[Effect of surgical and combined treatments on the state of the blood coagulating system in cancer of the corpus uteri].

The study of blood coagulability in the coarse of combination (77 cases) and surgical (17 cases) treatment of endometrial carcinoma showed that surgery is followed by an increased plasma tolerance to heparin, a raised fibrinogen level and a lowered fibrinolytic activity. Therefore, surgery promotes blood coagulability, thus increasing the risk of thromboembolic complications in endometrial cancer patients in postoperative period. Preoperative hormonotherapy seems to contribute to this risk since it increases blood coagulability and delays its normalization in postoperative period.

[Sympathetic-adrenal system in various types of anesthesia in surgical treatment of gastric and rectal cancer].

Under study was the functional activity of the sympatho-adrenal system in identical surgical interventions in 44 gastric cancer patients and 77 rectal cancer patients, operated under nitrogen monoxide-ether, nitrogen monoxide-fluothane anesthesia and under neuroleptanalgesia supplemented with nitrogen monoxide. The sympatho-adrenal activity was assessed, taking into account the content of total catecholamines (adrenaline and noradrenaline) in blood and urine prior to surgery, intraoperatively before anesthesia, during the most traumatizing moment of the operation and at 1, 3, 6 day after it. It was found that in identical operative procedures stimulation of the sympatho-adrenal system is mostly manifest in patients operated upon under either anesthesia and neuroleptanalgesia. Use of fluothane narcosis in gastric cancer patients produced no marked activation of the sympatho-adrenal system, while in rectal cancer patients it was not so significant as in either narcosis and neuroleptanalgesia.

[Diagnostic value of determining spermidine in the urine of patients with melanoblastoma of the skin].

To estimate the diagnostic value of determination of one of polyamines (spermidine) in the urine the authors examined 58 patients with skin melanoblastomas of different localization and 10 healthy persons. It was found that in 16 of 25 patients having only primary tumor, in all 9 patients having primary tumor with metastases in regional lymph nodes, and also in 14 of 20 patients, who after previous therapy developed recurrence or metastases in regional lymph nodes, spermidine excretion exceeded its normal level. In three patients, subjected previously to radical treatment and considered as being clinically recovered, normal spermidine level was excreted. The data obtained enabled the authors to use this test as an adjuvant criterion in examination and treatment of such patients.

[Precursors of HLDF differentiation factor and ribosomal protein RPS21 have a common N-terminal sequence].

The mature differentiation factor HLDF, isolated from culture fluid, comprises 54 aa, whereas the open reading frame of mRNA encodes a 97-aa protein. We presumed that the protein translation begins from the first ATG codon, whose environment mostly meets the requirements for the initiation point. Two more ATG triplets are localized in positions 48-50 and 100-102, i.e., in the area preceding the cDNA fragment that encodes the N-terminal fragment of the mature protein. The mRNAs of HLDF and the S21 ribosomal protein have previously been shown to be highly homologous, and, therefore, their differences appear to be derived from two point deletions in the cDNA of the HLDF-encoding sequence (a G residue in position 112 and a C residue in position 224). As a result, the mature differentiation factor and RPS21 may be the products of translation from different open reading frames, the differentiation factor may be synthesized in the cell as a precursor, and its N-terminal sequence may be identical to that of RPS21. To test this hypothesis, we prepared recombinant RPS21 and the polyclonal antibodies to HLDF, full-size RPS21, and the C-terminal RPS21 peptide. Immunochemical staining by specially produced antibodies of native HL-60 cells and the same cells brought into apoptosis or differentiation confirmed that the precursor of the differentiation factor and the ribosomal S21 protein have a common N-terminal sequence and different cellular localizations. Neither an intron-containing gene nor a pseudogene with the nucleotide sequence corresponding to the HLDF cDNA was detected in the human genome or in the HL-60 cell line genome. On the basis of these facts, we propose a hypothesis of the molecular mechanism of the HLDF mRNA biosynthesis by means of posttranslational modifications of pre-mRNA of RPS21. [The use of the two-hybrid cloning in yeast for functional characterization of protein kinase MAK-V].

Identification of interaction partners opens a way to direct functional characterization of proteins. Several cDNAs coding for potential partners of protein kinase MAK-V/Hunk were isolated using two-hybrid cloning in yeast. Based on the partner properties, MAK-V/Hunk was assumed to play a role in tumorigenesis and tumor progression. With the previous results of two-hybrid cloning, MAK-V/Hunk was shown to participate in vesicular transport.
