Presence of calcitonin gene-related peptide (CGRP) and substance P (SP) immunoreactivity in intraepidermal free nerve endings of cat skin.

Immunocytochemistry against neuropeptides contained within primary afferent neurons was used to study the morphology and distribution of intraepidermal free nerve endings in cat facial skin. Calcitonin gene-related peptide (CGRP) and substance P (SP) immunoreactivity was found in similar intraepidermal nerve endings of cat glabrous and hairy skin epithelia. The greatest density of immunoreactive intraepidermal nerve endings was located in the nose epidermis. Small limited areas with immunoreactive intraepidermal nerve endings were also found in facial hairy skin.

Immunocytochemical analysis of calcitonin gene-related peptide and vasoactive intestinal polypeptide in Merkel cells and cutaneous free nerve endings of cats.

Calcitonin gene-related peptide (CGRP)- and vasoactive intestinal polypeptide (VIP)-immunoreactivity were observed to coexist in Merkel cells of cats. No differences in peptide content were found between Merkel cells located in epithelia of the hard palate, in hairy and glabrous skin of the upper lip, and in vibrissae follicles. CGRP- and VIP-immunoreactive nerve fibres were also found near CGRP/VIP-immunoreactive Merkel cells. In the vibrissae follicles some CGRP- and VIP-immunoreactive nerve terminals end abutting on the glassy membrane. Other CGRP-immunoreactive nerve fibres penetrate the epithelium of the skin and end within it. Electron microscopy of vibrissae follicles revealed that Merkel cell neurites are not immunostained and that immunostained nerve fibres form unmyelinated bundles before ending freely. Thus, CGRP- and VIP-immunoreactive nerve fibres in cat skin do not end as Merkel cell neurites but as different kinds of free nerve endings.

Distribution of enkephalin-immunoreactive nerve fibres and terminals in the region of the nucleus basalis magnocellularis of the rat: a light and electron microscopic study.

This investigation was carried out on the distribution of enkephalin-containing nerve fibres and terminals in the region of the nucleus basalis magnocellularis (NBM) of the rat. At the light microscope (LM) level, enkephalin-immunoreactive sites and endogenous choline acetyltransferase (ChAT) were demonstrated by employing the two-colour immunoperoxidase staining technique, using highly specific monoclonal antibodies against enkephalin and ChAT. A pharmacohistochemical procedure to reveal acetylcholinesterase (AChE)-synthesizing neurons combined with the peroxidase-antiperoxidase (PAP) immunocytochemical technique to detect endogenous enkephalins, provided ultrastructural data on the relationships of neuronal elements containing AChE and enkephalins in the region of the NBM. At the LM level, cholinergic neurons of the NBM were surrounded by a dense network of enkephalin-immunoreactive nerve fibres. Electron microscopic (EM) observations of histochemically characterized structures, that were first identified in the LM, revealed that intensely AChE-stained structures in the region of the NBM received sparse synaptic inputs from enkephalin immunoreactive terminals. Synaptic inputs of immunoreactive terminals onto intensely AChE-stained neuron cell bodies were not detected. Synaptic contacts onto proximal AChE-positive dendrites were sparse, but the density increased on more distal regions of the dendrites. All immunoreactive boutons studied established symmetrical synaptic contacts with AChE-positive post-synaptic structures. The pattern of the synaptic input to these cells differs strikingly from that onto typical globus pallidus neurons. The perikarya and dendrites of the latter neurons were characteristically ensheathed in immunoreactive synaptic boutons. Results are consistent with the view that enkephalin-like substances in the rat might be synaptic transmitters or neuromodulators in the area of the NBM and that cholinergic neurons of the NBM (Ch4) are integrated into the circuitry of the basal ganglia. Enkephalins may play an important role regulating the extrinsic cholinergic innervation of the neocortex.

Blockade of muscarinic, histamine H1 and histamine H2 receptors by antidepressants.

The antagonistic effect of various antidepressants and of alprazolam on muscarinic and histamine H1 receptors has been studied in the guinea-pig ileum, and those on histamine H2 receptors on rat uterus. All antidepressants act like competitive antagonists on muscarinic and histamine H1 and H2 receptors. Alprazolam has shown competitive antagonism on muscarinic and histamine H1 receptors, and noncompetitive antagonism on histamine H2 receptors. Large differences in anticholinergic activity appear between the drugs studied, whereas all of them showed a higher histamine H1 receptor antagonistic activity. Most of the drugs studied showed a histamine H2 receptor antagonistic activity similar to that of cimetidine.

Verapamil and nifedipine effect on K+-induced contractions of guinea pig ileum during postnatal development.

The effects of verapamil and nifedipine on K+-induced contractions were studied in the isolated ileum of newborn, 30-day-old and adult guinea pigs. Both verapamil and nifedipine inhibited the K+-induced contractions, with nifedipine demonstrating the highest activity. The tonic component of the K+ response was more affected by the calcium antagonists than the phasic component. No changes in sensitivity to verapamil and nifedipine were recorded during the first month of postnatal development, thus suggesting that the function of calcium channels involved in the response to K+ does not change during this period.

Alcohol consumption among university students in Spain.

Alcohol consumption in 1984 among 2921 students of nine Faculties in the University of Valladolid (Spain) is reported. Alcoholic drinks were frequently consumed, especially by males and during weekends. Per capita consumption was 9.85 l of absolute alcohol/year for males and 4.971 for females. Beer, wine and gin were the favourite drinks. Alcohol consumption was similar in the different faculties and age groups but was to some extent related to age and place of residence in male students. Students' alcohol consumption was not as high as in the general Spanish population, and seems to have been stable during the last 10 years.

Effects of verapamil on acetylcholine and field stimulation induced contraction in postnatal development of guinea-pig ileum.

The effects of verapamil on acetylcholine and field stimulation induced contraction were studied in the isolated ileum of newborn, 5, 10, 15, 20 and 30 day old guinea-pigs. Verapamil inhibits the contraction evoked by acetylcholine or by field stimulation, being more affected the response to acetylcholine than to field stimulation. Verapamil IC50's against acetylcholine and field stimulation differ throughout the periods of development considered. These data suggest that the function of verapamil-sensitive calcium channels of guinea-pig ileum involved in the response to acetylcholine and field stimulation change during the period of development studied.

Effect of aluminum chloride on ascending colon of the rat.

Aluminum chloride (2.48 X 10(-4) to 1.24 X 10(-3) M) inhibits spontaneous motility of ascending colon of the rat. Resting tension was not affected. At concentrations between 2.07 X 10(-5) to 1.24 X 10(-3) M it inhibited in a dose-dependent manner the contractions induced by maximal doses of acetylcholine. Higher doses produced a very marked inhibition that was attributable to acidification of the medium. The inhibiting effect was more intense when the incubation medium contained an excess of calcium (3.6 and 4.3 mM) or theobromine (10(-4) and 10(-3) M). This inhibiting effect tended to be smaller in the presence of low calcium (0.6 and 1.0 mM) or verapamil (3 X 10(-8) and 10(-7) M).

Effect of trazodone on oxidative metabolism of rat brain in vitro.

Oxygen uptake of rat brain homogenate was reduced by 1 mM trazodone, a new atypical antidepressant. Na,K-ATPase activity and the associated oxygen consumption of rat brain slices were also reduced. Oxygen consumption of rat brain slices was enhanced by dopamine and this effect was blocked by 0.0001 mM trazodone. This drug uncoupled oxidative phosphorylation.

Medication and fitness to drive.

PURPOSE: The aim of this study is to analyze the consumption patterns of medicaments among motor vehicle drivers who attend 'Medical Driving Test Centres' and the relation between habitual consumption of medicaments and fitness to drive. METHODS: The study was carried out on 8043 drivers who attended 25 Medical Driving Test Centres. RESULTS: 24.7% of drivers chronically consume medicaments while 6.8% consume medicaments along with alcohol every day. Of those who chronically consume medicaments with a warning about the medications on driving, 65.8% were considered 'fit' to drive, 27.3% 'fit with restrictions', 5.1% 'suspended' and 0.4% 'unfit'. CONCLUSIONS: The results show how frequent the consumption of medicaments along with alcohol is and that the great majority of drivers who take medicaments are considered fit to drive.
