Postnatal development of semitendinosus muscle in the dog.

In this study the differentiation of postnatal muscle fibres in dog semitendinosus muscle has been characterized. Several histochemical techniques for myosin ATPase and metabolic activity were used in animals aged between 1 day and 2 months. The results show that at birth there are two types of fibre, whose ATPase activity gradually changes during postnatal development, so that several types of fibre can be identified after 2 months. These finally become the four types of the adult: I, IIA, IIDog, IIC. The main conclusions of the study are that the use of mATPase techniques is appropriate for showing the differentiation between muscle fibres, even at early stages of postnatal development, and that the origin of the four different fibres of the adult can be traced back to early postnatal stages.

Paraoxonase activity in human pericardial fluid: its relationship to coronary artery disease.

Pericardial fluid paraoxonase activity was compared with 3 biochemical markers of atherosclerosis (HDL, LDL/HDL ratio and Apolipoprotein A-I) and a significant association was found. When the paraoxonase activity in pericardial fluid samples was separated into 2 groups according to the degree of coronary atherosclerosis (slight and severe), most of the cases showing low levels of paraoxonase activity also showed severe coronary atherosclerosis. In addition, paraoxonase activity in pericardial fluid was found to be statistically correlated with HDL levels, which agrees with the results reported in serum.

Partial purification of paraoxonase from rat liver.

A method for the partial purification of rat liver paraoxonase is presented. The method consists of the following steps: preparation of microsomes, solubilization with Triton X-100, adsorption on hydroxylapatite and chromatography on DEAE-52 cellulose. A partially purified preparation of rat liver paraoxonase has been obtained, showing a specific activity of 422 mU/mg with a yield of about 22% and a purification factor of 77-fold.

Rat liver paraoxonase: subcellular distribution and characterization.

The subcellular localization and some biochemical properties of rat liver paraoxonase have been studied in order to establish a correlation with plasma enzyme. The whole paraoxonase activity was found in the microsomal fraction. Rat plasma and liver paraoxonase showed similar optimum pH (8.5), Km (0.4 mM) and calcium requirement, but differed in the response to several inhibitors.

Characterization of paraoxonase activity in pericardial fluid: usefulness as a marker of coronary disease.

In this study, the presence of paraoxonase activity in pericardial fluid was demonstrated. A comparison of some properties, such as optimum pH, stability versus pH, heat inactivation, effect of inhibitors, isoelectric point and kinetic parameters (Km and Vmax), between plasma and pericardial fluid paraoxonase was made. The properties studied were practically identical. The enzyme activity in pericardial fluid was tested as a marker in the postmortem diagnosis of myocardial infarction. The paraoxonase activity in the myocardial infarction group (47 cases) was lower than in the control group (40 cases), but the difference was not significant.

[Chronology of the postnatal ossification of the shoulder and elbow joints in the Siamese cat (Felis catus L.)].

A study was done by radiological techniques to show the chronology of ossification of the shoulder and cubit joints in the Siam cat, from birth up to the 25th week of postnatal development. For this experiment we used 40 little cats (19 males and 21 females) belonging to 11 litters subjected to different controls: pattern race, healthy, feeding, growing up and radioactivity. The time of the appearance and the evolution of the ossification centers is determined as well as the phenomenon of fusion during this time. The postnatal ossification from both joints of the Siam cat is compared with that of the common cat.

[Chronology of the postnatal ossification of the thoracic limb in the Siamese cat (Felis catus L.)].

A study was done by radiological techniques to show the chronology of ossification of the thoracic autopodo of the Siam cat from birth up to the 25th week of postnatal development. For this experiment we used 40 little cats (19 males and 21 females) belonging to 11 litters subjected to different controls: pattern race, healthy, feeding, grow up and radioactivity. The time of the appearance and evolution of the centres that form the basipodo, metapodo and acropodo of the thoracic limb is determined. Likewise it is analyzed the phenomenon of fusion during that time to define the main ossification sequence in the carpo. The aspects of the postnatal ossification of the thoracic autopodo in the Siam cat are compared with the literature of different authors about the common cat.

Effect of glutathione on canine myocardial ischaemia without reperfusion.

The present study was to evaluate the effect of exogenous glutathione on myocardial damage resulting from permanent (no reperfusion) coronary ligation (3 or 6 h) in anaesthetized dogs. Haemodynamics, infarct size and myocardial glutathione content were determined. Erythrocyte superoxide dismutase (SOD) activity was also determined in coronary venous blood samples. Glutathione was administered by the intraperitoneal route, 100 mg kg-1 as initial dose given 5 min before coronary ligation, and successive doses of 25 mg kg-1 every 40 min throughout the study period. Saline-treated dogs showed myocardial infarction, a decrease in myocardial glutathione content, and a transient increase in SOD activity. Three hours occlusion in glutathione-treated dogs resulted in a small reduction of infarct size, and no changes in myocardial glutathione content and SOD activity. By contrast, administration of glutathione failed to reduce infarct size and failed to prevent myocardial glutathione decrease in dogs subjected to 6 h occlusion. These results indicate that exogenous glutathione is of minor beneficial effect for myocardial damage resulting from permanent coronary occlusion and suggest that endogenous glutathione has a limited role in protecting against myocardial ischaemia without reperfusion.

Intra-articular therapy of experimental arthritis with a derivative of triamcinolone acetonide incorporated in liposomes.

Triamcinolone acetonide-21-palmitate was synthesized and incorporated into liposomes for intra-articular treatment of an experimentally-induced arthritis in the knee joints of rabbits. The liposomal formulation was more efficient than free triamcinolone acetonide in solution in suppressing the arthritis. Using radioactive tracers, it was found that triamcinolone acetonide-21-palmitate incorporated into liposomes was retained in the articular cavity, together with the liposomal lipids, for a much longer period than free triamcinolone acetonide, and this correlated with its anti-inflammatory effect.

Morphological and histochemical characteristics of muscle fibre types in the flexor carpi radialis of the dog.

The canine flexor carpi radialis is subdivided into 2 portions. The radial portion which occupies one third of the cross-section is located caudomedially and is composed only of large type I fibres (43.53 +/- 0.6 microns minimum diameter) in levels close to the insertion. The cubital portion is located craniolaterally and contains fewer and smaller type I fibres (26.10 +/- 0.3 microns minimum diameter). It is hypothesised that the radial portion of the muscle, with its larger slow fibre population, contributes to forelimb postural function. The cubital portion, with its generally fast fibre population (IIA), may be more important in the muscle's recognised function (flexion of the carpal joint during locomotion).

Skeletal muscle fibre types in the dog.

Using a variety of histochemical methods we have investigated the mATPase reaction of skeletal muscle fibres in the dog. Types I, IIA, IIDog (peculiar to the dog) and IIC fibres were identified. The results reveal that the interpretation of the fibre type composition depends on the methods used.

Inhibition by trains of subthreshold high-frequency stimuli: an experimental study in spatial limitation.

The aim of this study was to determine whether the spatial limitation of the inhibitory effect of subthreshold conditioning stimuli trains (TSc) could be overcome by their simultaneous emission through several electrodes surrounding the area where the suprathreshold extrastimuli (S2) are delivered. In seven anesthaetized open chest dogs the effective ventricular refractory period was determined before and after the introduction of unipolar cathodal TSc, using epicardial electrodes. TSc pulse frequencies tested were 100 Hz, 200 Hz, 400 Hz, 600 Hz and 800 Hz, and train intensity was 10% lower than the train diastolic threshold for every pulse frequency. S2 and TSc were delivered: (a) by the same electrode; (b) by two different electrodes 3 mm apart; and (c) TSc through six peripheral electrodes surrounding the central electrode that delivered S2 (heptapolar electrode). Trains of 400 Hz showed the highest diastolic threshold, permitting the use of the highest train intensities. When TSc and S2 were delivered through the same electrode the ventricle remained unexcitable during the entire cardiac cycle in six of the seven dogs. In turn, when TSc and S2 were delivered by two different electrodes, the effective ventricular refractory period (EVRP) could only be increased by greater than or equal to 10 ms in three of the seven dogs (18 ms, 62 ms and 10 ms). When TSc was delivered simultaneously through six peripheral electrodes the increments were higher (118 ms, 88 ms and 75 ms) in these three dogs, and there was one additional dog with 12 ms increments of of EVRP.(ABSTRACT TRUNCATED AT 250 WORDS)

Protective effect of N-acetylcysteine on ischaemia-induced myocardial damage in canine heart.

The glutathione redox pathway is an important antioxidant system in the myocardium. N-Acetylcysteine is a low molecular weight glutathione precursor that has been used clinically to replenish glutathione stores. The present study was aimed at evaluating the protective effect of N-acetylcysteine on myocardial damage resulting from permanent coronary occlusion (without reperfusion) in anaesthetized dogs. N-Acetylcysteine (150 mg kg-1 i.v.) administered 2 min before occlusion reduced infarct size in dogs subjected to 24 h ischemia. The infarct size as a percentage of the area at risk was 86.8 +/- 3.6% (n = 11) in control (saline-treated) dogs and 68.2 +/- 2.4% (n = 7; P less than 0.05 vs control) in N-acetylcysteine-treated animals. Haemodynamic variables (heart rate, mean arterial pressure and rate-pressure product) were similar in the control and the treated group. Regional myocardial blood flow was determined with radioactive microspheres in ischaemic and non-ischaemic zones before occlusion and 3 h post-occlusion. N-Acetylcysteine did not influence the regional distribution of myocardial blood flow. The myocardial content of reduced glutathione was significantly (P less than 0.05) decreased 3 h post-occlusion (0.53 +/- 0.19 mumol/g-1; n = 5) compared to either pre-occlusion values (0.94 +/- 0.03 mumol/g-1; n = 8) or values 3 h post-occlusion in sham-operated animals (0.93 +/- 0.15 mumol/g-1; n = 5). Depletion of myocardial glutathione 3 h post-occlusion was not observed in dogs treated with N-acetylcysteine (0.87 +/- 0.11 mumol/g-1; n = 5).(ABSTRACT TRUNCATED AT 250 WORDS)

Identification in porcine faeces of a novel virus with a bisegmented double stranded RNA genome.

Polyacrylamide gel electrophoresis of nucleic acids extracted from porcine faecal samples revealed in several samples the presence of two discrete bands. The bands were resistant to digestion with of DNase I and RNase T1, but not with RNase A in low salt conditions, indicating that they consisted of double stranded (ds) RNA. The two bands from different samples varied in sizes, in a range between 2.4-2.6 kbp and 1.7-1.9 kbp for the slow and fast moving band respectively. The bands cosedimented in CsCl gradients at an average density of 1.415 g/ml with icosahedral virus particles of a diameter of 34 nm and a triangulation number equal to 3. Aggregates of virus, which appeared to be immunocomplexes, were seen in one sample. From 244 faecal samples collected in one farm, 27 (11.1%) were found to contain the characteristic dsRNA pattern, with a higher prevalence in samples from animals 15 to 35 days old. The agent was equally distributed among samples from diarrhoeic or non-diarrhoeic animals. These results confirm the circulation among pigs of a novel virus, possibly of vertebrates, with a bisegmented double stranded RNA genome, similar to viruses previously described in humans, wild rats, guinea pigs, pigs, and chickens, for which the name "picobirnavirus" has been proposed.

A fatal case of oral ingestion of methanol. Distribution in postmortem tissues and fluids including pericardial fluid and vitreous humor.

A 41-year-old man ingested orally a large quantity of methanol and was found dead at home. The presence of methanol in body fluids and tissues was determined by head-space gas chromatography. The blood ethanol and acetone were negative. Tissue distribution of methanol showed that the kidney presented the highest content of methanol (5.13 g/kg) followed by liver (4.18 g/kg), vitreous humor (3.96 g/l), heart (3.45 g/kg), urine (3.43 g/l), pericardial fluid (3.29 g/l), blood (2.84 g/l) and finally stomach content (2.21 g/l).

Cell adhesion regulates melanoma specific differentiation and interactions with the 3' region of the tyrosinase gene.

Decreased attachment to substratum has now been found to increase melanosome formation and cell-cell interaction in B16 melanoma. Since melanosome formation involves tyrosinase gene expression, we assayed for differential RNA expression by hybridization with probes from the distal ends of this gene, detecting unequal reactivity only with the 3' end probe. The same DNA showed binding of 2 nuclear proteins of 50 and 60 kd in unanchored cells, in contrast with a decreased binding of the 60 kd species, in nuclear extracts from attached cells. No comparable differences were detected with a gamma-actin DNA of identical length, suggesting that the changes observed are sequence-specific. Our studies suggest that the adhesion-mediated modulation of pigmentation in B16 melanoma correlates with differential macromolecular interactions with the 3' end of the tyrosinase gene.

Body composition of goat kids during sucking. Voluntary feed intake.

The body composition of thirty-eight Granadina goat kids was measured. Six animals were slaughtered at birth while the remainder were kept individually at an environmental temperature of 24 +/- 2 degrees and a relative humidity of 60 +/- 5%. They were given goat's milk or a milk-substitute at two planes of nutrition until 15 or 30 d of age and then slaughtered. The goat's milk and milk-substitute contained 260.4 and 222.0 g digestible protein/kg and 23.23 and 20.85 MJ metabolizable energy/kg respectively. Voluntary feed intake as metabolizable energy was a function of metabolic body-weight (kg W0.75), equivalent to 2.42 and 2.44 times the energy requirement for maintenance for goat's milk- and milk-substitute-fed animals respectively. There was a high degree of correlation between the empty-body concentration of dry matter, fat and energy and empty-body-weight (P less than 0.001) or animal age (P less than 0.001), and between body-weight and animal age (P less than 0.001). The relationships between empty-body composition and empty-body-weight were independent of type of milk or plane of nutrition. In contrast relationships between empty-body composition or empty-body-weight and animal age were affected by the type of milk and, over all, by the plane of nutrition. All these results show that in these animals any body-weight will have a similar composition, but it will be reached earlier or later depending on dietary regimen and always with the limitation of voluntary intake.
