[Activity of heme synthesis enzymes in the bone marrow cells of rats with alimentary iron-deficient anemia].

An intensification of aminolevulinic acid (ALA)-synthase on the day of experiment and its tend to its further gradual inhibition noted to be in the bone marrow cells of rats with alimentary iron-deficient anemia (IDA). An activity of porfobilinogen (PB)-synthase was increasing in all the terms of the trial, that of ferrochelatase--on the day 10, and it was inhibited later on. Such changes in an activity of heme synthesis key enzymes result in an accumulation of intermittent products. An increase in MDA level, an inhibition of antioxidant defense enzymes found to be in all the terms of trial, SOD--in all the terms of studies, catalase--in late terms.

[Changes in some parameters of nitric oxide synthesis in the early periods of development of hepatocarcinogenesis].

Our objective in this study was to determine some indices for the synthesis of nitric oxide in the early periods of diethylnitrosoamine-induced (10 mg/kg 5 times a week for two months) hepatocarcinogenesis in mature male rats (n = 100) and to gain insights into their relation to lipid peroxidation process. During the time-related course of hepatocarcinogenesis the blood serum level of L-arginin has been found to change significantly with time of the study while that of malonic dialdehyde tended to be on the increase. The SOD activity increased somewhat during the period of initiation, with its significant reduction recordable during the period of promotion and a striking increase in the initial period of progression. Unlike SOD, catalase was showing a tendency to increase after some decline in its activity.

[The effect of cobavit on glutathione system in patients with bleeding gastroduodenal ulcer].

Index of glutathione system studied in 20 healthy persons (KG-1), in 20 with non-complicated duodenal ulcer (KG-2), in 210 patients with bleeding gastroduodenal ulcer, in 77 in early period after different operations. Results obtained showed that ulcer disease, complicated with bleeding was characterized by significant changes in glutathione system degree of which was depended on amount of the blood lost. Operations in early period enhanced disorders in glutathione system. Inclusion of cobavit into the complex of postoperative therapy provided recovery of glutathione pool activation of glutathiontransferase and glutathionreductase reaction that resulted in increase of withdrawal of toxic metabolites and activation of biosynthetic processes in patient's organism.

[The function of the liver mitochondria in rats with experimental cirrhosis undergoing an organ-preserving operation on the spleen].

The effect of splenectomy and of spleen-preserving operation with suture ligation of the left gastric artery on the functional status of liver mitochondria was studied by using 102 white male-rats of mixed population with experimental cirrhosis. The obtained data made it evident that in the immediate postoperative period (from 1 to 3 weeks after the procedure) in the animals of both the first and the second series, the disorder of energetic regulation of mitochondria hepatocytes respiration and the decrease in phosphorylation efficiency had the same tendency, which were seemingly caused by stress-action of the operative trauma. The data accumulated in the more distant postoperative period (8 weeks after the procedure) indicated that the resection of the lower splenic pole in combination with supplementary liver arterialization improved essentially the functional status of mitochondria hepatocytes and was more beneficial in contradistinction to splenectomy.

[A comparative study of the efficacy of pharmacological agents in regulating the level of NADPH in the hepatocytes in an acute lesion].

Experiments on mice with acute toxic damage to the liver induced by thioacetamide (200 mg/kg) demonstrated inhibited activity of enzymes glucose-6-phosphate dehydrogenase (G-6-PDH), NADPH-dependent isocitrate dehydrogenase and malate dehydrogenase in the hepatocyte mitochondrial and microsomal-cytosol fractions. Pharmacotherapy with membranostabilizers and cytochrome P-450 inducers activated the enzymes under study, the degree of activation depended on agents used.

Acute Disease↗

[Changes in the activity of enzymes of heme synthesis and catabolism, and level of microsomal hemoproteins during the liver acute intoxication by thioacetamide].

Thioacetamide administration to rats (20 mg/100 g) caused the development of toxic hepatitis which was accompanied by the increase of hepatic ALA-synthase and D-ALA that led to accumulating free porphyrines in the liver. At the same time an increase in activity of heme oxigenase was also found. A decrease in heme synthesis correlated with a decrease in content of cytochrome P450 and b5 in microsomal hepatic fraction of experimental animals.

[Comparative evaluation of antioxidant properties of neoselen and berberin in chronic heliotrinal hepatitis].

Prominent features of heliotrin-related chronic toxical disorders of the liver developing in hepatic cirrohosis include hyperfermentemia, hypoproteinemia, intensification of lipid peroxidation (LPO), as evidenced by decrease in the activity of the antioxidant defence. In chronic active liver disorders, hyperfermentemia and hyperlipoperoxidation are common, but it is later in the course of the experiment that hypoproteinemia and hypoalbuminemia and manifest depression of the antioxidant defence that come to be developing. Berberin experimental pharmacotherapy has been shown to attenuate the degree of hyperfermentemia, hyperlipoperoxidation and to increase levels of albumin and antioxidant defence enzymes, particularly so earlier in the course of the studies conducted but with aggravation of the pathological process the efficacy of the drug tended to be lower. Neoselen appeared to be effective in alleviating the cytologic syndrome, hyperlipoperoxidation, increasing the activity of the antioxidant defence enzymes. Patients are found to derive more benefit from neoselen therapy with prolongation of its use.

[Toxic effects of various water pollutants on structural and functional parameters of hepatocytes].

A prolonged intragastral intoxication of male and female rats by low doses of fluorine, zinc, chromium, arsenite, and combination of these compounds lead to developing cytolysis and hepatic-cellular insufficiency. These changes are more pronounced in female rats especially if chromium, arsenite and a combination of toxicants were used. They lead to developing hyper lipid peroxidation in male rats the most expressed by intoxication with arsenite and combined effect. Unlike to male rats LPO intensity in female rats was more pronounced and fixed by effect of all preparations. SOD activation was revealed to be in male rats by effect of chromium and especially zinc, in female rats by effect of fluorine and zinc. A distinctive inhibition of SOD activity determined to be by using arsenite and combination of toxicants. Activation of catalase especially in male rats was also revealed. Activation of membrane-destructive processes in the liver of experimental animals results in reducing pharmaco-metabolizing function of this organ.

[Effects of kavergal on indices of lipid peroxidation and the condition of the antioxidant system in patients with rheumatic heart disease presenting with circulatory insufficiency].

132 patients with rheumatic heart disease presenting with circulatory insufficiency displayed increased LPO both in the blood plasma and red cells, decline in the antioxidant enzymes activity varying with the circulatory insufficiency functional class degree of severity. Basic therapy with making use of antiinflammatory drugs, cardiac glycosides, diuretics together with drugs endowed with an antiarrhythmic activity and nitroglycerin (where indicated) was found to have practically no effect on LPO level or activity of the antioxidant system. The use of the drug kavergal, 1 g three times daily (total daily dosage being 3 g) in the complex therapy, has been shown to significantly decrease hyperlipoperoxidation both in the blood plasma and red cells increasing the activity of enzymes of the antioxidant defence.

[Effect of treatment with perftoran on the course of experimental uveitis, lipid peroxidation and activity of antioxidant defense enzymes].

Experimental uveitis is characterized by a pronounced lipid peroxidation (LPO) in damaged tissues of eye, which is complicated by reduced activity of the antioxidant enzymes, SOD and catalase. The intensification of LPO was also found in blood serum and hepatic tissue. However, hepatic lipid peroxidation was accompanied by compensatory increase of catalase activity. The activity of antioxidant defence enzymes decreased in blood, whereas catalase was activated in hepatic tissue. The therapeutic effect was accompanied by a decrease of LPO products and increase of activity of the antioxidant enzymes in all tissues. Treatment of uveitis with gentamycin and, especially, perftoran reduced inflammatory events.

[The state of the microsomal oxidative system and liver mitochondria in rats after splenectomy].

Activity of main microsomal enzymes and the functional state of liver mitochondria were studied in male rats within 4, 7, 10 and 30 days after splenectomy. Within early periods after the operation content of cytochromes P-450 and b5, activities of amidopyrine N-demethylase, aniline hydroxylase and NADPH cytochrome c reductase were decreased, while the functional state of rat liver mitochondria was impaired. Administration of splenine into the animals within 6 days normalized distinctly the microsomal oxidation and reduced completely the liver mitochondria functional state in the systems involving both substrates of oxidation.

[Functional state of rat liver mitochondria in experimental peritonitis].

Respiration and oxidative phosphorylation were studied in liver mitochondria of rats with experimental peritonitis. Peritonitis did not cause any distinct alterations in the functional state of liver mitochondria. Activity of main mitochondrial enzymes was only slightly altered. Inactivation of metabolic toxins occurred in cytoplasm of hepatocytes, i. e. out of mitochondria. Addition of peritoneal exudate at various dilutions to intact isolated mitochondria caused uncoupling of respiration and oxidative phosphorylation.

[Functional state of the rat liver mitochondria in acute kidney failure].

Activity of mitochondrial enzymes, respiration and oxidative phosphorylation in liver mitochondria were studied in rats with acute kidney insufficiency caused by bilateral nephrectomy. Depending on severity of kidney insufficiency, distinct alterations were observed in the functional state of animal mitochondria, especially during the low energetic shifting even down to uncoupling of respiration and oxidative phosphorylation; the phenomenon appears to be one of reasons of liver tissue failure in uremia.

[A comparative evaluation of the action of kvamatel and omez on the glutathione system in different sections of the digestive system in experimental duodenal ulcer].

Experiments on male albino rats demonstrated significant changes in the content of oxidized and reduced glutathione in the tissues of the liver, pancreas, stomach, duodenum, and small intestine in acetate ulcer of the duodenum. Experimental therapy with quamatel and particularly with omez led to activation of the glutathione system in the tissues under and in this way increased their resistance to the ulcerogenic effect.

[Disorders of the processes of protein biosynthesis and a decrease in the content of cytochromes P-450 and b5 in the microsomal fraction of rat kidney in the dynamics of development of acute pyelonephritis].

Noticeable decrease in microsomal protein level MOS terminal component--cytochrome P-450 and MOS median link--cytochrome b5 contents in microsomal fraction implicated in renal pathologic process was established in development of acute unilateral pyelonephritis. These findings reduction expression directly depended on severity and pathologic process development degree. One of the main causes led to reduction in cytochromes P-450 and b5 level and inhibition proteins biosynthesis may be influence of toxic products which are generated in substance exchange involved in prevalence of catabolic processes in injured kidney and intensification in endogenous intoxication syndrome on renal structures. Implication of many other factors effecting decrease in microsomal hemoproteins and protein levels in the damaged kidney in above pathology showed necessity to perform further investigations in the field given.
