Measurement of total-body 40potassium in patients with urate calculi.

Fifty-eight patients with urate calculi and 31 patients with other (non-urate) calculi were examined for total-body potassium by means of a total-body counter. The patients with urate calculosis were found to differ significantly in their total-body K from patients with other calculi as well as from normal subjects. The results indicate that total-body potassium deficiency is one of the factors causing urate calculosis.

[Marginal nephrectomy for the removal of coral calculi].

Efforts aimed at surgical solutions conserving the organs obtained importance also in renal surgery in recent years. This fact meant a milestone in the life of patients suffering from coral calculi, since it has been recommended to perform nephrectomy only in rare cases, and in the majority of cases young mothers were concerned. On the basis of the operation material comprising 12 cases--out of which some cases are described--the points of view of the marginal nephrectomy are discussed in detail. Among the modified techniques on the one hand the hairpin-shaped suture, on the other in the case of the local disease of the calyx system the marginal nephrectomy combined with wedge-shaped resection are explained, for the reduction of the danger of a recidivation of calculi an essential importance is ascribed to the last mentioned method.

Chemical and biochemical studies for the differentiation of coagulase-positive staphylococci.

The cell wall composition, the configuration of lactic acid produced from glucose under anaerobic conditions, the occurrence of fructose-1,6-diphosphate (FDP) activated L-lactate dehydrogenase (L-LDH), and the esterase pattern were determined from more than 80 strains of coagulase-positive staphylococci isolated from man and animal. Strains isolated from man, swine, bovines and hares form a rather homogeneous group. They exhibit a similar cell wall composition, produce predominantly D,L-lactate and have a characteristic and simple esterase pattern. Coagulase-positive staphylococci isolated from dogs, horses, minks and pigeons are quite distinct from typical Staphylococcus aureus strains. They exhibit a different cell wall composition, produce only L-lactate, possess an L-LDH which is specifically activated by FDP, and have a quite complex esterase pattern.

Hypertension in adult rats treated neonatally with angiotensin.

Rats treated neonatally with angiotensin showed labile hypertension in adulthood. This finding may suggest a permanently changed responsiveness of a central nervous controller of the cardiocirculatory system caused by angiotensin-induced hypertension during brain differentiation.

Sex hormone-dependent brain maturation and sexual behaviour in rats.

In male and female rats the endogenous steroid and gonadotrophin secretion was inhibited by injecting high doses of chlormadinone acetate (CmAc) from day 14 to 24 of life, i. e. during the period of brain maturation. In adulthood the males treated prepubertally with CmAc exhibited reduced sexual activity and fertility, whereas the females did not differ from the controls. More complete sex hormone deficiency during brain maturation was achieved by castration on day 14 of life. Controls were castrated at normal puberty time (40--60 days). Both groups were then substituted with androgens or oestrogens. In the females castrated on day 14 no impairment of sexual behaviour was observed as compared to the later castrated controls. In contrast, the early castrated males showed delayed onset of mounting behaviour. At autopsy, the weights of their sex organs were found to be lower than in the controls despite equal testosterone replacement for several months. These findings speak in favour of a permanently diminished responsiveness to androgens in males having been exposed to more or less severe androgen deficiency during sex specific brain maturation. Hence, the maturation of a male hypothalamus as well as the differentiation appears to depend at least in part on the presence of androgens, whereas in females it runs without hormonal influence.

Purification and properties of a fructose-1,6-diphosphate activated L-lactate dehydrogenase from Staphylococcus epidermidis.

L-(+)-lactate dehydrogenase (LDH) from Staphylococcus epidermidis ATCC 14990 was purified by affinity chromatography. The purified enzyme was specifically activated by fructose-1,6-diphosphate (FDP). The concentration of FDP required for 50% maximal activity was about 0.15 mM. The enzyme activity was inhibited by adenosine diphosphate (ADP) and oxamate. The inhibition by ADP appeared to be competitive with respect to reduced nicotinamide adenine dinucleotide (NADH). The catalytic activity of the LDH for pyruvate reduction exhibited an optimum at pH 5.6. The enzyme is composed of four, probably identical, subunits. Sephadex gel filtration and sedimentation velocity at pH 5.6 Yielded molecular weights of about 130 000 and 126 000, respectively. The molecular weight at pH 6.5 and 7.0 was found to be only about 68 000. Polyacrylamide gel electrophoresis in the presence of sodium dodecylsulfate and sedimentation velocity at pH 2.0 or 8.5 revealed monomeric subunits with an approximate molecular weight of 36000. The thermostability of the heat labile enzyme was increased in the presence of FDP, NADH and pyruvate. The purified LDH exhibited an anomalous type of kinetic behavior. Plots of initial velocity vs. different concentrations of pyruvate, NADH or FDP led to saturation curves with intermediary plateau regions. As a consequence of these plateau regions the Hill coefficient alternated between lower and higher n-values. Some distinguishing properties of the S. epidermidis LDH and other LDHs activated by FDP are discussed.
