Evolutionary perspective on a composite Shigella flexneri 2a virulence plasmid-borne locus comprising three distinct genetic elements.

Nucleotide sequence analysis of a Shigella flexneri 2a virulence plasmid-borne locus revealed that it comprised three distinct genetic elements: a stretch of colicin 1a/1b-linked sequence, a truncated IS911 element, and a third element containing two ORFs that shared a high level of similarity to a Salmonella-specific chromosomal sequence. Examination of other known IS911-like sequences showed that these sequences also were frequently associated with other accessory elements and appeared to be prone to partial deletion events. Analysis of the data led to a model of the evolution of this unusual composite locus.

Differential expression in human cells from the p6 promoter of human parvovirus B19 following plasmid transfection and recombinant adeno-associated virus 2 (AAV) infection: human megakaryocytic leukaemia cells are non-permissive for AAV infection.

Expression from the human parvovirus B19p6 promoter fused to the firefly luciferase ('Luc') reporter gene was evaluated in a non-erythroid human nasopharyngeal carcinoma cell line, KB, and a human megakaryocytic leukaemia cell line, MB-02, known to become permissive for B19 replication following erythroid-differentiation. The B19p6-Luc construct was introduced into KB and MB-02 cells, both in undifferentiated and differentiated states, either via DNA-mediated transfection, or via infection with recombinant adeno-associated virus 2 (AAV), a non-pathogenic human parvovirus known to possess a broad host-range. Although Luc activity was readily detected in KB cells following transfection of the B19p6-Luc plasmid DNA, no expression from the B19p6 promoter was observed following infection with recombinant virus. In addition, transfection of the reporter plasmid resulted in high-level expression of Luc in differentiated but not in undifferentiated MB-02 cells. However, no Luc activity was detected, even in differentiated MB-02 cells, following infection with recombinant virus. Further studies with an additional recombinant as well as wild-type (wt) AAV revealed that MB-02 cells were non-permissive for AAV infection. A second human megakaryocytic leukaemia cell line, M07e, was likewise resistant to infection by recombinant as well as wt AAV. Taken together, these studies identify the first human cell type that cannot be infected by AAV. They indicate that expression from the B19p6 promoter, in the context of an AAV genome, is restricted to primary human haematopoietic cells, perhaps because parvoviral DNA replication and transcription are intrinsically coupled.

[Influence of cerebral traumatic dementia treated with acupuncture at houxi and shenmen].

Sixty-two cerebral traumatic dementia patients were randomly divided into two groups acupuncture treatment group (Group A, 32 cases) and physical treatment group (Group B, 30 cases). The changes of MMSE scores and auditory P300 were observed before and after the treatment. It were found that the indexes in group A changed significantly (P < 0.05-0.001), and the indexes in group B didn't change significantly (P > 0.1-0.4). The therapeutic effect in group A was better than that in group B(P < 0.001). The results confirmed that the cognitive function of cerebral traumatic dementia patients was increased effectively with acupuncture at Houxi and Shenmen.

Physiological properties of neurons in superficial layers of superior colliculus of rabbits.

Neurons in superficial layers of the superior colliculus of the rabbit are classified into three types by their electrophysiological properties. Among them, two types belong to projecting neurons which send axons to the thalamic pulvinar (N = 52) and dorsal lateral geniculate nucleus (N = 54) respectively. All other neurons are pooled into the third type (N = 99). Projecting neurons of both types receive monosynaptic visual inputs via optic tract fibers of similar conduction velocity, indicating that in the superior colliculus of the rabbit, there is no difference in conduction velocity between the two pathways. They also receive trisynaptic inhibitory inputs, most likely via recurrent inhibitory circuits. The third type of neurons receives disynaptic optic and trisynaptic inhibitory inputs. The function of neurons of the third type is studied.

Kinetics of low threshold calcium channels of relay cells in cat lateral geniculate nucleus.

Kinetics of the low threshold T-type Ca2+ channel is studied with single electrode voltage clamp technique on brain slices of the cat lateral geniculate nucleus (LGN). Space clamp is dramatically improved by blocking various K+ and Na+ channels, decreasing Ca2+ current and selecting proper holding potentials. Results from this study are similar to those obtained from acutely dissociated LGN neurons of the rat, indicating that the kinetics of T-Ca2+ channels of the cat LGN neurons is the same as that of the rat LGN. The result reported previously on the cat LGN may result from a defect in space clamp.

[A study on the cumulative effect of repeated electroacupuncture on chronic pain].

Clinical and animal studies were performed to evaluate the effect of repeated transcutaneous electric nerve stimulation (TENS) applied on acupoints of pain patients and the effect of repeated electroacupuncture (EA) at acupoints of the rat on experimental arthritic pain models. Attempts were also made to evaluate whether substance P (SP), cholecystokinin octopeptide (CCK-8), and met5-enkephalin (MEK) are involved in the mechanism of the cumulative effect of repeated EA on experimental arthritis using neuropharmacological (receptor antagonists) and neurochemical (radioimmunoassay, RIA) approach. The main results show that repeated TENS of 100 Hz relieves spinal spasticity in a cumulative manner, while TENS of 2/15 Hz is effective in relieving chronic pain. In animal chronic pain models, repeated EA suppressed the hyperalgesia in arthritic rats in a cumulative manner. Further studies revealed that in pain patients there was a plastic change in the release and metabolic rate of spinal opioid peptides. In arthritic rats there was also a change in the releasing rate of spinal SP, CCK-8, and MEK as compared to control rats, an effect modulated by repeated EA. These plastic changes occured under chronic pain condition and their modulation by repeated acupoint stimulation may explain the mechanisms of the cumulative effect of acupuncture on chronic pain.

In vitro inhibition of Helicobacter pylori NCTC 11637 by organic acids and lactic acid bacteria.

In this study the effects of both pH and organic acids on Helicobacter pylori NCTC 11637 were tested. Lactobacillus acidophilus, Lact. casei, Lact. bulgaricus, Pediococcus pentosaceus and Bifidobacterium bifidus were assayed for their lactic acid production, pH and inhibition of H. pylori growth. A standard antimicrobial plate well diffusion assay was employed to examine inhibitory effects. Lactic, acetic and hydrochloric acids demonstrated inhibition of H. pylori growth in a concentration-dependent manner with the lactic acid demonstrating the greatest inhibition. This inhibition was due both to the pH of the solution and its concentration. Six strains of Lact. acidophilus and one strain of Lact. casei subsp. rhamnosus inhibited H. pylori growth where as Bifidobacterium bifidus, Ped. pentosaceus and Lact. bulgaricus did not. Concentrations of lactic acid produced by these strains ranged from 50 to 156 mmol l-1 and correlated with H. pylori inhibition. The role of probiotic organisms and their metabolic by-products in the eradication of H. pylori in vivo remains to be determined.

Adeno-associated virus 2-mediated gene transfer and functional expression of the human granulocyte-macrophage colony-stimulating factor.

It is becoming increasingly clear that the adeno-associated virus 2 (AAV)-based vector system may prove to be useful for high-efficiency gene transfer in human cells as well as for potential gene therapy in humans. A recombinant AAV vector containing the gene for a human hematopoietic growth factor, granulocyte-macrophage colony-stimulating factor (GM-CSF), was constructed and used to infect COS-1 cells, a monkey kidney cell line. COS-1 cells infected with the recombinant virus, but not mock-infected cells, expressed high levels of the human GM-CSF gene transcripts. Furthermore, in co-cultivation experiments with the recombinant virus-infected cells, but not in those with mock-infected cells, active proliferation of a GM-CSF-dependent human megakaryocytic leukemia cell line, M07e, could be obtained in the absence of exogenously added GM-CSF. The recombinant GM-CSF protein released into the supernatant was biologically active in progenitor cell assays carried out with primary human hematopoietic cells, and this activity was specifically abrogated by treatment of the supernatant with anti-GM-CSF antibodies. This recombinant virus may be potentially useful in the management and gene therapy of a variety of malignant disorders in the human hematopoietic system.

[Detection of human papilloma virus in esophageal squamous cell carcinoma and adjacent tissue specimens in Linxian].

A total of 87 human esophageal tissue specimens, comprising 35 squamous cell carcinomas and 52 adjacent tissue specimens (with no gross evidence of tumors) were obtained from patients who underwent surgery for cancer of the esophagus in Lin-xian county, Henen Province. The tissue specimens were analyzed for the presence of HPV DNA by Southern blot assay with HPV type 16 and 18 probes and by polymerase chain reaction (PCR) with HPV consensus primer. The esophageal epithelial specimens of human fetus, and condyloma tissue contained HPV type 6 were used as controls. The results showed that no HPV 16, 18 and other type were detectable in any case, either in the esophageal carcinomas or adjacent tissue specimens. Thus, the role of HPV in the carcinogenesis of esophagus remains unclear. The high incidence of esophageal squamous cell carcinoma in Lin-xian is probably related to other factors such as N-nitrosamines.

Novel cDNA clones obtained by antibody screening of a mouse cerebellar cDNA expression library.

In order to obtain cDNAs of genes that are expressed in cerebellar granule cells (GC), an antiserum was raised against GC isolated from mouse cerebella. Western blot analysis demonstrated that antibodies against multiple proteins were present and immunohistochemical analysis showed that at least some of these proteins were localized to cerebellar GC. The antiserum was used to screen an expression library derived from mouse cerebellar cDNA. Twenty-two granule cell antibody-positive (GCAP) clones were obtained. Of these, eight represented genes previously described and 14 were novel clones (not found in the GenBank database). In situ hybridization histochemistry showed that eight of the novel clones had moderate to strong expression in cerebellar GC and some of these clones were expressed also in the hippocampal formation. One such clone, GCAP-7, appears to represent a single-copy gene and the entire cDNA insert (2,688 bp) has been sequenced. The clone appears to consist primarily of the 3' untranslated portion, including a poly(A) tail and polyadenylation signals, of a 5 kb transcript. The GCAP clones should be useful for future studies of molecular biology of GC in normal individuals and in inherited neurologic disease with GC degeneration.

Crystal structure of murine cyclophilin C complexed with immunosuppressive drug cyclosporin A.

Cyclophilin is a cellular receptor for the immunosuppressive drug cyclosporin A (CsA). Cyclophilin C (CyPC) is highly expressed in murine kidney, making it a potential mediator of the nephrotoxic effects of CsA. The structure of murine CyPC complexed with CsA has been solved and refined to an R factor of 0.197 at a 1.64-A resolution. Superposition of the CyPC-CsA structure with the unligated cyclophilin A (CyPA) revealed significant migration of three loops: Gln-179 to Thr-189, Asp-47 to Lys-49, and Met-170 to Ile-176. The proximity of the loop Gln-179 to Thr-189 to the CsA binding site may account for the unique binding of a 77-kDa glycoprotein, CyPC binding protein (CyCAP), to CyPC. The binding of CsA to CyPC is similar to that of CsA to human T-cell cyclophilin A (CyPA). However, the conformation of CsA when bound to CyPC is significantly different from that when bound to CyPA. These differences may reflect conformational variation of CsA when bound to different proteins. Alternatively, the previous CyPA-CsA structure at low resolution may not provide sufficient details for a comparison with the CyPC-CsA structure.

[Determination of trace chloride and sulfate in drugs by ion chromatography].

Chloride and sulfate in magnesium trisilicate, activated charcoal, vesnarinone and bendazac were determined by the IC conductimetric detection. Analysis were performed by using a separating column (100 x 6 mm i.d.) packed with anion exchanger and with a stripper column (100 x 6 mm i.d.); 1.4 mmol/L Na2CO3 and 1.4 mmol/L Na2CO3-0.75 mmol/L NaHCO3 were used as eluents respectively. The peak height was linear with concentration from 10(-6) to 10(-3) mol/L for Cl- and from 10(-5) to 10(-3) mol/L for SO4(2-). Results showed no significant difference as compared with those determined by the CHP (1985) and CHP (1990) appendices methods. The IC method is specific, sensitive, simple and rapid.

[Enkephelinase inhibitors acetorphan and SCH34826 potentiate the analgesic effect of low frequency electroacupuncture in rats].

Influence of two novel enkephelinase inhibitors, Acetorphan and SCH34826, on the effect of electroacupuncture analgesia (EAA) has been observed in rats. Administered Acetorphan (2.0-4.0 mg/kg, i.p.) or SCH34826 (30-100 mg/kg, p.o.) potentiated and prolonged the effect of 2 Hz EAA significantly in the nonresponder in which an evident analgesic effect can not be induced by 2 Hz electroacupuncture. This means that che non-responder of 2 Hz EAA can be converted into the responder with these two drugs. The non-responder of 100 Hz EAA, however, can not be changed into the responder with Acetorphan. These results again support our previous reports that enkephelin plays an important role in low frequency EAA. Meanwhile, this suggests us a probable route for potentiating acupuncture analgesia in clinical practice.

[The measurement of radial bone mineral content in children].

The bone mineral contents of radius of 255 boys and 243 girls (7 to 16 year old of the Han nationality, living in Chengdu) were measured with Bone Mineral Analyzer (Type:SPA-I). The standard values of g/cm was made. There was a positive correlation between the value in boys or girls and chronological age, height or weight. The coefficients were respectively 0.83, 0.77, 0.64 in boys and 0.82, 0.82, 0.82 in girls (P less than 0.005). At the age of 13 (in boys or in girls) the value of the g/cm was the highest.

A rat model for severe limb ischemia at rest.

We sought an animal model able to discriminate metabolic and angiogenic processes in limb ischemia. For that we modified and evaluated a rat model of severe unilateral limb ischemia at rest. A two-stage surgical procedure entailing left femoral artery ligation preceded by interruption of collateral vessels originating from the infra-renal aorta and left iliac arteries was performed in Sprague-Dawley rats. The model was evaluated for up to 8 weeks with a transit-time flow meter, a laser Doppler perfusion imager, microspheres, arteriography and histology. It was found to be well tolerated with low mortality and perfusion in the foot skin was reduced up to 8 weeks, while collaterals were visible after 2 weeks. Histologic signs of ischemia were seen for up to 4 weeks. This rat model of severe limb ischemia at rest lasts up to 8 weeks and seems well suited for longitudinal studies of the pathophysiology of limb ischemia and healing mechanisms like angio- and arteriogenesis.

The intestinal absorption of camptothecin, a highly lipophilic drug, across Caco-2 cells is mediated by active transporter(s).

The intestinal transport properties and kinetics of camptothecin (CPT) through Caco-2 cell monolayers were characterized by determining concentration-, temperature-, and ATP-dependence as well as the effect of selected inhibitors. The absorptive permeability (Peff) of CPT was found to be concentration dependent and saturable (K(m) = 31.2 +/- 6.9 microM) indicating the involvement of a high affinity, low capacity transport system. CPT transport was also temperature dependent and inhibited by sodium azide plus 2-deoxyglucose, which deplete cellular ATP, further suggesting that an active, carrier-mediated transport system contributes to CPT absorption. Based on inhibition studies, the involvement of organic anion and cation transporters was implicated but not conclusively demonstrated. Total CPT transport decreased four fold with increasing pH from 5.5 to 8.5 indicating that CPT lactone contributed more significantly to overall CPT transport than CPT carboxylate. The results of these studies suggest that CPT absorption is mediated by multiple mechanisms including significant passive diffusion and active transport components. Since typical substrates for intestinal carriers are hydrophilic and charged, the involvement of putative absorptive carriers in the transport of CPT is a novel finding that may give insight into the erratic oral bioavailability of CPTs observed in the clinic.
