Expression of Ba currents in Xenopus oocyte injected with pregnant rat myometrium mRNA.

Using the single electrode voltage-clamp technique, stage V and VI Xenopus oocytes showed a Ba inward current (endogenous IBa) with a peak amplitude of -15 +/- 2 nA in a Cl and Na-free Ba methane sulphonate medium. When oocytes were injected with pregnant rat (18 days gestation) myometrium mRNA, an additional component of Ba current could be detected (exogenous IBa). This inward current could be distinguished from the native IBa by several means: i) peak amplitude (-75 +/- 3 nA); ii) activation voltage threshold; iii) steady-state inactivation parameter; iv) sensitivity to dihydropyridines. The features of the exogenous IBa were compared to those of the inward Ca channel current recorded with the tight-seal patch-clamp technique in single myometrial cells maintained in primary culture.

Possible effects of membrane polarization on calcium uptake by and release from sarcoplasmic reticulum vesicles.

Rates of calcium uptake by and calcium release from sarcoplasmic reticulum vesicles isolated from skeletal muscle of the crab seem to depend on membrane potential generated by potassium (K) and chloride (Cl) gradients. This does not appear to be due to an effect of the ions themselves since media of different ionic compositions leading to the same membrane potential, also lead to the same ATP hydrolysis and the same Ca uptake by SR vesicles. From a large positive intravesicular potential (conditions termed "normal" in this paper), membrane depolarization of passively Ca loaded vesicles, produced by changes in K and Cl concentrations in the media, resulted in: i) decrease in rate of calcium uptake; ii) decrease in calcium loading; iii) increase in rate of calcium release despite a decrease in the driving force for calcium ions. Moreover, the addition of caffeine (5 mmol/l) to the different polarization media resulted in a increase in calcium release.

[Rupture of the abdominal wall in highway accident victims wearing seat-belts].

Rupture of the rectus abdominis muscle represents one of the rare specific lesions due to seat-belts. Two cases are presented which illustrate diagnostic difficulties and the value of the warning provided by these lesions, which are very frequently associated with severe contusion of the small intestine or colon.

Suppression of mutation induction and failure to detect mutagenic activity with athabasca tar sand fractions.

5 different histidine-requiring strains of Salmonella typhimurium were used to test the mutagenic activity of 7 different fractions of Athabasca tar-sand. None of the 7 fractions (bitumen, maltenes, asphaltenes, saturated, monoaromatic, diaromatic and polyaromatic hydrocarbons), showed positive mutagenic response in any of the Salmonella typhimurium strains. We have tested a wide range of concentrations. The results obtained so far are consistent with the lack of mutagenic activity of all investigated fractions in the absence and in the presence of metabolic activation. Assuming that there might be an association between the absence of mutagenic activity and the complexity of the tar-sand fractions, we investigated the effect of the polyaromatic hydrocaron fraction on the mutagenicity of the carcinogenic agent 2-aminoanthracene. The data obtained indicate clearly that the polyaromatic hydrocarbon fraction suppresses the mutagenic activity of 2-aminoanthracene.
