Cellular blood fluorescence polarization: a possible prognostic tool in human acute lymphatic leukemia.

We measured the fluorescence polarization of 1-6 diphenyl hexatriene in mononucleated cells from the peripheral blood of 49 acute lymphoid leukemic patients in complete hematological remission with an Elscint MV-1 microviscosimeter. The measurements of fluorescence polarization were made at various times after the remission was achieved. Our results represent two years of this prospective study. We found that lymphoid leukemic patients in remission could be separated into two groups according to the lowest value of fluorescence polarization of their blood cells. 1) The first group of 26 patients showed values superior to 3.44 poises, in all samples, corresponding to the lowest value of normal controls. 2) The second group showed at least one value of blood fluorescence polarization lower than 3.44 poises. This second group had a significantly lower median duration of remission and survival compared to the first. We have subdivided these two groups according to other established prognostic parameters such as cytological subtype and initial volume of the disease: we found that cellular fluorescence polarization could give additional prognostic information. These data suggest that fluorescence polarization measurements have a prognostic value to follow acute lymphoid leukemia patients in complete remission: when a low value is found there is a significant risk of relapse.

Long term perifusion of rabbit ovarian follicles.

Preovulatory follicles were dissected from ovaries of estrous rabbits. Four follicles were placed in each chamber of a perifusion system. Medium 199, containing 10% rabbit serum and antibiotics, eluted through the chambers at a rate of 1 ml/10 min. Every 6 h for 36 h, 1 microgram of LH and 50 ng of FSH were injected into each chamber. P4, T and E2 were measured in the medium. P4 exhibited a peak after each of the injections at 6, 12, 18, 24 and 30 h. E2 and T gave major peaks after the injection at 0 time, with minor peaks after the injection at 6 h; thereafter T and E2 secretion gradually decreased to levels below those of baseline.

[Adenomyoma of the gallbladder].

The clinical, anatomopathological and radiological data concerning adenomyoma of the gall bladder are considered. The differential features of the pain in patients suffering from adenomyoma and calculosis are reviewed and a line of therapeutic conduct proposed in the light of modern pathogenetic views.

[A case of follicular carcinoma of the thyroid gland, with uptaking metastases, treated with surgery and isotope therapy].

A short review of the relevant literature is followed by the presentation of a case of follicular cancer of the thyroid with uptaking spinal metastases. Five months after almost total thyroidectomy for right macrofollicular goitre and left follicular cancer, a 131-I scintiscan showed uptake in the right lobe of the thyroid and IX and X dorsal vertabrae. Resection of the remainder of the thyroid was followed by the administration of 100 and then 150 mC of 131-I. The result at 10 months was satisfactory, though not final; no uptake at the Xth vertebra, reduced uptake at the IXth, disappearance of subjective and objective symptoms of spinal pain.

Characterization of normal breast epithelial cells in primary cultures: differentiation and growth factor receptors studies.

The growth and differentiation of normal human mammary epithelial cells (HMEC) were studied after propagation of serial cultures from breast tissue biopsies from 42 mammoplasty patients. Cells were grown for up to 7 mo. in low calcium medium. HMEC cultures displayed heterogeneous growth patterns, according to the average doubling time of 44 +/- 6 h for 32 generations. Proliferation peaked at Day 30. HMEC maintained a normal karyotype and were organized in ductlike structures when cultured in collagen gel matrix. The cultures retained several phenotype traits of the epithelial lineage, including the expression of cytokeratins 18 and 19, specific mammary gland antigens, as shown by indirect HMEC immunostaining by the monoclonal antibodies DF3, EMA, 7B10, and 1BE12. Estrogen receptors were undetectable, whereas progesterone receptors were present at very low density. High-affinity cell surface receptors for epidermal growth factor (EGF) (Kd = 1.1 x 10(-10) M) were observed at a density of 50,000 to 100,000 sites per cell. Accordingly, [3H]thymidine incorporation in HMEC was optimally stimulated by EGF at concentrations of 10(-11) to 10(-10) M. HMEC were also seen to possess functional VIP receptors linked to the adenylate cyclase system, as we previously observed in seven human breast cancer cell lines. These results show that long-term cultures of HMEC provide useful models for studying the growth and differentiation of the normal human mammary gland, and the role of growth factors and hormones in these functions.

Modulation of proliferation, estradiol receptors and estrogen regulated protein PS2/BCEI in human breast cancer cell lines by gamma interferon.

The effects of recombinant gamma interferon (IFN gamma) on proliferation, estrogen-receptor (ER) content, mRNA level and protein secretion of a breast cancer estrogen-induced protein pS2/BCEI were investigated in two human breast cancer cell lines, ZR75-1 and T47D. Both cell lines have estrogen and progesterone receptors and previously showed HLA class I and class II responses to IFN gamma (Int J Cancer 1990; 45: 1169). An antiproliferative effect of IFN gamma (1000-5000 U/ml) was observed in serum containing medium on ZR75-1 but not on T47D cells. Noninhibitory concentration of IFN gamma (100 U/ml) had sensitising antiproliferative effect with antiestrogens on ZR75-1 cells whereas IFN gamma did not modify the growth inhibition observed in T47D cells with antiestrogens. In serum-free, estradiol-free, phenol-red-free chemically defined medium (Cancer Res 1984; 44: 4553), IFN gamma abolished in ZR75-1 but not in T47D the 30% growth stimulation induced by estradiol. In ZR75-1 cells, IFN gamma induced a transitory 30-50% increase of ER content, as measured by ER-enzymoimmunoassay, at day 2 of culture, and reduced mRNA level and secretion of pS2/BCEI. In T47D cells, a 30-50% decrease of ER content was observed but only when cells were long term cultured (30 weeks) with IFN gamma. In this cell line, no transcription of pS2/BCEI was observed. Antiproliferative action of IFN gamma on ZR75-1 cells is associated with an inhibition of estradiol effects and a reduction of pS2/BCEI mRNA level and protein secretion.

[Villous adenoma of the rectum. Review of the literature and personal experience].

Potential malignancy of rectal villous adenomas is referred as varying from 20 to 30%; an appropriate surgical excision is required in order to insure an effective cancer prophylaxis and to improve the accuracy of the pathologic assessment. The Authors review their series concerning 15 patients with one or more polyps of villous type, located at the lower third of the rectum and with precise indications for transanal excision. In 13 cases histology demonstrated the presence of an adenoma with different grade of dysplasia, which simply entered a follow-up together with a case of an adenoma with carcinoma in situ. The latter, because of the presence of invasive carcinoma, underwent radical surgery. No postoperative morbidity was observed. Recurrence rate was 8%. After an accurate review of the Literature, a radical management of the rectal villous tumour performed on routine basis seems unjustified. The Authors suggest the indication for a modern proceeding.
