[Detection of human papillomavirus DNA by molecular hybridization in tube: interest in cervical neoplasia].

The presence of human papillomavirus (HPV) DNA in 79 cervical specimens obtained from 70 patients was studied by using a molecular hybridization technique performed in tube. The results were compared to those of the cytological and histological studies. The molecular hybridization technique in tube (Hybrid Capture I) detects two groups of HPV types. One group is highly associated with the development of cancer (types 16, 18, 31, 33, 35, 45, 51, 52, 56) whereas the second group (types 6, 11, 42, 43, 44) is not. Among 42 patients with cervical lesions before any treatment, high risk DNA of HPV was found in 50% of those with low grade cytology and 90% with high grade cytology. In total, 32 out of the 42 patients (76%) who presented histological lesions, were actually infected by HPV. Samples were obtained before and after treatment from 9 patients. Seven out of 9 presented high grade cervical intraepithelial neoplasia (CIN) and 2 other patients had low grade CIN. HPV DNA was not detected in any of the patients after treatment. Detection of HPV DNA by molecular hybridization in tube is simple, sensitive, standardized, inexpensive and is well adapted to screening programs. It can be used in complement of the cytological diagnosis, in the surveillance of equivocal cytological abnormalities, and in the follow-up of treated patients.

Adenocarcinoma↗

[Lymph node involvement of squamous cell carcinoma of the oesophagus. Contribution of thoracic radiograph (author's transl)].

The authors studied standard thoracic radiographs (PA and lateral) and esophageal X ray examination with barium in 116 patients with a squamous cell carcinoma of the esophagus. Diagnosis of lymph node involvement was done in 21 cases. Is was only probable in 33 other cases. In 43 cases, lymph node are located near the primary tumor but in 11 cases, they are located at a certain distance from the primary lesion. These X ray, simple findings, seem to be very important for the radiotherapeutic treatment planning.

Carcinoma, Squamous Cell↗

[Myofibromatosis in children. Histopathologic and ultrastructural study of a localized form with a spontaneously regressive course].

Report of a case of the localized form of infantile myofibromatosis, in a three-month-old boy with a brachial mass and osteolytic lesion of the adjacent humerus at radiographic examination. Diagnosis is made on the histopathological features of the lesion at biopsy. Ultrastructurally, the tumor cells have myofibroblastic differentiations. Evolution is regressive without any treatment. The anatomical, clinical, pathological, ultrastructural and evolutive features are discussed.

Gene alterations and apoptosis in rhabdomyosarcoma.

The aim of the study is to look retrospectively for gene alterations and evaluate apoptosis in rhabdomyosarcomas (RMSs) from 40 children including 24 patients not previously treated. Histological subtype was botryoid in 1 case, spindle cell in 2 cases, embryonal in 22 cases, alveolar in 10 cases, and undetermined in 5 cases. Gene expression was evaluated immunohistochemically for p53 tumor suppressor gene, MDM2 oncogene, and bcl-2 gene. N-myc amplification was detected by in situ hybridization. Apoptotic cells and bodies were recognized morphologically and stained by 3-OH end labeling. Intranuclear accumulation of p53 protein was obvious (> 25% of tumor cells) in two recurrent embryonal RMSs. Expression of the MDM2 gene was intense (80% of tumor cells) in a recurrent and metastatic embryonal RMS. Amplification of the N-myc gene was obvious (about 20% of tumor cells) in an alveolar RMS metastatic at diagnosis. Expression of the bcl-2 gene was intermediate (25-75% of tumor cells) in 26% of cases and high (> 75% of tumor cells) in 10% of cases either embryonal or alveolar. The percentage of tumor cells showing morphologically recognizable apoptosis was 0.2-7.5% (mean 2.9%). There was no correlation between apoptosis and histological subtype, bcl-2 expression, or previous treatment.
