Comparative pharmacokinetics of febantel and its metabolites in sheep and cattle.

The pharmacokinetics of febantel and its main metabolites were studied in cattle and sheep. Seven ewes and 4 heifers were given febantel orally in a single dose of 7.5 mg/kg, 25 mg/kg, or 45 mg/kg of body weight. Plasma concentrations vs time of febantel and individual metabolites were determined by high-performance liquid chromatography analysis. Intestinal absorption of febantel was faster and biotransformations were more active in sheep than in cattle.

Radioimmunoassay of 19 nor testosterone. Evidence of its secretion by the testis of the stallion.

Antiserum has been raised in rabbits treated with a 19 nor testosterone-hemisuccinate-bovine-serum-albumin conjugate and used for the development of a specific RIA of plasma 19 nor testosterone. Plasma samples are drawn from testicular and jugular veins of stallions during castration under general anesthesia. Results demonstrate a testicular secretion of 19 nor testosterone and a stress inhibition of this secretion correlatively with stress inhibition of testosterone secretion.

A correlation of toxicity of albendazole and oxfendazole with their free metabolites and bound residues.

The possible correlations between embryotoxicity, plasma kinetics of toxic metabolites and covalent binding of metabolites to foetal tissues were studied using two drugs, albendazole and oxfendazole. In the rat, the metabolic inhibitor, SKF-525A, induced changes in embryotoxicity which were well correlated with plasma levels of identified embryotoxic metabolites, but not with the levels of foetal tissue bound drug metabolites.

[Serum enzymology in the goat: the usual levels of CPK, LDH, ICDH and SDH].

Using about 100 blood samples of dairy goats selected for their high level of milk production, the authors determined the enzymic activities of CPK, LDH, ICDH and SDH. The limits of the practical values of these parameters are (expressed in IU/l): CPK: 14-62; LDH pregnancy: 217-417, lactation: 300-586; ICDH: 2.3-14.8; SDH: 2.6-11.6.

Kinetics of four metabolites of Febantel in cow's milk.

The mammary elimination of four potentially toxic metabolites of Febantel has been investigated by HPLC methods in the cow after the administration of a 7,5 mg/kg single oral experimental dose of this anthelmintic. The total of the four metabolites is equal to about 0.4 ppm 12 hours after the treatment. The individual levels fall down under the detection limit between the third and the sixth milking.

[Non-extractible residues of aryl-14C-albendazole in sheep].

The location of the intractable residues of 14C-ring labelled-albendazole has been investigated in the sheep. Numerous biochemical compounds have been isolated: pulmonary CO2, blood dextrose, glycogen, triglycerides, cholesterol, hemoglobin, plasmatic, cytosolic and microsomal proteins, nucleic acids . . . No incorporation into intermediary metabolisms has been put into evidence. On the contrary, a significant covalently bound radioactivity has been observed on hepatic and plasmatic proteins. No covalent binding to hepatic DNA seems to be produced.

[Experimental infection of the pregnant cow with Chlamydia psittaci].

Seven cows were inoculated with two strains of C. psittaci from bovine origin. Their chlamydial excretion, complement fixing (CF) antibody titer and hormonal pattern, at calving or abortion were monitored. Five of the six pregnant inoculated cows had a pathological parturition, and a CF antibody titer greater than 40 during more than two months post-abortion. Three of them excreted Chlamydiae at calving. The other inoculated pregnant cow, the inoculated non pregnant cow and the three contacts had CF titer less than 40 during the same period. The progesteronemy and the oestradiol 17 beta level of the two early aborted cows were abnormal.

[Metabolism-embryotoxicity relationship of febantel in the rat and the sheep].

The metabolism of febantel has been investigated in the rat and the sheep. The general pathway seems to be similar in both species: ten urinary metabolites are the result of cyclisation, oxidation and/or hydrolysis process. Using synthetic samples, the embryotoxic study of all individual metabolites has been performed in the rat. Unchanged febantel and two other metabolites are responsible for teratogenic effects. A four steps toxogenic pathway is described which finally leads to the ultimate known teratogen: methyl (5-(phenylsulfinyl)-1H-benzimidazole-2-yl) carbamate. Metabolic similarities with other anthelmintics and residual consequences are discussed.
