ADP-ribosylation factor 6 and endocytosis at the apical surface of Madin-Darby canine kidney cells.

We report that the small GTPase, ADP-ribosylation factor 6 (ARF6), is present only on the apical surface of polarized MDCK epithelial cells. Overexpression of a mutant of ARF6, ARF6-Q67L, which is predicted to be in the GTP-bound form, stimulates endocytosis exclusively at this surface. Surprisingly, overexpression of the mutant ARF6-T27N, which is predicted to be in the GDP-bound form, also stimulated apical endocytosis, though to a lesser extent. ARF6-stimulated endocytosis is inhibited by a dominant-negative form of dynamin, or a dominant-negative hub fragment of clathrin heavy chain, indicating that it is mediated by clathrin. Correspondingly, overexpression of either mutant of ARF6 leads to an increase in the number of clathrin-coated pits at the apical plasma membrane. When ARF6-Q67L is overexpressed in the presence of the dominant-negative dynamin, the ARF6-Q67L colocalizes with clathrin and with IgA bound to its receptor. We conclude that ARF6 is an important modulator of clathrin-mediated endocytosis at the apical surface of epithelial cells.

Identification and characterization of two novel lymphocyte function-associated antigens, L24 and L25.

We describe the function and cell distribution of two novel cell surface antigens, L24 and L25. These antigens are broadly distributed on human lymphocytes. Monoclonal antibodies specific for these molecules block lysis by Class I- and II-specific cytotoxic T lymphocytes, but do not affect any other T cell functions tested. Anti-L24 antibody immunoprecipitates a molecule composed of two disulfide-linked monomers of 140 kd each. Anti-L25 antibody immunoprecipitates three proteins of 150, 85, and 75 kd. The study of these and other function associated molecules may provide insight into mechanisms of cytotoxic T lymphocyte recognition and/or function.

Monoclonal antimacrophage antibodies: human pulmonary macrophages express HLA-DR (Ia-like) antigens in culture.

Study of human macrophages and their role in disease has been hampered by lack of well-characterized tissue macrophages. We describe a simple procedure to obtain viable adherent cells in good yield from resected lung, characterize the cells as greater than 95% macrophage by standard criteria, and document expression of marker properties previously described in rodent macrophages. Human pulmonary macrophages were used to produce monoclonal antibodies and to study expression of HLA-A,B,C, HLA-DR, and related antigens. A new monoclonal mouse antibody, 18, showed restricted binding to macrophages from human lung or blood and to B lymphoid cells, but did not bind to closely related cells. With antibody 18 and other monoclonal antibodies of known specificity, DA2 and Genox 3.53, we showed that a remarkably high proportion of human pulmonary macrophages express HLA-DR framework and DC1 polymorphic determinants in culture. Although the macrophages differed in source and in their state of activation, single cell analysis revealed no heterogeneity in antigen expression.

Nonthrombocytopenic purpura induced by carbromal.

A case of carbromal-induced purpura is described and the typical clinical features are presented. The diagnosis was confirmed by patch tests with 1 percent and 5 percent carbromal in propylene glycol and the positive results of a macrophage migration inhibitory factor test for carbromal. The rash gradually subsided following withdrawal of the drug.
