make a perfect recognition of the dysplasia possible; in consequence it is suggested that sonography should be used as a screening.

Isolation and partial characterization of a cystine-rich basic heparin-binding protein from bovine platelets.

We report the isolation and partial characterization of a so far unidentified basic platelet protein. Delipidated bovine platelets were extracted at pH 2.1. The extract was subjected to differential precipitation at pH 5.4-5.5 and by ammonium sulfate, then it was further purified by ion exchange chromatography on DEAE and CM cellulose columns in an urea containing medium. The major protein peak eluted from the CM cellulose column by NaCl gradient contained a protein in electrophoretically homogeneous form. It consists of a single polypeptide chain with an Mr of 28,000 as estimated by SDS PAGE. It was shown to be extremely rich in lysine and cystine and possessed a highly basic character (pI 9.8-10.1). On this basis the term cystine-rich basic protein (CRBP) was proposed for the new protein. Unlike some other low Mr basic proteins it did not bind calmodulin and troponin C, however, it showed significant heparin neutralizing activity.

The role of vasopressin in the pathogenesis of ethanol-induced gastric hemorrhagic erosions in rats. Is vasopressin an endogenous aggressor toward the gastric mucosa?

The role of vasopressin in the development of gastric hemorrhagic erosions induced by the oral administration of 1 mL of 75% ethanol in rats was studied. The area of the lesions in homozygous Brattleboro rats, having a defective vasopressin synthesis, was only 20% of that found in Wistar and heterozygous Brattleboro rats, which have normal vasopressin production. It is well known that vasopressin acts via the V1 (pressor) and V2 (antidiuretic) receptors. Administration of V1 and V2 vasopressin-receptor agonists and antagonists in this model showed that pressor-receptor activity is needed for the generation of all lesions in Wistar and heterozygous Brattleboro rats. Ethanol damage to the gastric mucosa was diminished by the V1 antagonist with similar efficacy as in the case of a vasopressin deficiency. Administration of the V1 antagonist and the absence of endogenous vasopressin were shown to protect the deeper layer of the gastric mucosa (assessed by histology) and to reduce significantly the ethanol-induced vascular injury and increase in vascular permeability (assessed by the monastral blue technique). Thus, endogenous vasopressin is clearly of great importance in the pathogenesis of gastric hemorrhagic lesions induced by ethanol. These results strongly suggest that vasopressin is an endogenous aggressor toward the gastric mucosa.

Derivative spectrophotometric assay of acetaminophen and spectrofluorimetric determination of its main impurity.

Second derivative UV spectrophotometry proved to be a useful tool for the determination of acetaminophen, even in the presence of high amounts of the impurity, 4-aminophenol and the background effect caused by dissolved excipients of some dosage forms. The fluorimetric method used for the estimation of 4-aminophenol impurity is less sensitive than the colorimetric methods used in the pharmacopeias. This can be attributed to an inner-filter effect.

[Value of ultrasonic diagnosis in transient coxitis].

In this paper the method of ultrasound examination of the hip and the results in 38 cases with acute and subacute hip complaints, suspect of transitory arthritis of the hip, are described. We could state that the increase of joint synovial fluid, causing typical complaints and clinical symptoms can be demonstrated with this method and its measure can be determined. The possibility to follow with objective method the whole time of regression of the process is also given. Based on our results we suggest the use of this method in the examination of cases suspect of the transitory arthritis of the hip as a supplement of the methods used until now.

Pathological and immunological study of goose embryos experimentally infected with duck plague virus.

A total of 240 embryonated goose eggs obtained from two susceptible flocks were used. Half of the eggs were inoculated into the allantoic cavity with a virulent strain (7593) of duck plague virus isolated from an acute outbreak, and the other half were inoculated with the attenuated vaccine virus (KAPEVAC). Ten, 100 or 1000 CPU/0.1 ml virus were given on days 12 and 20 of incubation. Embryos that died and surviving embryos killed at 5-day intervals were examined by light and electron microscopy. The yolk and the serum of embryos that survived until hatching were assayed for antibody content. Lymphocytes separated from the blood were used for the immuno-rosette formation and lymphocyte stimulation tests. Pathomorphological changes indicative of virus replication occurred in the liver, kidney, myocardium, gizzard muscle and chorioallantoic membrane (CAM) of the embryos in the case of both virus strains. The time of onset and severity of these changes and the time and rate of embryonic mortality depended on the virulence of the strain used for inoculation, the virus dose and the time of inoculation. Virus-neutralizing (VN) antibodies were demonstrable neither in the yolk nor in the serum of goslings exsanguinated after hatching. The lymphocytes recognized the virus antigen in the in vitro cellular tests and responded to it with blastogenic transformation. As opposed to adult birds, in the embryos duck plague virus infection did not cause damage to the digestive tract mucosa and the lymphoid organs.

[Ultrasonic examination of the hip joint in neonates and infants].

Sonography is a new procedure in the examination of the hip of newborns and infants. With this harmless, objective, image producing method, that can be used immediately after birth, the cartilaginous and bony development of the acetabulum, the intraarticular position of the femoral head, its coverage and its beginning decentralization, luxation can be investigated. With the use of this method such informations may be gained by the reproducible and image representation of the non osseous constituents of the joint that can otherwise be hoped previously only from invasive X-ray methods, meaning also radiological loading. This method must be considered of having great significance not only in the very early detection of dysplasias, but in the choice of the time and period of treatment and in the following of its effectiveness.

[Assessment, reproducibility and sources of error in sonographic examination of the hip].

To the sonographic examination of the hip middle-category devices with real-time working and 5 MHz linear probing heads are necessary. Special laying and documentation is suggested to the rapid and easy execution and assessment of the examinations. If the examination is carried out in a standard plane the points of orientation characterising the acetabulum are represented unequivocally and clearly, the results gained can be reproduced and compared.

Diagnostic Errors

[Granulomatous dermatitis caused by surfen in diabetics treated with Insulin-depot-S-Richter].

Two cases of local cutaneous reaction in diabetics treated with Insulin-depot-S-Richter are reported. Intracutaneous test and histological examination revealed that the granulomatous inflammation of the skin was due to surfen. Insulin-depot-S-Richter was changed to insulin preparation free from surfen (Monotard MC, Novo) and no further local reactions could be observed in the same patients.

Delayed-Action Preparations

[Direct and indirect immunofluorescence studies in congestive myocardial diseases].

Immunfluorescence examinations were made on 118 patients with congestive cardiomyopathy. Standard techniques of direct immunfluorescence (DIF) were employed on myocardial biopsy material in 61 cases, and of indirect immunfluorescence (IIF) on the serum in 89 cases. On the basis of the assumed aetiologies, the patients were classified into 6 groups: definitely alcoholic, presumably alcoholic, familial, peripartum, chronic myocarditic and idiopathic; studies were then made of the frequencies of pathological immunological processes in the individual groups. The highest incidences of DIF-positive cases were observed in the familial, peripartum, chronic myocarditic and idiopathic groups (67-87%). In the presumably alcoholic group the corresponding incidence was 37%, and in the definitely alcoholic group it was 25%, in contrast with expectations. Interestingly, very strong IIF-positive reactions too were observed in these alcoholic groups. It is possible that, besides the effect of alcohol, the fact that the immune background becomes pathological is also a significant factor in the emergence of the congestive cardiomyopathy in the alcoholic groups.

[Prediction of ovulation based on the serum estradiol level and the diameter of the follicle].

Experiences with the methods are summarized. Follicular growth was monitored by ultrasound folliculometry (USFM) and estimation of serum estradiol (E2 values in physiologic cycles. USFM was found to predict follicular rupture-ovulation--more precisely than E2 values. Cases of disturbed folliculogenesis (persisting and luteinized unruptured follicle, cysts) are described. The wider use of USFM in Hungary is urged, because appropriate equipment is available while the chances of continuous hormonal monitoring are rather limited.
