[Polymorphism of platelet glycoprotein Ib in the Spanish population ].

PURPOSE: To assess the polymorphism of the platelet membrane glycoprotein in castilians and gypsy people and to confirm the genetic transmission of such glycoprotein by means of family studies. MATERIAL AND METHODS: Two-hundred castilians and 87 gypsy subjects were studied. Glycoprotein Ib was analysed by means of SDS polyacrylamide gel electrophoresis followed by Western-Blotting and labelling with glycoprotein-specific monoclonal antibodies and lectins. The different populations were compared by using the chi-square method (Fisher's test). RESULTS: Only three groups of glycoprotein Ib, namely, B, C and D, were found in both population groups, no differences being observed in the expression of the different phenotypic variants. Lesser polymorphism was present in the gypsy group. The family studies showed that glycoprotein Ib is genetically determined, the two alleles being co-dominantly expressed in each individual. CONCLUSIONS: The polymorphism of glycoprotein Ib is similar to that of caucasians, type A being not found. No differences seem to exist between castilians and gypsy people. The heredity pattern is autosomal co-dominant.

Simultaneous effects of p-chloroamphetamine, d-fenfluramine, and reserpine on free and stored 5-hydroxytryptamine in brain and blood.

The effects of acute treatment with p-chloramphetamine, d-fenfluramine, and reserpine on intracellular (brain tissue and whole blood) and extracellular (CSF and platelet-free plasma) compartments of 5-hydroxytryptamine (5-HT) in the brain and blood of the same rats have been examined. These treatments affected 5-HT in brain tissue and whole blood similarly (r = 0.823). Reserpine significantly reduced both intracellular pools at 2 and 24 h. p-Chloroamphetamine and d-fenfluramine were more effective on brain tissue 5-HT. The concentration of 5-HT in CSF was significantly increased by all treatments. p-Chloroamphetamine induced a dramatic 70-fold increase of CSF 5-HT, paralleling a 42% decrease in brain tissue. d-Fenfluramine significantly increased CSF 5-HT to 212% of controls and reduced whole brain 5-HT (-23%). The effects of p-chloroamphetamine and d-fenfluramine on 5-HIAA in brain, CSF, and plasma were nonsignificant. Individual values of 5-hydroxyindoleacetic acid (5-HIAA) in CSF and brain were highly correlated (r = 0.855), indicating that CSF 5-HIAA reflects well the concentration of 5-HIAA in brain tissue. Yet the intra- and extracellular concentrations of 5-HIAA were unrelated to the 5-HT changes. This indicates that CSF 5-HIAA does not reflect the active (extracellular) compartment of 5-HT in brain.

Thimerosal induces calcium mobilization, fructose 2,6-bisphosphate synthesis and cytoplasmic alkalinization in rat thymus lymphocytes.

The effect of thimerosal on intracellular calcium ([Ca2+]i), pH (pHi) and fructose 2,6-bisphosphate (Fru 2,6-P2) in thymus lymphocytes was investigated. The effect of thimerosal on cell growth was also examined. Thimerosal produced a dose-dependent increase in [Ca2+]i, pHi and in the level of fructose 2,6-bisphosphate. Thimerosal was, however, unable to produce cell proliferation and inhibited [3H]thymidine incorporation when cells were challenged with PHA and costimulator. In the absence of external calcium, thimerosal produced only a slight increase in [Ca2+]i. In Na(+)-containing buffer, thimerosal induced an initial acidification (0.05 +/- 0.01 pH units), followed by an alkalinization of 0.08 pH units/min, whereas in Na(+)-free media, pHi decreased 0.2 +/- 0.02 units and this acidification was maintained for more than 40 min. When external calcium was removed the initial acidification was unchanged and no further increase in pHi was observed. Polymyxin B, an inhibitor of protein kinase C, did not modify the initial thimerosal-induced acidification although pH returned to basal levels after 10 min. It was concluded that alkalinization induced by thimerosal is probably due to activation of the Na+/H+ exchanger and that changes in internal Ca2+, pH and metabolic rate are not sufficient to induce cellular proliferation. The mechanism by which thimerosal inhibits thymocyte proliferation remains to be clarified.

Long-term pinealectomy alters hypothalamic serotonin metabolism in the rat.

In the present study, the effects of long-term pinealectomy on tryptophan, 5-hydroxytryptamine (5-HT or serotonin), 5-hydroxy-3-indoleacetic acid (5-HIAA), and tryptophan hydroxylase and monoamine oxidase activities were studied in preoptic area-anterior hypothalamus (POA-AH) and in the medial and posterior hypothalamus of the rat. After pinealectomy, 5-HT levels decreased significantly in medial hypothalamus but increased in the POA-AH. The levels of 5-HIAA decreased significantly in the POA-AH and medial hypothalamus. Tryptophan levels remained unchanged while tryptophan hydroxylase activity diminished significantly in POA-AH and medial hypothalamus. Monoamine oxidase activity remained unchanged in the hypothalamic regions. These results suggest that pinealectomy induces differential inhibitory actions on the serotoninergic terminal regions, mainly in anterior and medial hypothalamic areas.

Intradermal and intramuscular route for vaccination against hepatitis B.

A recombinant hepatitis B vaccine was administered to high-risk hospital personnel by intramuscular (20 micrograms) or intradermal (2 micrograms) injections for the primary immunization (n = 69) with three doses and booster immunization (n = 51) with one dose. Basic vaccination performed intramuscularly gave rise to significantly higher seroconversion levels (97.2% versus 78.1%) and geometric mean titres of antibody (1649 versus 126 IUl-1) as compared with the intradermal route. Intradermal administration did not boost antibody titres in patients previously vaccinated intradermally. Adverse reactions were not serious or severe. The intramuscular route is recommended as the procedure of choice when vaccinating against hepatitis B.

Subcutaneous metastasis following laparoscopy in gastric adenocarcinoma.

An 80-year-old male developed ascites as the first indication of an adenocarcinoma of the gastroesophageal junction. Laparoscopy was performed during the diagnostic study of the ascites and, 7 days later at the point of introduction of the laparoscope, a metastasis developed in skin and subcutaneous cellular tissue. This is the third case of subcutaneous neoplastic implantation following laparoscopy to appear in the literature. The two previously described cases dealt with patients with ovarian adenocarcinoma. The literature reporting cases of tumor dissemination following technical procedures is reviewed.

[Infectious food poisoning in the Autonomous Basque Community (1984-1986)].

The results of the study of 117 outbreaks of food poisoning in the Basque Autonomous Community during 1984-1986 are reported. In 76% of these outbreaks the causative agent could be isolated. Bacteria of the genus Salmonella accounted for 86.5% of the outbreaks with a known etiology, S. enteritidis being the most commonly found serotype. In 90% of the outbreaks caused by Salmonella, the transmission was by eggs or egg containing products. The features of food poisoning in the Basque Autonomous Community are discussed.
