Structure-sensitive resonance Raman bands of capped tetraphenylporphyrinatoiron complexes.

Resonance Raman spectra have been determined for the capped tetraphenylporphyrinatoiron complexes and their base adducts. The five-coordinate mono-base complexes of these porphyrin complexes display Raman spectra that are nearly identical to those previously determined for five-coordinate iron(II) tetraphenylporphyrin complexes in the marker regions around 1540, 1345, and 370 cm(-1). A similar correspondence is found between Raman bands of iron(II) complexes of the capped complexes and those of tetraphenylporphyrin. By contrast, the unusual pseudo-six-coordinate iron(II) complexes more closely resemble high-spin five-coordinate systems in the 1540 and 1345 cm(-1) regions, but in the 380 cm(-1) region the resemblance is closer to the low-spin six-coordinate tetraphenylporphyrin complexes. These results provide spectroscopic confirmation for the unique nature of these pseudo-six-coordinate complexes.

Sulfur base ligation to iron(II) and cobalt(II) porphyrins.

Equilibrium constants for the binding of sulfur bases to cobalt(II) porphyrins were measured in toluene solution by a spectrophotometric method. The order of decreasing binding strength of sulfur ligands to cobalt(II) porphyrins was found to be mercaptide ions >> thioethers > mercaptans. It is suggested that a similar stability order of these sulfur ligands should exist towards iron(II) porphyrins, but formation constants could be obtained only for the mercaptide ions.

Expression of calcitonin gene-related peptide in medullary thyroid cancer.

We studied the expression of calcitonin (CT) and calcitonin gene-related peptide (CGRP) in 18 patients with medullary thyroid cancer (MTC) in the neoplastic (primary or metastatic) tissue by immunohistochemistry and in the plasma by radioimmunoassay. CT immunoreactivity was found in 100% of the primary and metastatic MTC, CGRP was expressed in 66% of the primary tumors and in 73% of the metastases. Both the number of positive cells and the degree of staining were always higher for CT than for CGRP staining. While plasma CT concentrations were always increased in patients with metastases, 3 patients with metastases had undetectable plasma CGRP levels. A positive correlation was found between plasma CT and CGRP levels. These data indicate that CGRP is frequently expressed in MTC sections and that plasma CGRP measurement is an additional marker for MTC, although has no advantage with respect to CT measurements in monitoring the progression of the disease.

Modulation of neoangiogenesis in bronchial preneoplastic lesions.

We have previously demonstrated that vascular count significantly increases in the preneoplastic lesions of the bronchial tree, starting from very low levels in the normal epithelium to a significantly higher number of microvessels in moderate dysplastic lesions and in situ carcinomas. Vascular endothelial growth factor (VEGF) protein expression has shown to be strictly associated with neovascularization both in human cancer and in various type of preinvasive lesions. A number of studies have demonstrated that mutant p53 is involved in the regulation of angiogenesis, and immunohistochemical detection of the p53 protein is associated with p53 gene mutations. In this study we looked for possible correlation between p53 protein detection, VEGF expression and vascular count in a series of preneoplastic and neoplastic lesions of the bronchial tree in order to investigate the angiogenic pattern and its genetic control in the early steps of bronchial cancer development. Twenty-four retrospective bronchial lesions with different grades of dysplasia and a case of normal bronchial epithelium were analysed. Surgical specimens removed from patients either confirmed, or suspect for lung carcinoma were stained immunohistochemically for CD34, VEGF, and p53. There were significant increases in microvascular density (MVD), VEGF, and p53 expression from normal bronchial epithelium through moderate dysplasia to in situ carcinoma to invasive cancer and these factors were significantly associated with moderate dysplastic lesions. A statistically significant difference was observed in MVD between hyperplastic-metaplastic, moderate dysplastic lesions and in situ carcinoma. A similar pattern was also observed for VEGF and p53 protein expression but no significant difference was observed between moderate dysplastic lesions and in situ carcinoma with regard to VEGF protein expression. The association between MVD, VEGF expression, p53 mutations and preinvasive lesions of the bronchial tree suggests that neoangiogenesis is early in non-small cell lung cancer (NSCLC) development and that p53 may have an important role in promoting angiogenesis in this human model of carcinogenesis.

Optimal treatment for T1-3NOMO small cell lung cancer: surgery plus adjuvant chemotherapy.

To improve treatment results in patients with surgically resectable T1-3NOMO small cell lung cancer, 16 patients were randomized to receive either neoadjuvant (preoperative) or adjuvant (postoperative) chemotherapy (cyclophosphamide, epirubicin, and etoposide). The overall survival is 45% at a median follow-up of 28.5 months. Patients treated with surgery plus adjuvant chemotherapy (n = 8) showed a significantly higher median and overall survival, proportion of 2-year survivors, and lower systemic relapse rate. These results indicate that the optimal treatment for T1-3NOMO small cell lung cancer is represented by the combination of surgery plus adjuvant chemotherapy.

Parathyroid carcinoma. Report of a case.

Parathyroid carcinoma is a rare cause of hyperparathyroidism. Its prognosis is mainly related to an en-bloc resection of the tumor and of the involved structures. However because of the small number of cases reported in the international literature, biological behaviour of the disease is not well known. In this report we describe one case of parathyroid carcinoma seen in our Institution. Preoperative presumption of parathyroid carcinoma thanks to clinical features and ultrasound imaging helped us in deciding an adequate surgical resection.

[Hematoma and fat necrosis of the breast: mammographic and echographic features].

The mammary gland is a common site for traumas which often lead to the formation of intraglandular hematomas. The abundance of fat tissue in this gland accounts for the relatively high frequency of fat necrosis and self-digestion of fat by lipases after traumas, i.e., fat necrosis, cystosteatonecrosis, lipophagic granuloma. Broadly speaking, hematomas are easily diagnosed by both mammography and US. However, scarring and the formation of lipophagic granulomas due to hemorrhage result in the diagnostic features which are difficult to distinguish from malignant breast nodules. In our study, we examined 40 patients with a clinically detected nodule and/or hematoma with skin retraction or thickening related to trauma. In 26 women, the trauma dated to 7-60 days before our observation, in 4 women to a year before and in 4 other women to more than two years before; 6 women had undergone breast surgery in the last two years. Mammography and US were performed at first observation; later, every six months, the patients were followed-up with US and a single targeted radiograph. The mammographic features at first observation were classified as follows: a single nodule (9 patients, 22.5%), a patchy nodule (7 patients, 17.5%), diffusely increased gland density (8 patients, 20%), radiolucent nodules (10 patients, 25%), nodules with calcifications (6 patients, 15%), no findings (2 patients, 5%). US demonstrated a fluid collection in 12 patients (30%), a solid nodule in 6 patients (15%), a cystic nodule in 10 patients (25%), diffuse parenchymal abnormalities in 4 patients (10%), calcified nodules in 6 patients (15%) and no findings in 2 patients (5%). US-guided needle biopsy was performed in 10 patients. Five patients underwent surgical biopsy: 4 had a lipophagic granuloma and the other one a chronic inflammation. Follow-up at two years allowed hematoma evolution to a scar or cystosteatonecrosis to be monitored. To conclude, the authors analyze the diagnostic problems related to the different mammographic and US patterns trying to suggest, on the basis of their own experience, the best imaging follow-up for breast trauma patients.

c-Ha-ras transfection and expression of MDR-related genes in MCF-10A human breast cell line.

BACKGROUND: We studied the relationship between ras transfection and the drug resistance phenotype in the MCF-10A cell line (a non-transformed immortalized human breast epithelial cell line). MATERIALS AND METHODS: Parental (MCF-10A) and c-Ha-ras transfected (MCF-10A H) cell lines were tested for resistance to Doxorubicin, Maphosphamide and Cisplatinum, by evaluating the inhibition of [3H]-thymidine incorporation. Cells were also examined for the expression of different resistance mechanisms by immunocytochemical and RT-PCR methods. RESULTS: ras-transfected cells were much more resistant to Cis-platinum than parental ones. GST-pi the GST isoenzyme frequently involved in human cancers) increased only in transfected cells. No expression of any other resistance mechanisms (MDR, MRP, LRP) was found by immunocytochemistry either in parental or in transfected cells; nevertheless, the more sensitive RT-PCR tests detected mrp products in parental and in transfected cells: gene expression levels were low and equivalent in both cell lines. CONCLUSIONS: ras transfection can induce resistance to Cis-platinum by increasing GST-pi expression.
