DNA DSB induced by iron ions in human fibroblasts: LET dependence and shielding efficiency.

This paper reports on DNA DSB induction in human fibroblasts by iron ions of different energies, namely 5, 1 GeV/u, 414 and 115 MeV/u, in absence or presence of different shields (PMMA, Al and Pb). Measure of DNA DSB was performed by calibrated Pulsed Field Gel Electrophoresis using the fragment counting method. The RBE-LET relationships for unshielded and shielded beams were obtained both in terms of dose average LET and of track average LET. Weak dependence on these parameters was observed for DSB induction. The shielding efficiency, evaluated by the ratio between the cross sections for unshielded and shielded beams, depends not only on the shield type and thickness, but also on the beam energy. Protection is only observed at high iron ions energy, especially at 5 GeV/u, where PMMA shield gives higher protection compared to Al or Pb shields of the same thickness expressed in g/cm2.

Studies on homocysteine and dehydroepiandrosterone sulphate plasma levels in Alzheimer's disease patients and in Parkinson's disease patients.

Homocysteine (HC) and dehydroepiandrosterone sulphate (DHEAS) plasma levels have been evaluated in groups of male and female patients with Parkinson's disease (PD) and in a group of female patients with Alzheimer's disease (AD) and compared with the corresponding plasma levels observed in a group of age-matched subjects. It has been confirmed that HC plasma levels are enhanced in both PD and AD patients. As far as the DHEAS plasma levels are concerned no changes have been observed in PD patients while a marked decrease has been observed in AD patients. These results support the view that while the pro-oxidant effects of HC and its agonist action at NMDA receptors can play a role in both neurodegenerative diseases, the role of DHEAS is more complex and may be an important factor only in certain neurodegenerative diseases. Thus, according to the present study DHEAS is likely to be involved in AD but not in PD.

DNA fragmentation induced by Fe ions in human cells: shielding influence on spatially correlated damage.

Outside the magnetic field of the Earth, high energy heavy ions constitute a relevant part of the biologically significant dose to astronauts during the very long travels through space. The typical pattern of energy deposition in the matter by heavy ions on the microscopic scale is believed to produce spatially correlated damage in the DNA which is critical for radiobiological effects. We have investigated the influence of a lucite shielding on the initial production of very small DNA fragments in human fibroblasts irradiated with 1 GeV/u iron (Fe) ions. We also used gamma rays as reference radiation. Our results show: (1) a lower effect per incident ion when the shielding is used; (2) an higher DNA Double Strand Breaks (DSB) induction by Fe ions than by gamma rays in the size range 1-23 kbp; (3) a non-random DNA DSB induction by Fe ions.

Influence of a low background radiation environment on biochemical and biological responses in V79 cells.

We present the results of an experiment aimed at comparing the effects of different background radiation environments on metabolism and responses to gamma-rays and cycloheximide of cultured mammalian cells. Chinese hamster V79 cells were maintained in exponential growth in parallel for up to 9 months at the Istituto Superiore di Sanità (ISS) and at the INFN-Gran Sasso underground Laboratory (LNGS) where exposure due to gamma-rays and to radon was reduced by factors of about 70 and 25, respectively. After 9 months the cells grown at the LNGS (cumulative gamma dose about 30 microGy, average radon concentration around 5 Bq/m(3)), compared to the cells grown at the ISS (cumulative gamma-ray dose about 2 mGy, average radon concentration around 120 Bq/m(3)), exhibited i). a significant increase of the cell density at confluence, ii). a significantly higher capacity to scavenge organic and inorganic hydroperoxides but a reduced scavenging capacity towards superoxide anions and iii). an increase in both the basal hprt mutation frequency and sensitivity to the mutagenic effect of gamma-rays. The cells grown at the LNGS also showed a greater apoptotic sensitivity starting at the third month of culture, that was no longer detected after 9 months. Overall, these data suggest a role of background ionizing radiation in determining an adaptive response, although they cannot be considered conclusive.

Inactivation of human cells exposed to fractionated doses of low energy protons: relationship between cell sensitivity and recovery efficiency.

Within the framework of radiation biophysics research in the hadrontherapy field, split-dose studies have been performed on four human cell lines with different radiation sensitivity (SCC25, HF19, H184B5 F5-1 M10, and SQ20B). Low energy protons of about 8 and 20 keV/micron LET and gamma-rays were used to study the relationship between the recovery ratio and the radiation quality. Each cell line was irradiated with two dose values corresponding to survival levels of about 5% and 1%. The same total dose was also delivered in two equal fractions separated by 1.5, 3, and 4.5 hours. A higher maximum recovery ratio was observed for radiosensitive cell lines as compared to radioresistant cells. The recovery potential after split doses was small for slow protons, compared to low-LET radiation. These data show that radiosensitivity may not be related to a deficient recovery, and suggest a possible involvement of inducible repair mechanisms.

Cardiac and humoral changes induced by recreational scuba diving.

The aim of this study was to evaluate the prevalence and the possible clinical relevance of circulating bubbles after a recreational scuba dive. Twenty healthy subjects (18 male, 2 female; age range 25-36 yr) underwent a Doppler-echocardiographic study in basal conditions and 1.9+/-0.2 h after a recreational scuba dive. Venous blood samples were taken just before the two ultrasonic studies to obtain leukocyte and platelet counts and plasma activity of angiotensin-converting enzyme (ACE; assumed as pulmonary endothelial damage marker). Circulating bubbles were observed in the right heart chambers of 12 subjects after the dive. The echocardiographic and humoral data were evaluated before and after diving in subjects with and without circulating bubbles. At the postdive evaluation, a significant increase in right ventricular dimensions (37.4+/-3.9 vs. 40.7+/-4.0 mm; P < 0.01) and a significant reduction of early diastolic filling velocities of both right (59.1+/-16.4 vs. 48.9+/-6.9 cm x s-(-1); P < 0.05) and left (76.2+/-9.9 vs. 67.5+/-10.2 cm x s(-1); P < 0.02) ventricle were observed in the group with circulating bubbles. In the same group, significant increases in ACE activity (92.9+/-41.1 vs. 105.9+/-41.7 U x liter(-1); P < 0.05), platelets (217+/-34 vs. 232+/-35 10(3) x microl(-1); P < 0.01), and granulocytes (3,704+/-715 x microl(-1) vs. 5,212+/-1,995 x microl(-1); P < 0.001) were observed. The bubble-free group showed only a postdive significant decrease of left ventricular early diastolic filling velocity (74+/-6.8 vs. 62.6+/-4.5 cm x s(-1); P < 0.005). These data may indicate that circulating gas bubbles are associated with cardiac changes, suggesting a right ventricular overload and an impairment of ventricular diastolic performance. Postdive humoral and hematologic changes are consistent with the hypothesis that "silent" gas bubbles may damage pulmonary endothelium and activate the reactive systems of the human body.

[Blastocystis hominis infection: a case report].

Blastocytis hominis has long been described as a non-pathogenetic protozoan parasite until recently, when claims have been made that it could be the cause of intestinal disorders. The clinical picture of B. hominis consists of non specific abdominal pain, watery diarrhea, anorexia, vomiting and weight loss. Rarely a more invasive form of the disease with rectal bleeding can occur. We describe the case of a ten year-old girl who was admitted to our hospital for diarrhea, abdominal pain and fever. The presence of B. hominis was demonstrated in her stools. The patient responded favourably to treatment with metronidazole. We feel that our observation is an additional support to recognition of B. hominis as a human pathogen.

[Hypnosis in psychosomatic medicine].

Hypnosis is a form of psychotherapy open to any physician who has been suitably trained. Its diffusion is to be hoped for, especially since all doctors today consider that they should handle their patients from the psychological side as well, in keeping with the dictates of modern psychosomatology. Reference is made to the main indications for hypnosis and its contraindications, the optimum technique according to Spiegel, the desirability of resolving symptoms without excessive concern for that which they hide, symbolise, or defend, and the way in which hypnosis can be inserted at both the diagnostic and therapeutic level within the present stage of the history of psychosomatic medicine.

[Role of hypnosis in psychosomatic medicine].

In Italy, the attitude displayed by psychosomatists towards hypnosis and A.T. is decidedly favourable, since they are considered as forms of short-term psychotherapy, this being primarily indicated in the management of psychosomatosis. While some reservations may be expressed with regard to the employment of hypnosis solely for the suppression of psychosomatic symptoms before their full significance is assessed, support may be given for the insertion of hypnosis in a wider psychotherapeutical context that is psychodynamically oriented. Psychogenesis may have different degrees of depth and not all forms of psychosomatosis are open to hypnotic attack, or even to short-term psychotherapy. Psychological therapy is only effective in cases where it releases the patient for his neurotic, infantile way of life and allows him to mature in a suitable manner. A result of this kind can hardly be reached in a short period of time. Hypnosis is, on the other hand, strongly indicated in a number of situations; these are listed.
