Space-domain lock-in amplifier based on a liquid-crystal spatial light modulator.

We present a two-dimensional (2D) spatial lock-in amplifier that provides a contrast ratio of more than 10,000:1 for transmitted and blocked intensity patterns using a conventional liquid-crystal spatial light modulator. The device is based on spatial-domain modulation-demodulation of intensity patterns under coherent imaging conditions. The operation of the 2D lock-in amplifier is illustrated by implementing Young's double-slit arrangement for measurements of the mutual coherence between individual emitters of a 2D phase-coupled array of vertical cavity surface emitting lasers.

Synthesis of novel poly(dG)-poly(dG)-poly(dC) triplex structure by Klenow exo- fragment of DNA polymerase I.

The extension of the G-strand of long (700 bp) poly(dG)-poly(dC) by the Klenow exo(-) fragment of DNA polymerase I yields a complete triplex structure of the H-DNA type. High-performance liquid chromatography analysis demonstrates that the length of the G-strand is doubled during the polymerase synthesis. Fluorescence resonance energy transfer analysis shows that the 5' ends of the G- and the C-strands, labeled with fluorescein and TAMRA, respectively, are positioned close to each other in the product of the synthesis. Atomic force microscopy morphology imaging shows that the synthesized structures lack single-stranded fragments and have approximately the same length as the parent 700 bp poly(dG)-poly(dC). CD spectrum of the polymer has a large negative peak at 278 nm, which is characteristic of the poly(dG)-poly(dG)-poly(dC) triplex. The polymer is resistant to DNase and interacts much more weakly with ethidium bromide as compared with the double-stranded DNA.

Three-dimensional analysis of mode discrimination in vertical-cavity surface-emitting lasers.

Optical mode discrimination in vertical-cavity surface-emitting lasers that contain distributed Bragg reflectors (DBRs) and a spatially limited gain medium is analyzed numerically. It is assumed that the output field is linearly polarized owing to gain selectivity. The analysis employs a three-dimensional model and an angular spectrum of plane-wave decomposition with the proper polarizations. Two types of round aperture are considered, namely, a Gaussian aperture and a ring-peak aperture that represents gain saturation. Coupled with the DBRs, the former aperture yields nearly Laguerre-Gaussian modes, whereas the latter aperture significantly distorts the mode shapes. In both cases, narrowband DBRs provide the best mode discrimination.

Long term behavior of lithographically prepared in vitro neuronal networks.

We measured the long term spontaneous electrical activity of neuronal networks with different sizes, grown on lithographically prepared substrates and recorded with multi-electrode-array technology. The time sequences of synchronized bursting events were used to characterize network dynamics. All networks exhibit scale-invariant Lévy distributions and long-range correlations. These observations suggest that different-size networks self-organize to adjust their activities over many time scales. As predictions of current models differ from our observations, this calls for revised models.
