Gastric antral ulcers induced by a combination of acid, indomethacin and ischemia in rats.

Gastric antral lesions were produced with hemorrhages by vascular ligation-induced ischemia in the prepyloric regions in rats. Additional treatments with intraluminal acid application and indomethacin markedly aggravated the lesions. Histological examination showed that the incidence of ulcers which penetrated the muscularis mucosae was nearly 100% upon treatment with a combination of acid, indomethacin and ischemia. This model provides a useful tool for studying gastric ulcer etiology.

[Brain metastasis of hepatoblastoma--a case report and review of literature].

Hepatoblastoma is a rare abdominal tumor which affects infant, especially two or three year old boy. The authors experienced a rare case of cerebral metastasis of hepatoblastoma. The patient was a 1 and one thirds year old boy who was admitted because of left hemiparesis, somnolent consciousness, and abdominal tumor in September 21, 1982. At admission physical examination showed massive epigastric tumor with emaciated and dehydrated condition. Neurological evaluation indicated left spastic hemiparesis, somnolent consciousness and bilateral choked disc. Laboratory data were leukocytosis, marked thrombocytosis and slight abnormality of the liver function test. Alpha-fetoprotein level was 61 X 10(4) ng/ml and human chronic gonadotropin 20 IU/l. Chest X-P was suspected to be multiple metastatic lesions. Celiac angiogram showed a giant malignant tumor of the liver. Cranial CT scan revealed an intracerebral mass in the right frontal lobe which was visualized an enhanced high density area by contrast enhancement with a cystic component. The patient was diagnosed metastatic tumors of the brain and the lung with primary hepatoblastoma. En bloc tumor removal with right frontotemporal craniotomy was performed. Histological diagnosis of the removed tumor was a well differentiated type of the hepatoblastoma. Postoperative course was fine as for consciousness and hemiparesis. With recent development of chemotherapy and medical treatment for infantile malignancy, the prognosis of hepatoblastoma has been improved. This report is believed to be the first operative case for metastatic brain tumor of hepatoblastoma.

Salivary gland tumors in atomic bomb survivors, hiroshima, japan. II. Pathologic study and supplementary epidemiologic observations.

A pathological investigation was undertaken in Hiroshima on cases seen between 1945 and 1971 to determine the relationship between salivary gland tumors and exposure to atomic (A) bomb radiation. Of the 208 cases of histologically confirmed salivary gland tumors, 62 were A-bomb survivors and 146 were nonexposed. The relative risk based on the rate in the nonexposed for malignant tumors of salivary glands among the exposed in Hiroshima Prefecture was 11.0, while that of the submaxillary gland was 13.8 and that of the parotid gland was 9.8. They were all highly significant by X2 test (P less than 0.001). The latent period of malignant salivary gland tumors was shorter in the exposed than in the nonexposed. Four cases of benign salivary gland tumor, all being benign mixed tumors of the parotid gland, were observed in children whose parents had been exposed. The incidence of mixed tumors of the salivary gland among the exposed in Hiroshima City was 2.0 times higher than that among the nonexposed and showed a tendency to increase with proximity to the hypocenter (P less than 0.01).

Effect of healing on small internal diameter arterial graft compliance.

Porosity is an essential component for long term function of small internal diameter synthetic vascular prostheses. Theoretically, porosity is required for healing by providing a scaffold for ingrowth of periprosthetic tissues. Porosity may be required for transfer of fluids and ions in prostheses even if tissue ingrowth does not occur. Increased permeability of vascular grafts has been shown to enhance tissue incorporation. Implants with pore sizes greater than 10 microns but less than 45 microns become ingrown with fibrohistiocytic tissue and capillaries. Implants with pore sizes greater than 45 microns are ingrown with organized fibrous tissue and minimal histiocytic response. The phenomenon of differential ingrowth may have important functional significance. Prostheses which heal with organized fibrous tissue have the potential for long term contracture, strangulation of vascularity and calcification. Fibrohistiocytic tissue has low mechanical strength and does not appear to contract or calcify. Elastomeric microporous vascular prostheses which are minimally ingrown or ingrown with fibrohistiocytic tissue maintain compliance after months of implantation. This study was designated to determine the effects of healing on the compliance of small internal diameter vascular replacements and to correlate the compliance to patency rates.

Human chorionic gonadotropin--like material: presence in normal human tissues.

With the use of radioreceptor assay for gonadotropin and a beta-chain radioimmunoassay for human chorionic gonadotropin (hCG), we have been able to demonstrate the presence of hCG-like material in all normal human tissues tested. This activity was completely absorbed by a hCHG-antibody affinity column and was demonstrated to have altered carbohydrate content by studies showing this material did not bind to concanavalin A (con A)-Sepharose affinity columns. In identical studies greater than 90% of placental hCG was bound to con A. However, if placental hCG is rendered carbohydrate free it also does not bind to con A. It is known that carbohydrate-free hCG is cleared rapidly from the circulation and thus possesses little or no biological potency in vivo. Accordingly, we hypothesize: (1) that this hCG-like material in normal tissues has the protein structure of hCG but does not possess the carbohydrate moieties of placental hCG and probably has little or no bioactivity in vivo and (2) that the trophoblastic cell is not unique in its ability to synthesize hCG but has developed the ability to glycosylate hCG, transforming a ubiquitous cellular protein into a hormone, hCG might better be called human cellular gonadotropin.

A mode of incipient growth in chemically induced signet ring cell carcinoma of the canine stomach.

A 31-month-old female mongrel dog was orally administered with 50 mg or 100 mg of N-nitrosobutylurea (NBU) in gelatin-capsule 3 times per week for 19 months with interposing periods of complete suspension. Thirty-four foci of signet ring cell carcinoma were found in the antral region of the stomach. The majority of the foci (31 foci) were early cancer, and the remaining foci were invasive cancer. In addition to these lesions, there was "a single gland cancer" in which a row of cancer cells was confined to a single gland. The whole gland was composed of two cell layers; the inner layer facing the lumen was normal gastric cells and the outer layer was atypical or neoplastic cells underlaid by the basement membrane. Mitosis was frequently observed on the bottom of the gland. Atypical or neoplastic cells seemed to mature gradually through a process of upward migration with increase in cytoplasmic Alcian blue-PAS and HID-AB positive mucin. Some of the cells rich in mucin moved into the lamina propria. The other cells remained in the flow of the regular cell renewal system of the normal gastric cells and reached the top of the gland. This observation revealed a mode of incipient gastric cancer growth, which starts and spreads within a single gland, before it invades the surrounding lamina propria.

[Pharmacological effects of dobutamine and several catecholamines on various smooth muscles (author's transl)].

Pharmacological effects of dobutamine (DOB), a positive inotropic drug, on smooth muscles were compared with those of catecholamines such as norepinephrine (NE), epinephrine (E), isoproterenol (ISO) and dopamine (DA). DOB acted as a partial agonist and simultaneously as an antagonist on alpha-adrenergic receptors in the thoracic artery. Large doses of DOB induced uterine contraction in rabbits, but antagonised the contraction of the vas deferens of guinea pigs, as was induced by electrical stimulation. However, DOB showed a potent activity, similar to that seen with NE or E as an alpha-adrenergic agonist, on the portal vein. Doses of DOB required to produce a relaxation in the trachea of guinea pigs both in vitro and in vivo were 1/250 and 1/600 of ISO, respectively, and were considerably less regarding inhibition of uterine motility in rats. Inhibiting activity of DOB on the transport rate of intestinal contents in mice and gastro-intestinal motility of rabbits was much less than the activity seen with ISO, E and NE. Spasmolytic activity of DOB was also less than that of ISO, NE and E, but was more than that of DA. It is concluded that DOB is a more selective beta 1-agonist than ISO because of its weaker activity on alpha- and beta 2-receptors. The alpha-agonist activity of DOB in the portal vein was however, potent.

Effect of ovariectomy on x-ray carcinogenesis in rats.

Tumor induction by fractionated whole-body X-irradiation (400 rad) was studied in spayed Sprague-Dawley rats. Ovariectomy was chosen as an intensifying factor for radiation leukemogenesis. Ovariectomized rats gained more body weight and responded more quickly (but transiently) in the recovery of WBC levels after the last (3rd or 4th) X-irradiation. A total of 26 tumors developed in 21 out of 47 ovariectomized rats, 11 tumors in 6 out of 13 ovariectomized and ovary-grafted rats, and 44 tumors in 25 out of 29 sham-ovariectomized rats during the observation period up to 64 weeks after starting X-irradiation. Eighty per cent of tumors were of mammary gland origin in the latter two groups with intact or grafted ovaries. By contrast, 61.1% of tumors in the spayed rats were derived from the subcutaneous mesenchymal tissue and the hematopoietic tissue. This may imply that some forms of mesenchymal tumors including leukemia are under the suppressive influence of female sex hormones.

Induction of atrophic gastritis in ICR mice by the administration of an allogenic antigen.

In an attempt to produce experimental autoimmunity in small animals the experiment was sought to induce atrophic gastritis in ICR/JCL mice. The stomach antigen of ICR mice was extracted and emulsified with an equal volume of Freund's complete adjuvant. This was subcutaneously injected in 5-week-old ICR/JCL mice at 1 week intervals for a total of 1 to 4 administrations. The stomach antibody in the serum gradually increased up to 2(6) until four weeks after the last injection of the stomach antigen. At the same time pyknosis and a decrease in number of the gastric mucosal cells, which ultimately led to the atrophying of gastric mucosa, developed. Thereafter, concomitant with the decrease in serum antibody against mucous cells, regeneration of mucous cells was especially remarkable, but atrophy of the fundic gland continued.

Morphological and biochemical changes in the gastric mucosa of A/HEJ mice injected with a xenogeneic stomach antigen.

Subcutaneous injection of W/Fu rat stomach antigen emulsified with an equal volume of Freund's complete adjuvant at 1 week interval for a total of 2 administrations induced atrophic gastritis in A/HeJ mice. From the second week after the last injection the parietal cell mass decreased. At the same time, Alcian blue-PAS positive cells and Paneth-like cells appeared. Furthermore, the activity of alkaline phosphatase and disaccharidase of the small intestinal mucosa was biochemically demonstrated in the atrophic glandular stomach.

Induction of rectal carcinoma in mice by local x-irradiation.

The pelvic region of random bred female ICR-JCL mice and CF1 mice was irradiated with various doses of X-rays at 1-week intervals to determine the relationship between the X-ray dose and induction of rectal carcinoma. The incidence of rectal carcinoma in ICR mice was zero after a single dose of 2,000 rad of X-rays, but 31% after a single dose of 3,000 rad, 6% after 2 doses of 1,500 rad, 25% after 3 doses of 1,500 rad, 42% after 2 doses of 2,000 rad, and 95% after 3 doses of 2,000 rad. This cancer developed in 70% of CF1 mice exposed to 2 doses of 2,000 rad. No case of this tumor was observed in the control animals not exposed to X-rays. The development of this tumor was found to depend on the X-ray exposure dose. Local X-irradiation of the pelvic region is one of the effective methods for inducing rectal carcinoma in mice. Rectal cancers induced by X-irradiation were adenocarcinoma of the tubular, papillary, and mucinous type, and frequently showed invasive growth into the deep layers of the rectal wall.

Salivary gland tumors in atomic bomb survivors, Hiroshima, Japan. I. Epidemiologic observations.

An epidemiologic investigation was undertaken in Hiroshima to study the relationship of salivary gland tumors to exposure to ionizing radiation. Two hundred and eleven cases examined between 1953-1971 were confirmed by a review of histologic specimens. Of these, 66 cases occurred in residents of Hiroshima City who were born before the A-bomb: 30 among A-bomb-exposed, and 36 in non-exposed individuals. The incidence of salivary gland tumors among the exposed individuals was significantly higher, 2.6 times greater than that among the nonexposed individuals for all tumors, and 10 times greater for malignant tumors. The incidence increased with proximity to the hypocenter for the directly exposed. An increased incidence was also found among early entrants, particularly those entering the city within 3 days of the A-bomb.

Effect of experimental immune atrophic gastritis on the induction of gastric carcinoma by X-irradiation in ICR mice.

Divided doses of 6,000 or 8,000 rad of X-ray were given to the gastric region of ICR/JCL female mice with immune atrophic gastritis produced by the injection with allogenic stomach antigen. The carcinogenic effect of X-rays for inducing gastric carcinoma was significantly increased by this method. Two points can be presented as its reason. First, the pyloric gland mucosa regenerating from injuries by immunization was exposed to the divided dose of X-rays. Second, the marked requirement of gastrin secretion attributable to severe injuries of parietal cell mass by immunization and local X-irradiation acted as a promoting factor on the induction of gastric carcinoma by X-ray for a long time through the trophic effects on the pyloric gland.

Development of adenocarcinoma of the prostate in ICR mice locally irradiated with X-rays.

The pelvic region of male ICR/JCL mice 5 approximately 7 weeks of age was irradiated with 1,000 rad of X-rays for a total of 8 exposures ot determine whether such irradiation would induce prostatic carcinoma. Prostatic adenocarcinoma was observed in 5 of 135 animals which survived more than 12 weeks after the first X-irradiation. Because spontaneous development of prostatic adenocarcinoma has not been reported in mice, development of this carcinoma in these animals can be considered to be attributable to X-irradiation.
