[Diagnosis of cancer of the thyroid gland].

The clinical diagnosis of carcinoma of the thyroid gland (CTG) is a difficult and responsible problem. The importance of cytologic and histologic methods of examination has grown, they determine the therapeutic tactics and the volume of the operative intervention. Clinical examination and operations were carried out on 484 patients with various diseases of the thyroid. The examination included inspection of the thyroid gland, radioisotope scanning, radiological examination of the neck, mediastinum and lungs, ultrasonic study, and aspiration biopsy with further cytologic study of the material. Among the patients treated by surgery 58 (11.9%), 39 females and 19 males (2:1), were found to have CTG. Intraoperative emergency cytologic and histologic study was conducted in 30 patients. Differentiated CTG was revealed in all cases. Among the patients who were operated on 5 died from CTG in a 10-year follow-up period. The importance of aspiration biopsy, under conditions of an outpatient clinic in particular, is emphasized. The results of operative treatment of differentiated CTG show that early diagnosis and appropriate surgical intervention are conducive to a favourable prognosis in relation to the patients survival period.

[Electron microscopic study of various salts of proteoglycan aggregates].

Chemically isolated preparations of acid and normal Mg2+, Ca2+-, and guanidine+ salts of cartilage proteoglycan aggregates were investigated by electron microscopy. Some differences between acid and normal salts depend on the amount, valency and distribution of basic ion as well as between carboxyl and sulphate groups.

[Electron microscopic study of isolated proteoglycans].

Chemically isolated separate preparations of the non-aggregating protein-chondroitin-keratin sulphate (PCKS) fraction from the hyaline cartilage and hyaluronic acid (HUA) of the vitreous body and of the umbilicus were investigated by electron microscopy. PCKS and HUA in films without cytochrome c were present in the form of granules and differed by structural organization. The proteoglycans of the cytochrome c films were seen as finely-filamentous-cellular network, and were distinctly differentiated by their macromolecular organization. A mixture of both proteoglycans formed complexes as a result of a noncovalent interaction. Uranyl acetate ensured a good contrasting of proteoglycans, especially of PCKS, without cytochrome c.

[Regional features of the architectonics of the dermal fibrous framework of human skin].

Collagenous carcass of human derma is formed by interconnected fibrils, fibrillar fasciculi, fibers and their fasciculi. The latter form a three-dimentional network or spatial carcass (collagenous dermal pattern) subdivided into three main types (layer-forming, rhombic and complex-looped) and several mixed types of pattern. Every pattern has its own definite topographic-anatomical localization in the human body, depends on the functional loading experienced by every part of the body and is historically connected with the erect posture and labour activity.

[Effect of parenteral administration of melatonin on the biochemical composition of the rat granulation tissue].

The influence of long term parenteral melatonin administration on the biochemical composition of granulation tissue of surgical wounds in rats during healing was investigated. Physiological solution and solutions containing various concentrations of melatonin were subcutaneously injected to the animals within 3 weeks. Control and injected animals were wounded. Samples of gramilation tissue were investigated on the 5-th and the 8-th day of healing. The contents of oxyprolin, uronic acids, hexosamines, total lipids and their fractions, fractional composition of glycosaminoglicans and proteins composition of salt extracts were determined in the se samples. Repeated injections during three weeks caused the changes in biochemical composition of researched samples which were characteristic for stressful reaction of connective tissue. The specific changes are most expressed at long term introduction of a physiological solution to animals. The introduction of melatonin during similar period cansed protective effect, partially defending biochemical composition of granulation tissue from changes, which were induced by stressful situation.

[Characteristics of the activation of lysosomal enzymes in the rat liver after death and during resuscitation].

Alterations in activity of lysosomal acid phosphatase and cathepsin D, as well as of hepatocyte ultrastructure were studied in rat liver tissue after experimental heart arrest within 10 and 30 min and during the early post-resuscitation period. Cathepsin D free activity in a supernatant fraction as well as both enzymatic activities in lysosomal fraction were increased after 30 min heart arrest. Activity of acid phosphatase and cathepsin D in lysosomes was decreased, while the activity of the free enzymes was increased within 1 and 4 hrs after resuscitation. Triton X-100 (0.025%) caused labilization of lysosomal membranes. Alterations in ultrastructure of hepatocytes were observed within 30 min of the heart arrest and within first hour of the post-resuscitation period. The lysosomal membranes tended to normalization within 24 hrs after the post-resuscitation period, whereas the enzymatic activity remained elevated. Role of lysosomes in regulation of intracellular metabolism is discussed.
