The radioimmunoassay of a pregnancy specific-beta 1-glycoprotein in plasma as a pregnancy test for subfertile women.

We used a pregnancy specific-beta 1-glycoprotein (SP1) radioimmunoassay to monitor 72 menstrual cycles of 38 apparently subfertile women who were trying to become pregnant. Blood samples were taken up to day 42 from the start of the previous menstrual cycle. Using serum SP1 levels greater than 6 micrograms/1 as indicative of pregnancy, we obtained 16 positive results of which 11 were later confirmed by a human chorionic gonadotrophin haemagglutination pregnancy test. Three of the five women whose pregnancies were not confirmed had a subsequent history of spontaneous abortion.

Kinetic immunoturbidimetry: the measurement of pregnancy specific beta 1 glycoprotein.

The extension of kinetic immunoturbidimetry to the measurement of low concentration proteins has been described using the protein pregnancy-specific beta 1 glycoprotein. The technique has a precision superior to that of all other methods currently available. The assay is rapid, cheap and compares well with other published methods. The assay further emphasises the usefulness of the technique of kinetic immunoturbidimetry.

The relation between pregnancy specific beta 1 glycoprotein (SP1) levels and abnormal glucose tolerance in pregnancy.

Pregnancy specific beta 1 glycoprotein (SP1) levels were measured in 54 potentially diabetic patients in the third trimester of pregnancy using a laser nephelometric method. All patients were tested by the 50 g oral glucose tolerance test. The results show that a significantly greater number of patients with abnormal glucose tolerance had SP1 levels above the mean compared with patients with normal glucose responses. All patients with a diabetic type response had SP1 levels above the mean. There was no significant difference in SP1 levels in the fasting and two-hour plasma samples. The study suggests that patients with asymptomatic glucose intolerance in pregnancy have higher SP1 levels than patients with normal glucose tolerance. It is possible that one of the factors controlling the placental output of SP1 is the maternal blood glucose level.

Beneficial effect of post -thawing osmoconditioning on the recovery of cryopreserved coffee (Coffea arabica L.) seeds.

Osmoconditioning-controlled rehydration of seeds in a solution with low osmotic potential -has been shown to reinvigorate aged seeds. The present work aimed at investigating the effect of osmoconditioning on the germination of cryopreserved seeds of Coffea arabica, whose viability and vigour are drastically affected by cryopreservation. For cryopreservation, seeds were desiccated to 0.21 g H2O/g dw, cooled at 1 degree C/min to -50 degree C, then immersed rapidly in liquid nitrogen. After rapid rewarming, seeds were osmoconditioned for 1 to 6 weeks using solutions with osmotic potentials between -1 and -4 MPa. The time to produce half of the final percentage of normal seedlings, T50, was about three fold lower with osmoconditioned seeds than with non-osmoconditioned seeds (12-14 d vs 36 d). Moreover, after a 6-week osmoconditioning treatment with solutions with osmotic potential of -1 and -1.25 MPa, the percentage of seedlings recovered from cryopreserved seeds was 64-74%, against 13-16% only for cryopreserved seeds which were not osmoconditioned.
