A histochemical study of some enzymes of glucose metabolism was performed on the heart conduction system of rat, dog, rabbit, pig, calf and lamb. Histochemical activities revealed a higher rate of anaerobic metabolism and a lower rate of aerobic metabolism in the conducting cells in comparison with the working myocardial fibres. An increase of the histochemical activities from the atrioventricular node to the distal portions of bundle branches was noted. The importance of the high glycogen content and the high phosphorylase activity in the heart conduction system was discussed.
DNA synthesis of human hepatoblastoma HepG2 cells is reversibly inhibited by butyrate. When butyrate is removed from the culture medium, cells re-enter the cell cycle, synthesizing DNA with a time lag of about 12 h. HepG2 cells, growth-inhibited for 30 h with butyrate, synthesize and accumulate a nuclear protein, called D. Protein D synthesis is inhibited in cells which, released from the butyrate block, have resumed DNA synthesis as well as in growing cells never exposed to butyrate. Protein D has been purified from growth-arrested cells and partially sequenced. The amino acid sequences of five internal trypsin peptides indicate that protein D is a novel nuclear protein.

The present research deals with the localization of the interdigitating cells (IDCs) in normal and in Down's human thymuses. Eight thymic biopsy samples of normal children from 16 months to 10 1/2 years and six samples of Down's children from 2 months to 6 1/2 years were stained by the indirect immuno-peroxidase method using an anti-S-100 protein serum. IDCs are localized in the medullary zones, always numerous in all the Down's thymuses and in an age-related decreasing number in normal thymuses. The interrelationships between the physiological and pathological role of IDCs in inducing self-tolerance and T-cell activation and the numerical distribution in normal and in Down's human thymuses are discussed.
Chronic gastritis, which is frequent in subjects over 50 years old, is caused by the concurrence of predisposing and congenital conditions and exogenous harmful factors, in particular foods. In etiopathogenetic terms it is worth considering autoimmune diseases and duodenogastric back-flow separately. Lesions develop progressively from superficial gastritis to atrophic gastritis and finally to gastric atrophy; they are frequently found together with intestinal metaplasia, formed by areas of the epithelium with the morphological and histochemical characteristics of intestinal mucosa, which are the expression of a modified regeneration of the gastric wall. It is acknowledged that chronic atrophic gastritis is a precancerous phenomenon which is the majority of cases leads to the onset of intestinal cancer, passing through the stages of chronic gastritis, metaplasia and dysplasia. Identification of this lesion may therefore help to prevent cancer: diagnosis is essentially performed using endoscopy (together with histocytological tests and bioptic staining) and laboratory tests (enzyme and CEA assays in the gastric juices). Rather than prescribing generic medical therapy or surgical treatment, which is only possible in selected cases of alkaline gastritis, attention is focused on curing unhealthy habits and on an endoscopic follow-up (every 2 years in cases of gastritis, and more frequently in cases of metaplasia or dysplasia).
The addition of sodium butyrate, a differentiation-inducing agent, to the culture medium of human hepatoblastoma-derived (Hep G2) cells, produced a dose-dependent and time-dependent increase in albumin (ALB) and decrease in T4-binding globulin (TBG) synthesis and secretion. In the presence of 0.01 to 2.0 mM sodium butyrate, newly synthesized [35S]ALB progressively increased up to 139% of control cultures grown in the absence of sodium butyrate, whereas TBG synthesis was already slightly inhibited using the lowest concentrations of this agent and further diminished thereafter. The use of 5 mM and 10 mM sodium butyrate inhibited the synthesis of both proteins, probably as a consequence of toxic effects on cell cultures. The addition of 1 mM sodium butyrate for variable time intervals caused an increase in the amount of ALB recovered in the medium up to 146% after 72 h, and a decrease of TBG up to 44% of controls. These different effects on ALB and TBG occurred concomitantly with an inhibition of cell growth, as shown by the reduction in the cell number/flask compared to control cultures. At the highest sodium butyrate concentrations, a relevant impairment in the secretion of newly synthesized TBG, but not of ALB, also occurred. These divergent effects on ALB and TBG synthesis by Hep G2 cells might be related to biochemical differentiation induced by sodium butyrate in this tumoral cell system, suggesting that TBG synthesis is increased in Hep G2 cells because of their neoplastic nature.
A case of an aneurysm of the left sinus of Valsalva ruptured in the right atrium is reported. This anomaly was diagnosed in a 30 year old woman who had a continuous systo-diastolic murmur and no signs of cardiac failure. The diagnosis of aneurysm of the left sinus of Valsalva was been determined using 2-D echocardiography, Cardioangiography, in addition, enabled us to recognize the fistula which connected the left sinus of Valsalva with the right atrium. The rarity of this case is supported by two pieces of data: A) aneurysms of the left sinus of Valsalva are the rarest of the sinus of Valsalva aneurysms; B) generally the sinus of Valsalva aneurysms rupture into the adjacent cardiac cavities, while the anomalous fistolous conduits are quite uncommon. The case reported confirms that from an aneurysm of the sinus of Valsalva an atypical fistolous conduit may arise. This could be detected by cardioangiography. In addition echocardiography, is very important, at least as a preliminary examination, for the diagnosis of the sinus of Valsalva aneurysms.
24 patients (19 females and 5 males) between 45 and 81 years of age (average age 61.6) with chronic left bundle branch block and with no signs of reduced left ventricular function have been studied by M-Mode echocardiography. These patients have been split into three groups: A) pts. with normal indexes of left ventricular function and normal thickness of the left ventricular walls; B) pts. with normal indexes of left ventricular function and thickness of the interventricular septum at upper limits or increased; C) pts. with different cardiac pathologies but with no signs of reduced ventricular function. The contractility indexes and the motion of the interventricular septum in both the basal and the medial portion have been studied. The results did not show any important variation concerning the ventricular function compared to the normal values. The study of the septal motion pointed out, as a main feature of the left bundle branch block, a preejective posterior motion followed by: a) a normal posterior systolic motion of the interventricular septum, or b) a posterior systolic motion followed by a further posterior protodiastolic motion (delayed motion), or c) an anterior systolic and posterior diastolic motion (paradoxical motion). The alterations of the septal movement ranked in different degrees. The clearest alterations were peculiarly found in the patients of the first group. The patiens of the third group, besides, have shown the lightest alterations. The results allowed to deduce that patients with left bundle branch block can have both normal motion or alterations of the interventricular septum motion of different degree.(ABSTRACT TRUNCATED AT 250 WORDS)

Parathyroid surgery needs an appropriate diagnosis and a preoperative localization. We conducted a prospective study to compare the efficacy of 4 different imaging modalities in 17 patients: thallium-technetium subtraction scintigraphy, ultrasonography, computed tomography and arteriography. The sensitivity was: scintigraphy 58%, echotomography 86%, Tc 92% and arteriography only 33%. Neck exploration confirmed the imaging results. We found 15 cases of adenomas (2 cases of double adenoma) and 1 case of hyperplasia; in 1 patient, no lesions were found. We conclude that the association of such techniques appears to be the optimal strategy in about 100% of the patients.
An anti-thymostimulin (TS) serum was tested on rabbit, guinea-pig, lamb and pig thymuses to study the localization of the hormonal factor. The immuno-peroxidase method, with some modifications, was applied to tissue fixed in Bouin's or in Bouin-PAF liquids and embedded in paraffin wax. Immuno-reactivity was shown in reticulo-epithelial cells in the cortex, in the medulla and in the external cells of the Hassall's corpuscles. The intensity of the immuno-reaction is stronger in pig thymus, whereas in lamb, rabbit and guinea-pig thymuses it is more uniform and pale, but to varying degrees. Comparison with the localization of TS in thymuses of other mammals and man and the problem of species-specificity were discussed.

H1 histones were purified by preparative sodium dodecyl sulfate polyacrylamide gel electrophoresis from human lung carcinoma (line DMS79), human hepatoblastoma (HepG2), human adult lung and human adult and fetal liver. The purified human H1 histones were analyzed for their amino acid composition and terminal residues. The comparative analysis of the amino acid compositions of the different human H1 histones showed that: all the H1 preparations have the characteristically high lysine content associated with a low arginine content, which distinguishes outer histones from core histones; H1 is distinguishable from other H1 histones by the presence of methionine and histidine; H1 histones from human adult, fetal and cancer cells are very similar in amino acid composition, and in cancer cells the level of the H1 histone is not inversely related with cell growth rate nor with the expression of the alpha-fetoprotein gene.
Macromolecules and supramolecular structural components that are incorrectly synthesized or are damaged by radiation or by reactive chemicals are either repaired or selectively degraded and resynthesized. In addition, turnover rates for macromolecules and supramolecular structures can be elevated by alternation of fasting and feeding periods and can be influenced by metabolic regulatory mechanisms which are governed by steady-state concentrations of labile macromolecules.
